Proximal convoluted tubule cells-derived exosomes attenuate aging-related vascular dysfunction

Aging is an identified risk factor for cardiovascular disease. Aging is associated with increased prevalence of endothelial dysfunction and arterial stiffening. Exosomes are bi-lipid nanoscale vesicles that play important roles in intercellular communications. The purpose of this study was to determine whether administration of renal proximal convoluted tubule cells (PCT)-derived exosomes affects vascular and renal function in aging mice. Aging (24 months old) and young mice (3 months old) were treated with PCT-derived exosomes via non-invasive injection into the kidney 3 times a week for 2 weeks. Administration of PCT-derived exosomes did not affect blood pressure or urinary Na+, K+, and Ca2+ excretions, indicating that PCT-derived exosomes did not affect kidney function. Pulse wave velocity (PWV) was increased in aging mice, indicating arterial stiffening. Interestingly, treatment with PCT-exosomes effectively reduced aging-associated arterial stiffening. In addition, aortic endothelium-dependent relaxing (EDR) responses to acetylcholine (Ach) were reduced in aging mice, indicating endothelial dysfunction. Treatment with PCT-derived exosomes rescued aging-associated aortic endothelial dysfunction. Mechanistically, aortic expression of VCAM1, ICAM1, TNFα, and MCP-1 was upregulated in aging mice, indicating vascular inflammation. Notably, aging-associated vascular inflammation was largely attenuated by delivery of PCT-derived exosomes. Our results suggest that PCT-derived exosomes improves aging-associated arterial stiffening and vascular dysfunction partly via inhibiting vascular inflammation. The findings may help develop novel therapeutic strategies for aging-associated vascular dysfunction using PCT-derived exosomes. Immunity in the Progeroid Model of Cockayne Syndrome: Biomarkers of Pathological Aging.

Cockayne syndrome (CS) is a rare autosomal recessive disorder that affects the DNA repair process. It is a progeroid syndrome predisposing patients to accelerated aging and to increased susceptibility to respiratory infections. Here, we studied the immune status of CS patients to determine potential biomarkers associated with pathological aging. CS patients, as well as elderly and young, healthy donors, were enrolled in this study. Complete blood counts for patients and donors were assessed, immune cell subsets were analyzed using flow cytometry, and candidate cytokines were analyzed via multi-analyte ELISArray kits. In CS patients, we noticed a high percentage of lymphocytes, an increased rate of intermediate and non-classical monocytes, and a high level of pro-inflammatory cytokine IL-8. In addition, we identified an increased rate of particular subtypes of T Lymphocyte CD8+ CD28- CD27-, which are senescent T cells. Thus, an inflammatory state was found in CS patients that is similar to that observed in the elderly donors and is associated with an immunosenescence status in both groups. This could explain the CS patients' increased susceptibility to infections, which is partly due to an aging-associated inflammation process.

Inflammatory Signaling Mediated By Gasdermin D Underlies Tet2-Mutant Clonal Hematopoiesis-Associated Tissue Dysfunction

Aging is characterized by the progressive accumulation of somatic mutations. Acquisition of certain somatic mutations in hematopoietic stem cells (HSCs) confers on them a fitness advantage enabling them to outcompete other HSCs by clonal expansion in a phenomenon called clonal hematopoiesis (CH). CH is associated with a 40% increase in all-cause mortality linked to elevated risk for aging-associated morbidities such as cardiovascular disease (CVD), chronic kidney disease, COPD, and chronic liver disease. However, the mechanism by which CH exerts its adverse effects on disparate tissues is poorly understood. Here, we use CH-associated cardiac dysfunction as a paradigm to uncover general principles by which CH leads to the decline of tissue function. CH was modeled in mice by transplanting whole bone marrow (WBM) cells deficient for Tet2, a frequently mutated CH gene, into irradiated WT mice. We observed an elevated incidence of atrial fibrillation (AF) in Tet2 -/- WBM recipients relative to WT WBM recipients without any surgical or dietary intervention (Fig. A). The Tet2 -/- WBM recipients also had reduced left ventricular ejection fraction (LVEF), an indicator of heart failure. Mice exhibiting elevated incidence of AF and reduced LVEF are defined as having cardiac dysfunction. In addition, we observed that Tet2 -/- WBM recipients had increased systemic inflammation compared to controls, as evidenced by elevated levels of proinflammatory cytokines in the serum. Furthermore, their cardiac tissue exhibited upregulation of the inflammasome signaling pathway including the activation of Gasdermin D, the effector of pyroptotic cell death. Inflammation in the cardiac tissue was concomitantly associated with increased cardiac fibrosis in Tet2 -/- WBM recipients. Since inflammation is a known driver of CVD, we hypothesized that abrogating Gasdermin D-mediated inflammatory signaling could ameliorate CH-induced cardiac dysfunction. Indeed, Gsdmd -/- recipient mice were protected from cardiac dysfunction upon transplantation with Tet2 -/-WBM cells (Fig. A). Importantly, expansion of mutant hematopoietic cells was also mitigated in the absence of GSDMD: transplantation of Tet2 -/- WBM cells along with either WT or Gsdmd -/- WBM cells revealed that GSDMD deficiency restrained the rate of clonal expansion of Tet2 -/- cells (Fig. B). This suggests that instead of being mere bystanders, WT cells in the environment may play an active role in regulating the rate of mutant cell expansion via intercellular communication mediated by GSDMD. Identification of proinflammatory factors mediating clonal expansion and cardiac dysfunction is of significant interest for potential therapeutic intervention. Thus, we sought to determine factors that were upregulated in CH mouse models while being downregulated in Gsdmd -/- mice. Luminex multiplexed assay enabled us to identify several understudied proinflammatory cytokines present in serum satisfying the above criteria. In addition to elevated levels in the serum, some of these cytokines were upregulated in the cardiac tissue of Tet2 -/- WBM recipient mice but not in WT WBM recipients. Furthermore, some are reported to induce inflammasome signaling. CH, even in the absence of known drivers, is associated with higher CVD-associated mortality, suggesting the existence of pathways common to multiple forms of CH; systemic inflammation is one potential common mechanism. We tested whether aged WT WBM, which has low levels of mutant clone expansion but exhibits several features of CH, could cause cardiac dysfunction. We transplanted either aged or young WT WBM into young WT recipients and found that the aged WT WBM recipients exhibited elevated AF and reduced LVEF. These data suggest that aging-associated systemic inflammation may be sufficient for cardiac dysfunction. Importantly, one of the cytokines we uncovered was also upregulated in aged WT mice and has been associated with inflammation and fibrosis in multiple tissues. This cytokine is being further investigated as a top candidate for therapeutic intervention since it is a common feature of CH with and without identified drivers. Overall, our findings demonstrate the critical role of Gasdermin D-mediated inflammatory signaling in the pathogenesis of Tet2-mutant CH-induced cardiac dysfunction. We have also uncovered a cytokine that has emerged as a promising therapeutic target for CH-associated pathologies.

Mitochondrial OGG1 expression reduces age-associated neuroinflammation by regulating cytosolic mitochondrial DNA.

Aging is accompanied by a decline in DNA repair efficiency, which leads to the accumulation of different types of DNA damage. Age-associated chronic inflammation and generation of reactive oxygen species exacerbate the aging process and age-related chronic disorders. These inflammatory processes establish conditions that favor accumulation of DNA base damage, especially 8-oxo-7,8 di-hydroguanine (8-oxoG), which in turn contributes to various age associated diseases. 8-oxoG is repaired by 8-oxoG glycosylase1 (OGG1) through the base excision repair (BER) pathway. OGG1 is present in both the cell nucleus and in mitochondria. Mitochondrial OGG1 has been implicated in mitochondrial DNA repair and increased mitochondrial function. Using transgenic mouse models and cell lines that have been engineered to have enhanced expression of mitochondria-targeted OGG1 (mtOGG1), we show that elevated levels of mtOGG1 in mitochondria can reverse aging-associated inflammation and improve functions. Old male mtOGG1Tg mice show decreased inflammation response, decreased TNFα levels and multiple pro-inflammatory cytokines. Moreover, we observe that male mtOGG1Tg mice show resistance to STING activation. Interestingly, female mtOGG1Tg mice did not respond to mtOGG1 overexpression. Further, HMC3 cells expressing mtOGG1 display decreased release of mtDNA into the cytoplasm after lipopolysacchride induction and regulate inflammation through the pSTING pathway. Also, increased mtOGG1 expression reduced LPS-induced loss of mitochondrial functions. These results suggest that mtOGG1 regulates age-associated inflammation by controlling release of mtDNA into the cytoplasm.

Aging aggravates acetaminophen-induced acute liver injury and inflammation through inordinate C/EBPα-BMP9 crosstalk

BackgroundPrevious studies have shown that bone morphogenetic protein 9 (BMP9) is almost exclusively produced in the liver and reaches tissues throughout the body as a secreted protein. However, the mechanism of BMP9 action and its role in aging-associated liver injury and inflammation are still unclear.ResultsAging significantly aggravates acetaminophen (APAP)-induced acute liver injury (ALI). Increased expression of CCAAT/enhancer binding protein α (C/EBPα) and BMP9 was identified in aged livers and in hepatocytes and macrophages (MФs) isolated from aged mice. Further analysis revealed that excess BMP9 was directly related to APAP-induced hepatocyte injury and death, as evidenced by activated drosophila mothers against decapentaplegic protein 1/5/9 (SMAD1/5/9) signaling, an increased dead cell/total cell ratio, decreased levels of ATG3 and ATG7, blocked autophagy, increased senescence‐associated beta‐galactosidase (SA‐β‐Gal) activity, and a higher rate of senescence‐associated secretory phenotype (SASP) acquisition. In contrast, Bmp9 knockout (Bmp9−/−) partially alleviated the aforementioned manifestations of BMP9 overexpression. Moreover, BMP9 expression was found to be regulated by C/EBPα in vitro and in vivo. Notably, BMP9 also downregulated autophagy through its effect on autophagy-related genes (ATG3 and ATG7) in MΦs, which was associated with aggravated liver injury and SASP acquisition.ConclusionsIn summary, the present study highlights the crucial roles played by C/EBPα-BMP9 crosstalk and provides insights into the interrelationship between hepatocytes and MΦs during acute liver injury.

PS-BPB10-5: POSSIBLE INVOLVEMENT OF RENAL TUBULAR NFAT5 IN AGING-ASSOCIATED RENAL DYSFUNCTION AND SALT-SENSITIVE HYPERTENSION

Objective: Japan is currently facing a super-aging society, where an incidence of lifestyle-related diseases has become a greater social burden. The number of patients with chronic kidney disease (CKD) and hypertension is being increased with age; however, it is not clearly defined why and how aging causes renal dysfunction and hypertension. Nuclear factor of activated T-cells 5 (NFAT5) is a transcription factor that is activated upon hypertonic conditions as observed in the renal medulla. We have shown that the renal tubular cell-specific NFAT5 conditional knockout (KO) mice exhibit salt-sensitive hypertension, while the mice exhibit impaired urine concentrating ability and are susceptible to renal fibrosis. These phenotypes resemble aging-associated renal dysfunction, i.e., urine concentrating disorder, salt-sensitive hypertension, and renal fibrosis. We therefore investigated the possible involvement of NFAT5 in aging-related changes of the kidney. Methods: Male wild-type (WT) and KO mice (12 weeks old) were subjected to unilateral ureteral obstruction (UUO) or sham operation. After 7 days, mice were sacrificed and kidneys were removed for analysis. Renal fibrosis was evaluated by AZAN staining. Senescence-associated beta-galactosidase (SA-β;-Gal) activity was examined by fluorescence study in the whole kidney slices. mRNA expression for senescence-associated secretory phenotype (SASP)-related factors (TGF-β;1, COL1A1, ICAM1, PAI-1, p21, p16, and p53) in the whole kidney tissue was examined by real-time PCR. Results: AZAN staining showed that UUO induced renal fibrosis both in WT and KO mice. The UUO-induced fibrotic area was significantly larger in KO mice than in WT mice in the medulla. In sham-operated mice, overall SA-β;-Gal activity was larger in KO mice, in which the activity was dominant in the corticomedullary region. In UUO groups, overall SA-β;-Gal activity was low compared to sham-operated groups. The mRNA expression of SASP-related factors was not different between WT and KO mice in sham-operated groups. UUO upregulated the mRNA expression of all SASP-related factors both in WT and KO mice. In UUO groups, the expression of TGF-β;1, COL1A1, ICAM1, and PAI-1 was significantly larger in KO mice than in WT mice. The expression of p21, p16, and p53 tended to be larger in KO mice than in WT mice. Conclusions: Renal tubular NFAT5 deficiency showed senescence-related phenotypes at baseline. Deficit of renal tubular NFAT5 may further induce cell senescence upon insult, which might accelerate aging-associated renal inflammation and dysfunction in CKD.

Aging impairs arterial compliance via Klotho-mediated downregulation of B-cell population and IgG levels.

Aging is associated with compromised immune function and arterial remodeling and stiffness. The purpose of this study is to investigate whether in vivo AAV-based delivery of secreted Klotho (SKL) gene (AAV-SKL) improves aging- and senescence-associated immune dysfunction and arterial stiffness. Senescence-accelerated mice prone strain 1 (SAMP1, 10months) and old mice (20months) were used. Serum SKL levels, B-cell population and serum IgG levels were markedly decreased in SAMP1 and old mice. Rescue of downregulation of serum SKL levels by in vivo AAV2-based delivery of SKL gene (AAV-SKL) increased B-cell population and serum IgG levels and attenuated arterial stiffness in SAMP1 and old mice. Thus, Klotho deficiency may play a role in senescence- and aging-associated humoral immune dysfunction and arterial stiffness. Vascular infiltration of inflammatory cells and expression of TGFβ1, collagen 1, scleraxis, MMP-2 and MMP-9 were increased while the elastin level was decreased in aortas of SAMP1 and old mice which can be rescued by AAV-SKL. Interestingly, treatment with IgG effectively rescued arterial inflammation and remodeling and attenuated arterial stiffness and hypertension in aging mice. In cultured B-lymphoblast cells, we further showed that SKL regulates B-cell proliferation and maturation partly via the NFkB pathway. Aging-associated arterial stiffening may be largely attributed to downregulation of B-cell population and serum IgG levels. AAV-SKL attenuates arterial stiffness in aging mice partly via restoring B-cell population and serum IgG levels which attenuates aging-associated vascular inflammation and arterial remodeling.

The biological mechanisms of metformin effects on aging-associated inflammation in obesity

Abstract Obesity prevalence within the 60+ years-old (yrs) population is drastically increasing. This alarmingly high rate of obesity compels comparative studies to identify common players involved in aging-derived and obesity-associated inflammation. Blood CD4+ T cells from lean older humans show an upregulated Th17 signature accompanied by a defective redox system. Treatment of these cells with the anti-diabetic drug metformin (100μM) promoted a more young-like anti-inflammatory/antioxidant phenotype. Here, we investigate whether the protective effects of metformin observed in CD4+ T cells from lean aging adults persist in obesity. Blood CD4+ T cells from obese older (60+ yrs) donors secrete twice the amount of the Th17 cytokines IL-17F and IL-21 when compared to their younger counterparts (25–35 yrs). Glutamate production, which supports Th17 cytokines secretion, is 1.5-times higher in cells from older compared to younger subjects. Mitochondrial superoxide production in these cells was 25% higher than in younger cells, with no changes in total peroxide. Contrary to findings from cells of lean subjects, metformin failed to reduce Th17 cytokines in cells from obese older subjects, perhaps due to metformin’s inability to reduce glutamate production. However, metformin reduced hydrogen peroxide production 1.75-fold, with no effect on superoxide, in cells from obese older subjects. These results collectively indicate that 1) obesity-associated upregulation of Th17 cytokines is mechanistically linked to glutamine metabolism and mitochondrial superoxide production and 2) metformin’s action targets peroxide production rather than superoxide, which is insufficient for reducing age-associated inflammation in obesity. Cordycepin from Medicinal Fungi Cordyceps militaris Mitigates Inflammaging-Associated Testicular Damage via Regulating NF-κB/MAPKs Signaling in Naturally Aged Rats

Inflammaging in male reproductive organs covers a wide variety of problems, including sexual dysfunction and infertility. In this study, the beneficial effects of cordycepin (COR), isolated from potential medicinal fungi Cordyceps militaris, in aging-associated testicular inflammation and serum biochemical changes in naturally aged rats were investigated. Male Sprague Dawley rats were divided into young control (YC), aged control (AC), and COR (5, 10, and 20 mg/kg) treated aged rat groups. Aging-associated serum biochemical changes and inflammatory parameters were analyzed by biochemical assay kits, Western blotting, and real-time RT-PCR. Results showed a significant (p < 0.05) alteration in the total blood cell count, lipid metabolism, and liver functional parameters in AC group when compared with YC group. However, COR-treated aged rats ameliorated the altered biochemical parameters significantly (p < 0.05 and p < 0.01 at 5, 10, and 20 mg/kg, respectively). Furthermore, the increase in the expression of inflammatory mediators (COX-2, interleukin (IL)-6, IL-1β, and tissue necrosis factor-alpha) in aged rat testis was significant (p < 0.05) when compared with YC group. Treatment with COR at 20 mg/kg to aged rats attenuated the increased expression of inflammatory mediators significantly (p < 0.05). Mechanistic studies revealed that the potential attenuating effects exhibited by COR in aged rats was mediated by regulation of NF-κB activation and MAPKs (c-Jun N-terminal kinase, extracellular signal-regulated kinase 1/2, and p38) signaling. In conclusion, COR restored the altered serum biochemical parameters in aged rats and ameliorated the aging-associated testicular inflammation proving the therapeutic benefits of COR targeting inflammaging-associated male sexual dysfunctions.

Differential Association of Viral Dynamics With Disease Severity Depending on Patients' Age Group in COVID-19.

Despite a clear association of patient’s age with COVID-19 severity, there has been conflicting data on the association of viral load with disease severity. Here, we investigated the association of viral load dynamics with patient’s age and severity of COVID-19 using a set of respiratory specimens longitudinally collected (mean: 4.8 times/patient) from 64 patients with broad distribution of clinical severity and age during acute phase. Higher viral burden was positively associated with inflammatory responses, as assessed by IL-6, C-reactive protein, and lactate dehydrogenase levels in patients’ plasma collected on the same day, primarily in the younger cohort (≤59 years old) and in mild cases of all ages, whereas these were barely detectable in elderly patients (≥60 years old) with critical disease. In addition, viral load dynamics in elderly patients were not significantly different between mild and critical cases, even though more enhanced inflammation was consistently observed in the elderly group when compared to the younger group during the acute phase of infection. The positive correlation of viral load with disease severity in younger patients may explain the increased therapeutic responsiveness to current antiviral drugs and neutralizing antibody therapies in younger patients compared to elderly patients. More careful intervention against aging-associated inflammation might be required to mitigate severe disease progression and reduce fatality in COVID-19 patients more than 60 years old.

Effects of volume-matched resistance training with different loads on glycemic control, inflammation, and body composition in prediabetic older adults.

The purpose of the investigation was to examine the influence of resistance training (RT) with equal volume and varying load on glycemic control, inflammation, and body composition in non-obese prediabetic older adults. Non-obese older adults with prediabetes were randomized into 2 groups, high-load (80% of 1RM) and low-load (40% of 1RM) RT (n = 12/group), both with the same training volume. Oral glucose tolerance test (OGTT) and blood samples were collected at baseline and again after 10weeks of RT. Fasting blood glucose (103.8 vs. 99.9mg/dL) and the area under the curve (AUC) of OGTT (0-30min) decreased significantly in older adults with prediabetes after 10weeks of volume-matched RT (p< 0.05). Serum levels of MCP-1 (138.7 vs. 98.5 pg/mL) and TNF-α (1.8 vs. 1.3 pg/mL) showed significant decrease after 10 weeks of high-load RT (p< 0.05). There were no changes in IL-10, IL-6, and CRP levels in both groups. Leptin showed significant decrease after 10weeks of low-load RT (p< 0.05). Changes in fasting glucose and AUC of OGTT (0-120min) were positively correlated with changes in MCP-1 and TNF-α (p< 0.05). Lean body mass (39.6 vs. 40.3kg) increased significantly after 10weeks of volume-matched RT (p< 0.05). Results indicate that equal-volume RT at different loads is beneficial to glycemic control and muscle growth, and high-load RT shows more prominent anti-inflammatory effects. Novelty: Short-term high-load resistance training can help older adults bring their blood sugar level back to normal. High-load resistance training attenuates aging-associated chronic inflammation.

Oxidative Stress and Mitochondrial Damage in Dry Age-Related Macular Degeneration Like NFE2L2/PGC-1α-/- Mouse Model Evoke Complement Component C5a Independent of C3.

Simple SummaryAge-related macular degeneration (AMD) is an eye disease that results in permanent loss of vision due to degeneration in the central portion of the retina called the macula. Patients with severe visual loss have reduced quality of life and the risk of death is 2.4 times higher than the general population. Currently, there is no treatment to stop or cure dry AMD. Aging-associated chronic oxidative stress and inflammation are known to be involved in AMD pathology. To investigate the molecular mechanism behind the cause and to develop novel therapy, we have created and validated an animal model mimicking clinical features of dry AMD. Here, we show previously unknown thrombin-mediated complement component C5a activation in the degenerative retina without upregulation of C3. Our model might provide insight into AMD progression and help to develop novel therapies.Aging-associated chronic oxidative stress and inflammation are known to be involved in various diseases, e.g., age-related macular degeneration (AMD). Previously, we reported the presence of dry AMD-like signs, such as elevated oxidative stress, dysfunctional mitophagy and the accumulation of detrimental oxidized materials in the retinal pigment epithelial (RPE) cells of nuclear factor erythroid 2-related factor 2, and a peroxisome proliferator-activated receptor gamma coactivator 1-alpha (NFE2L2/PGC1α) double knockout (dKO) mouse model. Here, we investigated the dynamics of inflammatory markers in one-year-old NFE2L2/PGC1α dKO mice. Immunohistochemical analysis revealed an increase in levels of Toll-like receptors 3 and 9, while those of NOD-like receptor 3 were decreased in NFE2L2/PGC1α dKO retinal specimens as compared to wild type animals. Further analysis showed a trend towards an increase in complement component C5a independent of component C3, observed to be tightly regulated by complement factor H. Interestingly, we found that thrombin, a serine protease enzyme, was involved in enhancing the terminal pathway producing C5a, independent of C3. We also detected an increase in primary acute phase C-reactive protein and receptor for advanced glycation end products in NFE2L2/PGC1α dKO retina. Our main data show C5 and thrombin upregulation together with decreased C3 levels in this dry AMD-like model. In general, the retina strives to mount an orchestrated inflammatory response while attempting to maintain tissue homeostasis and resolve inflammation.

Aging-Associated Inflammation Due to Elevated Levels of TNF-α Increases Activation of the Integrin αIIbβ3 and the Procoagulant Potential of Platelets

Background: Aging is an independent risk factor for thrombosis and the molecular mechanisms underpinning this higher thrombosis risk in aging remains largely unknown. Chronic inflammation due to elevated levels of TNF-α contributes to the pathobiology of aging. Nevertheless, whether increased TNF-α levels during aging alter platelet function to promote thrombosis remains unexamined.Aims: To investigate the mechanism(s) by which aging-associated inflammation by TNF-α may promote platelet hyperreactivity in mice and humans.Methods: We analyzed megakaryocyte maturation, platelet number and activation indices by FACS, light transmission aggregometry (LTA) and a microfluidic assay in the following murine models of aging: 1) aged C57BL/6J mice (>18 months old, where circulating TNF-α levels are high); 2) young mice (~8 weeks old) with constitutive overexpression of TNF-α (TNFΔARE) ; and 3) young C57BL/6J mice injected in vivo with TNF-α or IL1b (for up to 20 days). In select experiments, we also neutralized TNF-α in vivo in aged mice. To determine if murine findings were recapitulated in humans, we examined platelet integrin activation in younger and older adults. Finally, to establish whether inflammation alters the molecular signature of megakaryocytes (MKs-the platelet parent cell) during aging, we performed next-generation RNA-sequencing (both bulk and single-cell) in bone marrow MKs from young and aged mice.Results: Aged mice exhibit thrombocytosis and accelerated megakaryocyte maturation as determined by ploidy analysis. In addition, platelets from aged mice, TNFΔARE mice, and from young mice injected with TNF-α exposed significantly higher amounts of membrane phosphatidylserine and activated integrin αIIbβ3 faster after stimulation with thrombin and convulxin. Treating mice in vivo with IL1b did not recapitulate these effects. Moreover, platelets from old mice adhered faster to collagen and formed larger clots under flow when assessed in a microfluidic assay using reconstituted whole blood (pooled plasma and erythrocytes in addition to platelets from young or old mice). Similarly, integrin αIIbβ3 activation was significantly greater in activated platelets from older adults (mean age 78.0 ± 8.4) compared to younger adults (mean age 35.0 ± 10.1). Interestingly, platelets from old mice injected with an anti-TNF-α antibody had a markedly decreased activation profile. Bulk RNA-seq analysis of native megakaryocytes from aged mice and from young mice treated with TNF-⍺ both had a robust TNF-⍺ transcriptional signature. Analyses of single cell RNA-seq of bone marrow MKs demonstrated dysregulation of the mTOR and mitochondrial dependent pathways in aged mice. These transcriptional findings indicate there is MK reprogramming in aging, perhaps due to chronic inflammation, and serve to identify potential candidates regulating aging-associated platelet hyperactivity.Conclusions: TNF-associated inflammation of aging is an important factor in the development of platelet hyperreactivity during aging and may contribute to the increased incidence of age-associated thrombosis. Thioredoxin interacting protein regulates age-associated neuroinflammation

Immune system hypersensitivity is believed to contribute to mental frailty in the elderly. Solid evidence indicates NOD-like receptor pyrin domain containing-3 (NLRP3)-inflammasome activation intimately connects aging-associated chronic inflammation (inflammaging) to senile cognitive decline. Thioredoxin interacting protein (TXNIP), an inducible protein involved in oxidative stress, is essential for NLRP3 inflammasome activity. This study aims to find whether TXNIP/NLRP3 inflammasome pathway is involved in senile dementia. According to our studies on sex-matched mice, TXNIP was significantly upregulated in aged animals, paralleled by the NLRP3-inflammasome over-activity leading to enhanced caspase-1 cleavage and IL-1β maturation, in both sexes. This was closely associated with depletion of the anti-aging and cognition enhancing protein klotho, in aged males. Txnip knockout reversed age-related NLRP3-hyperactivity and enhanced thioredoxin (TRX) levels. Further, TXNIP inhibition along with verapamil replicated TXNIP/NLRP3-inflammasome downregulation in aged animals, with FOXO-1 and mTOR upregulation. These alterations concurred with substantial improvements in both cognitive and sensorimotor abilities. Together, these findings substantiate the pivotal role of TXNIP to drive inflammaging in parallel with klotho depletion and functional decline, and delineate thioredoxin system as a potential target to decelerate senile dementia.

Adult mouse kidney stem cell‐derived exosomes attenuate blood pressure and improve kidney functions in aged mice

The physiological function of stem cell-derived extracellular vesicles remains elusive. Most recently, we isolated kidney stem cells (KSC) from adult mouse kidneys and studied the role of KSC-derived exosomes in blood pressure regulation in hypertensive aged mice. We treated aged mice with KSC-exosome (~1.26×1011 ± 1.94×109 particles/mouse/day) or PBS by intraperitoneal injection for 5 days. We found that the blood pressure was reduced (from 150.8±12.2 to 128.6±3 mmHg) at day 6 in KSC-exosome-treated aged mice, while the blood pressure remained unchanged in PBS-treated aged mice. Blood pressure continued to decrease to 116±8.2 mmHg at day 14 after KSC-exosome treatment. Hypertension reemerged gradually and relapsed at day 28 after treatment. To assess the kidney functions, we collected 6-hour urine at day 5 of the treatment. KSC-exosome treatment significantly reduced the increases in urine albumin and creatinine in aged mice, indicating that KSC-exosomes improve aging-associated kidney dysfunction. Notably, the glomerular filtration rate (GFR) was significantly increased in KSC-exosome-treated aged mice, suggesting improved glomerular function. Urinary cytokine levels were increased in aged mice which were largely reduced by KSC-exosomes. KSC-exosomes effectively reduced the kidney injury markers TIM1 and adiponectin and inflammation marker NGAL which were confirmed by ELISA. These data suggest that KSC-exosome treatment attenuates aging-associated renal inflammation and kidney injury. To explore the mechanisms by which KSC-exosome attenuates blood pressure and improves kidney functions in aged mice, we performed proteome analysis of KSC-exosomes by mass spectrometry. We found several pathways that involve in the kidney development were upregulated in KSC-exosome compared to renal tubule cells (RTC) derived-exosomes. Interestingly, KSC-exosome induced cell proliferation and cell migration after injury possibly through activation of AKT signaling demonstrated in vitro. In conclusion, our finding provided the first evidence that the adult mouse kidney stem cell-derived exosomes attenuate aging-associated hypertension and kidney dysfunctions. KSC-exosome treatment may be a novel therapeutic strategy for hypertension-associated kidney injuries in aging.

Inflammaging: Blame the sons. Relationships between the number of sons and the level of inflammatory mediators among post-reproductive women.

Reproduction is costly, but sons and daughters differently influence maternal physiology, also in older age. In particular, having sons may negatively influence maternal health and may be associated with a shorter life span of mothers. Sons may also contribute to increased inflammaging, a chronic sub-clinical systemic inflammatory state characterized by elevated levels of serum inflammatory mediators. The aim of this study was to examine the impact of the total number of children, and the number of daughters and sons separately on concentrations of C-reactive protein (CRP), and proinflammatory cytokines such as interleukin 6 (IL-6) and tumor necrosis factor-α (TNF-α). The participants were 378 women aged 45-92 who had 3.9 (SD 2.12, median = 4, range = 0-13) children, including 2.1 (SD 1.46, median = 2, range = 0-8) sons and 1.8 (SD 1.44, median = 2, range = 0-7) daughters on average. We found a positive relationship between the overall number of children and IL-6 levels. CRP and IL-6 concentrations were positively associated with the number of sons but not with the number of daughters. Each son increased maternal CRP level by 11%, and IL-6 level by 6%. Neither the total number of children nor the number of daughters or sons were related to the TNF-α concentration. Aging-associated inflammation in post-reproductive mothers with a higher number of sons supports the hypothesis of trade-offs between reproduction and health. Furthermore, these results provide new evidence contributing to the idea that having sons may have more detrimental effects on the maternal organism than having daughters.

Aging-associated inflammation and fibrosis in arachnoid membrane

BackgroundThe physiological and pathological significance of the arachnoid membrane (AM) is still unknown. In this study, we investigated various characteristics of the AM, focusing on the influence of inflammation and fibrosis.MethodsSmall pieces of AM sample were obtained during neurosurgical procedures from 74 cases. The clinical and pathological characteristics of the hyperplastic AM group (≥ 50 μm) and the non-hyperplastic AM group (< 50 μm) were compared. Then, potential correlations between AM thickness and clinical characteristics were analyzed. Moreover, VEGFα, TGFβ, and TGFα levels were quantitated by real time PCR. Then, the potential correlations between AM thickness and these inflammatory or anti-inflammatory markers, and the influence of the original disease were calculated.ResultsThe median age of the patients in hyperplastic AM group was significantly older than that of the non-hyperplastic AM group. Moreover, the number of fibroblasts, CD68+ cells, CD86+ cells, and CD206+ cells in the hyperplastic AM group was significantly higher than that in the non-hyperplastic AM group. The AM thickness was significantly correlated to age and number of fibroblasts, CD68+ cells, CD86+ cells, and CD206+ cells. The thickness of the AM was significantly correlated to the messenger RNA expression levels of VEGFα (ρ = 0.337), and the VEGFα expression levels were significantly correlated with TGFβ and TNFα.ConclusionsThe AM hyperplasia was influenced by aging and could be a result of inflammation and fibrosis through cytokine secretion from the inflammatory cells and fibroblasts in the AM.

Signatures of immune dysfunction in HIV and HCV infection share features with chronic inflammation in aging and persist after viral reduction or elimination

Chronic inflammation is thought to be a major cause of morbidity and mortality in aging, but whether similar mechanisms underlie dysfunction in infection-associated chronic inflammation is unclear. Here, we profiled the immune proteome, and cellular composition and signaling states in a cohort of aging individuals versus a set of HIV patients on long-term antiretroviral therapy therapy or hepatitis C virus (HCV) patients before and after sofosbuvir treatment. We found shared alterations in aging-associated and infection-associated chronic inflammation including T cell memory inflation, up-regulation of intracellular signaling pathways of inflammation, and diminished sensitivity to cytokines in lymphocytes and myeloid cells. In the HIV cohort, these dysregulations were evident despite viral suppression for over 10 y. Viral clearance in the HCV cohort partially restored cellular sensitivity to interferon-α, but many immune system alterations persisted for at least 1 y posttreatment. Our findings indicate that in the HIV and HCV cohorts, a broad remodeling and degradation of the immune system can persist for a year or more, even after the removal or drastic reduction of the pathogen load and that this shares some features of chronic inflammation in aging.

Microbiota profiling in aging-associated inflammation and liver degeneration

BackgroundThe number of people above the age of 60 years is raising world-wide being associated with an increase in the prevalence of aging-associated impairments and even diseases. Recent studies suggest that aging is associated with alterations in bacterial endotoxin levels and that these changes may add to low-grade inflammation, the so-called ‘inflammaging’, and aging-associated liver degeneration. However, mechanisms involved, and especially, the interaction of intestinal microbiota and barrier in the development of aging-associated inflammation and liver degeneration have not been fully understood. ObjectiveThe aim of the present study was to determine if intestinal microbiota composition changes with age and if these alterations are associated with changes of markers of intestinal barrier function and the development of inflammation and liver degeneration. MethodsBlood, liver, small and large intestinal tissue of male 2-, 15-, 24- and 30-months old C57BL/6 mice fed standard chow were obtained. Intestinal microbiota composition, expression levels of antimicrobial peptides in small intestine and markers of intestinal barrier function were measured. Furthermore, indices of liver damage, inflammation and expression levels of lipopolysaccharide binding protein (Lbp) as well as of toll-like receptors (Tlr) 1-9 in liver tissue were assessed. ResultsPairwise comparisons of the microbial community in the small intestine showed differences between 2- and 24-, 15- and 24-, as well as 15- and 30-months old animals while Shannon’s diversity, species richness and evenness indexes did not differ in both small and large intestine, respectively, between age groups. Concentrations of nitric oxide were significantly lower in small intestine of 15-, 24- and 30-months old mice compared to 2-months old mice while mRNA expression of the antimicrobial peptides defensin alpha 1 and lysozyme 1 was unchanged. In contrast, in liver tissue, older age of animals was associated with increasing inflammation and the development of fibrosis in 24- and 30-months old mice. Numbers of inflammatory foci and neutrophils in livers of 24- and 30-months old mice were significantly higher compared to 2-months old mice. These alterations were also associated with higher endotoxin levels in plasma as well as an increased mRNA expression of Lbp and Tlr1, Tlr2, Tlr4, Tlr6 and Tlr9 in livers in older mice. ConclusionDespite no consistent and robust changes of microbiota composition in small and/or large intestine of mice of different age were observed, our data suggest that alterations of markers of intestinal barrier function in small intestine are associated with an induction of several Tlrs and beginning hepatic inflammation in older mice and increase with age.

Senescent T Cell Induces Brown Adipose Tissue "Whitening" Via Secreting IFN-γ.

Aging-associated chronic inflammation is a key contributing factor to a cluster of chronic metabolic disorders, such as cardiovascular disease, obesity, and type 2 diabetes. Immune cells particularly T cells accumulate in adipose tissue with advancing age, and there exists a cross talk between T cell and preadipocyte, contributing to age-related adipose tissue remodeling. Here, we compared the difference in morphology and function of adipose tissue between young (3-month-old) and old (18-month-old) mice and showed the phenomenon of brown adipose tissue (BAT) “whitening” in old mice. Flow cytometry analysis suggested an increased proportion of T cells in BAT of old mice comparing with the young and exhibited senescent characteristics. We take advantage of coculture system to demonstrate directly that senescent T cells inhibited brown adipocyte differentiation of preadipocytes in adipose tissue. Mechanistically, both in vitro and in vivo studies suggested that senescent T cells produced and released a higher level of IFN-γ, which plays a critical role in inhibition of preadipocyte-to-brown adipocyte differentiation. Taken together, the data indicate that senescent T cell-derived IFN-γ is a key regulator in brown adipocyte differentiation.
