Kaempferol stimulates WNT/β-catenin signaling pathway to induce differentiation of osteoblasts

Flavonoids, a group of natural compounds found in a variety of vegetables and herbal medicines, have been intensively reported on stimulating bone mineral density and bone formation. Among them, kaempferol has been reported to assist bone formation in vitro and in vivo, but its precise mechanism of action for stimulating bone forming abilities of osteoblasts remained elusive. In SaOS-2 osteoblasts, treatment of kaempferol increased early and late osteogenic parameters significantly, including alkaline phosphatase (ALP) activity, collagen synthesis, and mRNA expression levels of Runx2, osterix, osteopontin and bone sialoprotein. Interestingly, kaempferol promoted osteoblastic differentiation via the activation of the WNT signaling pathway. The stimulation of SaOS-2 cells by kaempferol resulted in an increased activity of WNT signaling responsive reporter construct, Axin-2, and, subsequently, stabilization of WNT signaling mediated transcription factor β-catenin, probably leading to the activation of WNT-targeted genes for osteogenesis. In corroboration, the kaempferol-induced ALP activity was fully abolished by FH 535, an inhibitor of WNT signaling pathway. Kaempferol mediated activation of WNT signaling pathway through estrogen signaling pathway, as the application of ICI 182,780 (an inhibitor for estrogen receptors) markedly inhibited kaempferol-induced WNT signaling activation and osteogenic marker like ALP activity in SaOS-2 cells. Immunohistochemical studies in drill-hole defect model showed increased expression of Runx2 and β-catenin staining after kaempferol treatment. Thus, it may be concluded that kaempferol stimulates estrogen signaling followed by WNT signaling pathway activation to achieve its potential for bone-sparing effects.

Biocompatibility and mineralized nodule formation of Neo MTA Plus and an experimental tricalcium silicate cement containing tantalum oxide.

To evaluate the biocompatibility and mineralized nodule formation of an experimental tricalcium silicate cement with tantalum oxide (TSC/Ta2 O5 ) as radiopacifier, Neo MTA Plus (Avalon Biomed Inc., Bradenton, FL, USA) and MTA (Angelus, Londrina, PR, Brazil) on human osteoblast-like cells (Saos-2). Biocompatibility was evaluated by 3-(4,5-dimethyl-thiazoyl)-2,5-diphenyl-tetrazolium bromide (MTT) and neutral red (NR) assays, after exposure of Saos-2 to cement extracts at 1:1, 1:2, 1:4 and 1:8 dilutions for 24h. Bioactivity was evaluated by alkaline phosphatase (ALP) activity, and calcium deposits were detected with alizarin red staining (ARS). Statistical analysis was performed with analysis of variance and Bonferroni or Tukey post-test (α=0.05). The MTT assay revealed lower cytotoxicity for NEO and MTA (P<0.05), and higher for TSC/Ta2 O5 at 1:1 and 1:2 dilutions when compared to serum-free medium - control (P>0.05). At 1:4 dilution, the TSC/Ta2 O5 cytotoxicity was similar to the control (P>0.05). At 1:8 dilution, cell viability was significantly greater than the control (P<0.05). Saos-2 cell viability performed using the NR assay at all dilutions revealed no cytotoxic effect of MTA, NEO and TSC/Ta2 O5 . ALP activity at 1 and 3days was similar to the control (P>0.05). TSC/Ta2 O5 had significantly greater ALP activity at 7days when compared with the control (P<0.05). All materials induced the production of mineralized nodules, and NEO produced significantly more mineralized nodules than MTA and TSC/Ta2 O5 (P<0.05). Neo MTA Plus and TSC/Ta2 O5 were biocompatible and induced ALP activity in Saos-2 cells. Both materials induced mineralized nodule formation by Saos-2 with Neo MTA Plus producing significantly more.

Effect of low level laser therapy and zoledronate on the viability and ALP activity of Saos-2 cells

A limited number of clinical studies indicate the supportive role of low level laser therapy (LLLT) on medical and/or surgical approaches carried out in treatment modalities for bisphosphonate related necrosis of jaws (BRONJ), the most common side effect of bisphosphonates used to inhibit bone resorption. The purpose of this study was to investigate the effects of LLLT on cell proliferation and alkaline phosphatase (ALP) activity of human osteoblast-like cells (Saos-2) treated with different doses of zoledronate, the most potent bisphosphonate. Saos-2 cells were treated with different concentrations of zoledronate and were irradiated with diode laser (wavelength 808nm, 10s, 0.25 or 0.50W). Cell numbers and ALP activity of the cells were determined. LLLT mildly increased the proliferation rate or ALP activity, while zoledronate reduced both. When applied together, LLLT lessened the detrimental effects of zoledronate and improved cell function and/or proliferation. Based on the results of this study, it was concluded that LLLT has biostimulative effects on Saos-2 cells, even after treatment with zoledronate. LLLT may serve as a useful supportive method for BRONJ treatment through enhancement of healing by osteoblasts.

The effects of pulsed electromagnetic fields on the cellular activity of SaOS‐2 cells

Although pulsed electromagnetic fields (PEMFs) have been used for treatments of nonunion bone fracture healing for more than three decades, the underlying cellular mechanism of bone formation promoted by PEMFs is still unclear. It has been observed that a series of parameters such as pulse shape and frequency should be carefully controlled to achieve effective treatments. In this article, the effects of PEMFs with repetitive pulse burst waveform on the cellular activity of SaOS-2 osteoblast-like cells were investigated. In particular, cell proliferation and mineralization due to the imposed PEMFs were assessed through direct cell counts, the MTT assay, tissue nonspecific alkaline phosphatase (ALP) and Alizarin Red S (ARS) staining. PEMF stimulation with repetitive pulse burst waveform did not affect metabolic activity and cell number. However, the ALP activity of SaOS-2 cells and mineral nodule formation increased significantly after PEMF stimulation. These observations suggest that repetitive pulse burst PEMF does not affect cellular metabolism; however, it may play a role in the enhancement of SaOS-2 cell mineralization. We are currently investigating cellular responses under different PEMF waveforms and Western blots for protein expression of bone mineralization specific proteins.

Fabrication of calcium phosphate films for coating on titanium substrates heated up to 773 K by RF magnetron sputtering and their evaluations

Calcium phosphate films were fabricated on titanium substrates heated up to 773 K using radiofrequency (RF) magnetron sputtering. The deposition rate, phase and preferred orientation of the calcium phosphate films were studied. Immersion tests for the films were conducted using Hanks' solution and PBS(−), and the surface reactions on the specimens coated with the calcium phosphate films were investigated. The bonding strength between the coating films and the titanium substrates before and after the immersion tests was evaluated; the bonding strength decreased after the immersion tests. The alkaline phosphatase (ALP) activity of SaOS-2 cells on a titanium plate coated with a calcium phosphate film was examined by conducting a culture test. Calcium phosphate coating increased the ALP activity of SaOS-2 cells cultured for 3 and 7 days. Titanium cylinders were coated with an amorphous calcium phosphate film and implanted into the mandibles of beagle dogs. An increase in the extent of bone-implant contact for the coated titanium cylinders was confirmed 8 to 12 weeks after implantation and compared with the case for uncoated titanium cylinders.

Evaluation of Calcium Phosphate Coating Films on Titanium Fabricated Using RF Magnetron Sputtering

Calcium phosphate coating films fabricated on commercially pure titanium (CP-Ti) substrates using radiofrequency (RF) magnetron sputtering were evaluated in vivo and in vitro for investigating their applications in dental and medical implants. For the in vitro evaluations of the calcium phosphate coating films, the bonding strength and alkaline phosphatase (ALP) activity were examined. The bonding strength of the coating films to a polished titanium plate exceeded 60 MPa. When compared with an uncoated titanium plate, the increase in the ALP activity of SaOS-2 cells (a well-characterized osteosarcoma human cell line exhibiting osteoblast-like properties) on a titanium plate coated with a calcium phosphate film was confirmed by a culture test. Titanium cylinders coated with an amorphous calcium phosphate film were implanted into the mandibles of beagle dogs. The percentage of bone-implant contact in coated titanium was greater than that in uncoated titanium.

Phosphate regulates the stability of skeletal alkaline phosphatase activity in human osteosarcoma (SaOS-2) cells without equivalent effects on the level of skeletal alkaline phosphatase immunoreactive protein.

Inorganic phosphate (P(i)) can regulate the level of skeletal alkaline phosphatase (ALP) activity in human osteoblast-like cells, but not by means of changes in transcription or release from the cell surface. The current studies were intended to determine whether (1) P(i) affected the inactivation of ALP activity in human osteosarcoma (SaOS-2) cells; and (2) P(i)-dependent changes in ALP-specific activity were associated with equal, concomitant changes in the level of ALP immunoreactive protein. The results of these studies revealed that P(i) increased the stability of skeletal ALP activity without equivalent effects on the level of ALP immunoreactive protein. An increase in P(i) (from 0 to 1.8 mmol/liter) caused a time-dependent increase in the amount of skeletal ALP activity in the SaOS-2 cells, without a parallel increase in the amount of skeletal ALP immunoreactive protein, and a decrease in P(i) (from 1.8 to 0 mmol/liter) caused a time-dependent decrease in the amount of ALP activity, without a significant decrease in the total cellular content of ALP immunoreactive protein. Together, these observations suggest that P(i) may alter the level of skeletal ALP activity in SaOS-2 cells by inhibiting a process of irreversible inactivation that does not effect equal, concomitant changes in the level of skeletal ALP immunoreactive protein.

Skeletal alkaline phosphatase specific activity is an index of the osteoblastic phenotype in subpopulations of the human osteosarcoma cell line SaOS-2

During continuous culture with serial passage, the human osteosarcoma cell line SaOS-2 showed a time-dependent decrease in skeletal alkaline phosphatase (ALP) activity. Because this was indicative of heterogeneity, subpopulations of SaOS-2 cells were isolated from replicate low-density cultures. The subpopulations were less heterogeneous and more stable (with respect to ALP) than the parent population. ALP specific activity in the subpopulations ranged from 0.05 to 2.3 U/mg protein, and cytochemical analyses indicated multiple steady-state levels of ALP activity per cell. The amount of ALP activity in SaOS-2 subpopulations was proportional to collagen production ([ 3H]proline incorporation into collagenase-digestible protein; r = .84, P < .005), and to parathyroid hormone (PTH)-linked synthesis of cyclic adenosine monophosphate (cAMP) ( r = .88, P < .01). From these data, we inferred that ALP activity in SaOS-2 cells can provide a useful index of the osteoblastic phenotype, and that ALP activity, collagen production, and PTH-linked adenylate cyclase were coordinately regulated in these osteoblast-like osteosarcoma cells (ie, selection of subpopulations for ALP activity coselected for collagen synthesis and PTH-linked synthesis of cAMP). Further comparative studies showed that micromolar fluoride concentrations stimulated cell proliferation ([ 3H]thymidine incorporation into DNA) in low-ALP SaOS-2 subpopulations, but not in high-ALP cells ( P < .001), and that this differential sensitivity to fluoride was associated with an inverse correlation between fluoride-sensitive acid phosphatase and ALP activities ( r = −.91, P < .001).
