Comprehensive insights into hydrothermal pretreatment of rice straw from physicochemical structure, organic matter transformation and hydrolysate reuse

Anaerobic digestion is an effective means of biomass utilization from solid waste, but the complex structure and low biodegradability of lignocellulose severely limit the anaerobic degradation conversion efficiency. In this study, hydrothermal pretreatment combined with chemicals was conducted to investigate physicochemical structure, chemical composition and changes, and enzymatic hydrolysis performance of rice straw under various combination hydrothermal pretreatment. The transformation mechanism of main components during alkaline-thermal pretreatment and the potential reuse of hydrolysate were further explored. Results showed that the total sugar concentration after hydrothermal pretreatment was 1.49 times higher than that after dissolution pretreatment. Hydrothermal-KOH pretreatment could destroy the waxy and siliceous layer on the straw surface. The roughness increased from 0.525 to 1.170 nm, and the average pore size 6.20 to 8.99 nm. Alkaline-thermal pretreatment could facilitate lignin dissolution and retained more cellulose and hemicellulose in the solid phase, and sugar concentration after pretreatment could reach 40.82 g/L. Porosity was affected by the dissolution of chemicals, and the pore structure also affected subsequent enzymatic hydrolysis. Dissolution of cellulose and hemicellulose resulted in a reduction in particle size and the formation of micropores, while re-aggregation and precipitation of lignin after dissolution led to the pore collapse and an increase in pore size. Furthermore, the hydrolysates contained nutrients, humic acids, plant hormones, and micronutrients, which had the potential to be developed into foliar fertilizers and reused. The results of this study will provide new insights and technical support for high-value utilization of straw.

Anaerobic digestion of sewage sludge using anaerobic dynamic membrane bioreactor under various sludge composition and organic loading rates

This study investigates the effects of sludge compositions and organic loading rates (OLRs) on stable biogas production during sludge digestion. Batch digestion experiments evaluate the effects of alkaline-thermal pretreatment and waste activated sludge (WAS) fractions on the biochemical methane potential (BMP) of sludge. A lab-scale anaerobic dynamic membrane bioreactor (AnDMBR) is fed with a mixture of primary sludge and pretreated WAS. Monitoring of volatile fatty acid to total alkalinity (FOS/TAC) helps maintain operational stability. The highest average methane production rate of 0.7 L/L·d is achieved when the OLR, hydraulic retention time, WAS volume fraction, and FOS/TAC ratio are 5.0 g COD/L·d, 12 days, 0.75, and 0.32, respectively. This study finds functional redundancy in two pathways: hydrogenotrophic and acetolactic. An increase in OLR promotes bacterial and archaeal abundance and specific methanogenic activity. These results can be applied to the design and operation of sludge digestion for stable, high-rate biogas recovery.

Efficient caproate production from ethanol and acetate in open culture system through reinforcement of chain elongation process

The open culture system exhibits several advantages in bioconversion of complex substrates into caproate, yet it is urgent to develop an advanced approach to overcome the low fermentative efficiency. In this study, three reinforcement techniques were employed to reinforce the chain elongation in the open culture system, including bioaugmentation, pretreatment of anaerobic sludge (AS) and addition of cell carrier. Results showed that the caproate production was improved to 48.9 mmol/L using bioaugmentation technology. With removal of methanogen by alkaline-thermal pretreatment, the caproate production increased to 94.8 mmol/L. The highest caproate production of 130.5 mmol/L was obtained with addition of cell carrier, which was explained by the formation of biofilm. The genus Clostridium_sensu_stricto capable of medium chain fatty acids production dominated the microbiome of the AS. This study provides an insight that efficient caproate production can be achieved in the open culture system by reinforcing the pathway of chain elongation.

Enhanced methane production using pretreated sludge in MEC-AD system: Performance, microbial activity, and implications at different applied voltages

Although various pretreatment methods are employed to promote sludge hydrolysis and thereby promoting methane production in the subsequent microbial electrolysis cell assisted anaerobic digestion (MEC-AD) system, the questions arise are, “which pretreatment method on waste activated sludge (WAS) maximises the sludge hydrolysis and what is the optimal applied voltage on anaerobic digestion (AD) to stimulates the direct interspecies electron transfer (DIET) performance and thereby accelerating the methane production fed with pretreated WAS?” was still unanswered. Herein, firstly, a series of pretreatment methods to hydrolyse and mineralise the organic matter of WAS was performed to evaluate solubilization efficiency and thereafter, the influence of different applied voltages (0.3 V, 0.6 V, and 0.9 V) on coupled MEC-AD reactors fed with pretreated WAS was investigated to apprehend the DIET promotion for methane production. The results indicated that in MEC-AD reactors, the methane yield increased by 27.2%, 44.8%, and 37.3% when the applied voltages were 0.3 V, 0.6 V, and 0.9 V, respectively. Therefore, the alkaline-thermal pretreatment (ATP) enhanced the sludge hydrolysis in WAS, followed by an applied voltage of 0.6 V in the MEC-AD reactor fed with pretreated WAS, enhanced methane production under DIET stimulation induced by the increased abundance of electroactive microorganisms (EAM) and the advanced electron transfer. Besides, the energy balance estimation validates that with an applied voltage of 0.6 V in MEC-AD could achieve higher net energy input.

Sludge disintegration and anaerobic digestion enhancement by alkaline-thermal pretreatment: Economic evaluation and microbial population analysis

Alkaline-thermal pretreatment was examined for waste activated sludge (WAS) disintegration and subsequent anaerobic digestion (AD). Pretreatment at 60 °C was estimated to provide better economic benefits than higher temperature conditions. The maximum methane yield of 215.6 mL/g COD was achieved when WAS was pretreated at 60 °C and pH 10 for 24 h, which was 46.6% higher than untreated WAS. The pretreatment condition also provided the maximum net savings. The degree of sludge disintegration, considering both loosely bound-extracellular polymeric substance and soluble COD, would be a better indicator to predict anaerobic digestibility than the solubilization rate that considers soluble COD alone. Microbial analysis implied that pretreatment facilitated the growth of hydrolytic bacteria, phyla Bacteroidetes and Firmicutes. In addition, sludge pretreatment enhanced the growth of both acetoclastic and hydrogenotrophic methanogens, genera Methanosaeta and Methanobacterium. The mild AT-PT would be useful to enhance the digestion performance and economic benefit of WAS digestion.

Alkaline thermal pretreatment of waste activated sludge for enhanced hydrogen production in microbial electrolysis cells

Resource utilization of waste activated sludge (WAS) has become a mainstream development direction. Alkaline thermal pretreatment (TPT) was found to greatly promote the bioaccessibility and biodegradability of the sludge. The organic matter including soluble chemical oxygen demand (SCOD), soluble carbohydrate, soluble protein and volatile fatty acids (VFAs) after low temperature (90 °C) pretreatment was 4.8%–65.9% higher than that after high temperature (180 °C) pretreatment. These increasements could be contributed by the alkaline treatment condition and the longer treatment time. The alkaline condition reduced the resistance of cell wall to the temperature. The pretreatment time at 90 °C was two times of that at 180 °C, allowing more organic matter to be released. But the total energy consumption of low temperature pretreatment (2580.7 kJ/L) was 30.5% lower than that of high temperature pretreatment (3711.8 kJ/L). The sludge fermentation liquid (SFL) was then employed as the substrate in microbial electrolysis cells (MECs), and the utilization efficiency of acetic acid was the highest (74.9%–83.2%). The hydrogen yield using low temperature pretreated sludge was 0.44 m3/(m3·d), which was higher than that of using high temperature pretreated sludge (0.31 m3/(m3·d)). These results suggested that alkaline TPT at 90 °C was an effective way to hydrolyze sludge and further enhance hydrogen production in MECs.

Anaerobic Digestion of Digested Manure Fibers: Influence of Thermal and Alkaline Thermal Pretreatment on the Biogas Yield

Anaerobic digestion (AD) of animal manure converts only half of the organic material into biogas due to the presence of a significant amount of lignocellulosic materials in manure. In this study, alkaline thermal pretreatment was used for improving anaerobic digestion of residual manure fibers after AD. Anaerobic digestion of pretreated manure fibers was done in batch culture under mesophilic and thermophilic conditions. The results of the study showed that degradation of manure fibers was improved ca. 43.6% as a result of alkaline thermal pretreatment with 3% w/w NaOH added. Methane yield improved by 143.5 and 180.2% under mesophilic and thermophilic conditions, respectively. Compositional analysis of the effluent after AD showed the percentile conversion of 50.8% of cellulose, 59.5% of hemicellulose, 39.9% of acid-soluble and 21.7% of acid-insoluble lignin to methane under mesophilic conditions. Under thermophilic conditions, 57.3% of cellulose, 70.1% of hemicellulose, 39.4% of acid-soluble and 19.4% of acid-insoluble lignin were converted to methane. The result showed that alkaline thermal pretreatment of manure fibers can enhance the performance of AD while shortening the time needed to recover the maximum amount of biogas from AD.

Comprehensive study on the effects of process parameters of alkaline thermal pretreatment followed by thermomechanical extrusion in sugar liberation from Eucalyptus grandis wood

Abstract A combination of alkaline thermal pretreatment followed by thermomechanical extrusion was studied as a novel sequential pretreatment process for an effective breakdown of the lignocellulosic structure of Eucalyptus grandis wood (EW). The first step was studied by analysing the influence of two factors: the NaOH-to-dry biomass ratio or NaOH loading (NaOH/DM) and the liquid-to-solid ratio (L/S). Optimization of these two parameters provided good results in terms of enzymatic hydrolysis at 5% (w w−1) solids loading, obtaining a total sugar concentration of 24.9 g L−1 and a total sugar production of 36.9 g 100 g−1 raw EW after pretreating the biomass at 20% NaOH/DM and L/S = 1/1. The second step of extrusion, when followed by a final washing step, provided a significant increase in glucose and xylose production when working at 10% NaOH/DM. For a soda loading of 20%, there was a clear improvement in sugars conversion yield after extrusion and washing: 71% for glucan conversion and 89% for xylan.

Alkaline-thermal pretreatment of spectinomycin mycelial residues: Insights on anaerobic biodegradability and the fate of antibiotic resistance genes

Alkaline-thermal (AT) pretreatment is an economical and efficient pretreatment method to improve anaerobic biodegradability of biowaste. This study investigated the effect of AT pretreatment of spectinomycin mycelial residues (SMRs) for promoting anaerobic biodegradability along with the reduction of antibiotic resistance genes (ARGs), and thus obtained the optimal conditions of AT pretreatment. Biomethane potential (BMP) test was conducted to evaluate the anaerobic biodegradability of untreated and pretreated SMRs, and the fate of ARGs was tracked by quantitative polymerase chain reaction. Results showed that the modified Gompertz model fitted the results of BMP tests satisfactorily. Furthermore, AT pretreatment promoted BMP (B0) and reduced lag phase (λ) effectively. These were attributed to the solubilization of SMRs. The analyses of the changes in dissolved organic matter indicated that AT pretreatment could facilitate the solubilization of both biodegradable (e.g. protein) and recalcitrant matter (e.g. humic-like, analyzing by EEMs-PARAFAC), which had a significant corresponding positive (Person correlation, p < 0.01) and negative (Partial correlation, p < 0.01) influences on anaerobic biodegradability. However, the positive effects surpassed the negative effects, promoting the overall anaerobic biodegradability of SMRs. In addition, a considerable reduction of ARGs (by 0.62–1.36 log units) was observed at pH ≥ 12, attributed to the hydrolysis of phosphodiester bond of DNA in strong alkaline solution. Considering both anaerobic biodegradability and ARGs, the optimal AT condition was concluded as pH 12, temperature 90 °C and time 120 min.

Production of a bioflocculant by using activated sludge and its application in Pb(II) removal from aqueous solution

AbstractIn this study, the characteristics of a bioflocculant produced by using activated sludge as raw materials were investigated. The performance of this bioflocculant in the removal of Pb(II) from aqueous solution and the corresponding mechanisms were determined as well. After cultivating a bioflocculant-producing strain in an alkaline thermal pre-treatment sludge for 60 h, approximately 4.45 g of bioflocculant containing a protein backbone was harvested from 1 L of fermentation broth. This bioflocculant can remove 98.5% of Pb(II) from aqueous solutions under optimal conditions, which include a bioflocculant dosage of 6 mg/L and a CaCl2 concentration of 70 mg/L at a pH of 6.5.

Enhanced anaerobic digestion of sewage sludge by thermal or alkaline-thermal pretreatments: Influence of hydraulic retention time reduction

The influence of hydraulic retention times (HRTs) reduction from 25 days to 15 days on the enhancement effects of two pretreatments (thermal pretreatment and alkaline-thermal pretreatment) on the continuous anaerobic digestion (AD) of sewage sludge was studied in a long-term experiment (196 days). The operation of the semi-continuous AD fed with raw sludge or pretreated sludge was stable at the three HRTs. The methane production increased from 70.6 to 165.8 ml/L·d to 75.2–172.6 ml/L·d and the methane yield decreased from 98.9 to 234.9 ml/g added volatile solid to 65.6–144.9 ml/g added volatile solid when the HRT reduced from 25 days to 15 days. The two pretreatments reduced the HRT of raw sludge AD by over 40%, and the effects of the alkaline-thermal pretreatment were greater than those of the thermal pretreatment. The reduction of HRT from 25 days to 15 days increased the enhancement effects of the two pretreatments on the removal of organic matter (4.7–15.9% for volatile solid), average hydrolysis ratio (36.9–116.4%), and specific hydrolysis rate (44.1–155.6%) but decreased the enhancement effects of the pretreatments on the methane production (0.9–4.6%) and yield (4.0–15.8%), average reaction ratios (0.4–8.2%), and specific rates of the last three AD steps (0.1–13.9%). The influence of HRT reduction on the enhancement effects of the alkaline-thermal pretreatment for sludge AD was slightly greater than on the enhancement effects of the thermal pretreatment.

Hydrothermal and alkaline thermal pretreatment at mild temperature in solid state for physicochemical properties and biogas production from anaerobic digestion of rice straw

The effect of hydrothermal pretreatment (HTP) and alkaline thermal pretreatment (TA) at mild temperature in the solid state was comparatively studied, through the analysis of physicochemical properties of rice straw and its biogas production during anaerobic digestion. The results indicated that TA visibly increased the biogas yield from HTP. The accumulative biogas production of volatile solids was the highest for TA3 up to 411.1 mL/g VS, in which increased 24.04% compared to control. Therefore, the use of TA is more favorable to improve biogas production rates of anaerobic fermentation with rice straw than HTP, however, the additives of calcium hydroxide are too high, but it is not conducive to promoting the biogas yield of rice straw. The best additives amount of Ca (OH)2 was 2% to consider the cost of pretreatment with straw. The research results provide technical support for pretreatment of anaerobic fermentation of lingo-cellulosic materials such as straw.

Effect of biomass pretreatment on the product distribution and composition resulting from the hydrothermal liquefaction of short rotation coppice willow

A major challenge for the implementation of hydrothermal liquefaction (HTL) as a continuous process is the formulation of lignocellulosic feedstock, which is prone to phase separation into water and biomass parts when pressurized. One approach to remedy such phase separation is to reduce the dry matter content; however, as this approach is detrimental to process cost efficiency, designing an appropriate pretreatment step to ensure pumpability at high dry matter content is preferable. This paper evaluated the effect of various pretreatment methods on product distribution and composition resulting from the HTL of willow and proposes short rotation coppice as an alternative biomass feedstock for biofuels production. Alkaline–thermal pretreatment, besides making high dry matter pumpable feedstock slurries, also led to an increase in the production of the bio-crude product with an oxygen content lower than 8wt% and a higher concentration of aromatics and phenolic compounds.

Influence of Alkaline-Thermal Pretreatment on High-Solids Anaerobic Digestion of Dewatered Activated Sludge

The influence of alkaline (23.77 mg NaOH/g total solid), thermal (134.95 °C), and combined alkaline-thermal pretreatment on parameters of dewatered activated sludge (DAS) during high-solids anaerobic digestion was investigated. Soluble chemical oxygen demand (SCOD), soluble proteins, and VFAs (volatile fatty acids) concentrations were significantly higher (by two-fold) in pretreated DAS samples than in the control. During subsequent anaerobic digestion, the concentrations first increased and then decreased. Total chemical oxygen demand (TCOD) decreased by 28.40% to 40.92%, and ammonia nitrogen accumulated during anaerobic digestion. For the alkaline, thermal, and alkaline-thermal pretreatments, daily methane yield significantly increased in mid-anaerobic digestion and cumulative methane yield (CMY) increased by 9.92, 35.25, and 52.95%, respectively, relative to the control. There were clear synergistic effects of alkaline-thermal pretreatment, resulting in a 17.20% increase in CMY compared with the sum of the separate alkaline and thermal pretreatments. Therefore, alkaline-thermal pretreatment was helpful for high-solids anaerobic digestion of DAS and was an effective pretreatment method. Gompertz model fitting to the CMY curve produced determination coefficients (R2) greater than 0.9931 for all pretreatments, which was better than for a first-order kinetic model fitting curve.

Alkaline thermal pretreatment at mild temperatures for biogas production from anaerobic digestion of antibiotic mycelial residue

This paper aims at lowering the temperature for thermal pretreatment (TPT) of antibiotic mycelial residue (AMR) by alkali addition but without significantly worsening subsequent anaerobic digestion (AD) for biogas. Batch TPT and AD experiments were conducted in a bench-scale autoclave and several bench-scale anaerobic digesters, respectively. The results showed that the methane yield (<200ml·(gVS)−1) was visibly lower with lowering pretreatment temperature, compared to that (290ml·(gVS)−1) for TPT at the optimal temperature of 120°C, while it rebounded to 231ml·(gVS)−1 when proper amounts of alkali were employed (to adjust the pH of the AMR to 12) for TPT at 80°C. Further analysis indicated that low-temperature alkaline TPT was significantly less energy-consumption compared to only TPT, at cost of small amounts of alkali. It was more convenient and economical to implement AD of AMR in combination with alkaline TPT at mild temperatures for biogas.
