Aggregation-induced emission nanoparticles facilitating multicolor lateral flow immunoassay for rapid and simultaneous detection of aflatoxin B1 and zearalenone

Herein we developed a multicolor lateral flow immunoassay (LFIA) test strip for rapid and simultaneous quantitative detection of aflatoxin B1 (AFB1) and zearalenone (ZEN). Three differently colored aggregation-induced emission nanoparticles (AIENPs) were designed as LFIA signal tags, with red and green AIENPs for targeting AFB1 and ZEN at the test line, and yellow AIENPs for indicating the validity of the test strip at the control (C) line. After surface functionalization with antibodies, the developed AIENP-based multicolor LFIA allows simultaneous and accurate quantification of AFB1 and ZEN using an independent C-line assisted ratiometric signal output strategy. The detection limits of AFB1 and ZEN were 6.12 and 26 pg/mL, respectively. The potential of this method for real-world applications was well demonstrated in corn and wheat. Overall, this multicolor LFIA shows great potential for field screening of multiple mycotoxins and can be extended to rapid and simultaneous monitoring of other small molecule targets.

Ultra-sensitive fluorescent immunoassay based on bright AIE nanoparticle for biomarkers detection and quantification

Fluorescence immunoassay (FIA) is one of the most immunoassay diagnosis methods in clinic for biomarker quantification in recent years. The development of reliable FIA strategy with high-sensitivity, large-rang and outstanding efficiency for biomarkers rapid diagnosis, especially in complicated samples, is still a great challenge. Herein, an ultra-sensitive FIA combined with magnetic separation was established to achieve efficient point-of-care diagnosis in quantitative behavior based on bright aggregation-induced emission nanoparticles (AIE NPs). By utilizing swelling method, AIE NPs with controllable size, relatively high luminescence efficiency and large Stokes shift were prepared, moreover, the obtained NPs also exhibit narrow size distribution and well-defined spherical morphology. Stability studies prove that AIE NPs not only possess quite high photostability, but also present good resistance towards harsh acid-alkaline steadily. By using AIE NPs as the reporter, a reliable and efficient FIA sensor was successfully constructed. The precise detection of C-reaction protein (CRP) antigen was acquired with the linear range of 0.5 ng·mL−1 ∼ 1000 ng·mL−1 and the limit of detection (LOD) of 0.35 ng·mL−1, which is comparative with the conventional chemiluminescence immunoassays. Moreover, the whole detection operation could be accomplished within 10 min, which is relatively rapid and desirable for point-of-care diagnosis. In addition, this strategy also presented excellent specificity towards CRP antigens among various inflammatory antigens. These results confirm that AIE NPs-based FIA strategy provides an ideal and reliable approach for precise and quantitive biosensing of in vitro diagnostics.

A New Theranostic Platform Against Gram-Positive Bacteria Based on Near-Infrared-Emissive Aggregation-Induced Emission Nanoparticles.

Infections induced by Gram-positive bacteria pose a great threat to public health. Antibiotic therapy, as the first chosen strategy against Gram-positive bacteria, is inevitably associated with antibiotic resistance selection. Novel therapeutic strategies for the discrimination and inactivation of Gram-positive bacteria are thus needed. Here, a specific type of aggregation-induced emission luminogen (AIEgen) with near-infrared fluorescence emission as a novel antibiotic-free therapeutic strategy against Gram-positive bacteria is proposed. With the combination of a positively charged group into a highly twisted architecture, self-assembled AIEgens (AIE nanoparticles (NPs)) at a relatively low concentration (5µm) exhibited specific binding and photothermal effect against living Gram-positive bacteria both in vitro and in vivo. Moreover, toxicity assays demonstrated excellent biocompatibility of AIE NPs at this concentration. All these properties make the AIE NPs as a novel generation of theranostic platform for combating Gram-positive bacteria and highlight their promising potential for in vivo tracing of such bacteria.

Dual-color nanospheres based on aggregation-induced emission and catalytic hairpin assembly for simultaneous imaging of acrylamide and miR-21 in living cells

Acrylamide (AA) is a heat-processed potent food carcinogen that is widely used in industry, posing a significant risk to human health. Therefore, it is necessary to investigate the toxic effects and mechanism of AA. miR-21 is a representative biomarker during AA-induced carcinogenesis. Here, dual-color aggregation-induced emission nanoparticles (AIENPs) were developed for the detection and simultaneous imaging of AA and miR-21. AIENPs were synthesized by combining aggregation-induced emission (AIE) dyes and a poly (styrene-co-maleic anhydride) (PSMA) amphiphilic polymer modified with hairpin DNA. Upon AA intervention and aptamer recognition, cDNA was dissociated, leading to miR-21 overexpression and initiating the catalytic hairpin assembly cycle. Consequently, fluorescence quenching was observed due to FRET between AIENPs and labeled quenchers. The relative fluorescence intensities of dual-color AIENPs displayed good linear relationships with logarithmic AA and miR-21 concentrations. Moreover, there was a gradual decrease in dual-color AIENP fluorescence as the HepG2 cell concentration of AA (0–500 μM) and stimulation time (0–12 h) increased, making it possible to simultaneously image AA and AA-induced miR-21. The findings of this work are valuable for revealing the cytotoxic mechanism of AA.

Near‐infrared laser‐activated aggregation‐induced emission nanoparticles boost tumor carbonyl stress and immunotherapy of breast cancer

AbstractThe induction of tumor carbonyl stress is reported to efficiently revert immune suppression in the tumor microenvironment and enhance cancer immunotherapy. However, low oxygen concentration due to inherent tumor hypoxia limits its catalytic effect. Herein, an injectable thermosensitive hydrogel system (named APH) is developed for co‐loading of near‐infrared (NIR) aggregation‐induced emission (AIE) nanoparticles and plasma amine oxidase (PAO) for boosting carbonyl stress and enhancing antitumor immunity. Upon 808 nm NIR laser irradiation, the AIE nanoparticles trigger a mild‐temperature (around 45°C) photothermal effect in the tumor site, which significantly relieves tumor hypoxia and promotes the catalytic effect of released PAO to inhibit the growth of Myeloid‐derived suppressor cells. Remarkably, the synergistic therapeutic effect of APH is verified through a significant inhibitory effect on the distant tumor, enhanced immune memory, and effective suppression of postoperative recurrence, rechallenge, and metastasis. Overall, the combined effect of AIE‐mediated photothermal therapy and carbonyl stress by APH upon NIR irradiation therapy can significantly activate cancer immunotherapy, making it a promising treatment approach for cancer treatment.

An aggregation-induced emission sensor combined with UHPLC-Q-TOF/MS for fast identification of anticoagulant active ingredients from traditional Chinese medicine

Xuebijing injection (XBJ) has a good therapeutic effect on the patients with severe coronavirus disease, but the material basis of XBJ with the anticoagulant effect to improve the coagulopathy and thromboembolism is still unclear. Herein, we developed a new strategy based on aggregation-induced emission (AIE) for monitoring thrombin activity and screening thrombin inhibitors from XBJ. The molecule AIE603 and the thrombin substrate peptide S-2238 were formed into AIE nanoparticle (AIENP) which emitted notable fluorescence due to the restriction of intramolecular motions. In the presence of thrombin, AIENP was specifically hydrolyzed and AIE603 was released from AIENP, leading to the decrease of fluorescence intensity. Furthermore, AIENP was combined with ultra-high performance liquid chromatography-fraction collector (UHPLC-FC) and ultra-high performance liquid chromatography quadrupole-time-of-flight mass spectrometry (UHPLC-Q-TOF/MS) for separation, preparation, screening and identification of the thrombin inhibitors from XBJ, a total of 58 chemical constituents were identified, among which 6 compounds possessed higher anticoagulant activity. Notably, the overall inhibition rate of the 6 mixed standards was equivalent to about 60% of the inhibition rate of XBJ. Therefore, this work provides a novel, cheap and simple method for monitoring thrombin activity and is promising to screen active substances from traditional Chinese medicines.

AIE Bioconjugates for Accurate Identification and In Vivo Targeted Treatment of Bacterial Infection Based on Bioorthogonal Reaction.

Targeted killing multidrug-resistant bacteria with high efficiency is urgently needed for the treatment of infection with minimal collateral damage. Herein, a new near-infrared (NIR) fluorescence nanoprobe is designed and synthesized with aggregation-induced emission (AIE) features, which also is excellent reactive oxygen species (ROS) generator. The as-prepared AIE nanoparticles (NPs) present outstanding sterilizing rate on methicillin-resistant Staphylococcus aureus (MRSA) and kanamycin-resistant Escherichia coli (KREC). Meanwhile, considering the differences in the surface structure of animal cells and bacteria, a non-invasive image-guided strategy for precise treatment of bacterial infection has been successfully implemented based on bioorthogonal reaction which can perform and control unnatural chemical reactions inside living organisms. The AIE NPs are thus specifically trapped on the bacterial surface while not on the normal cells, realizing real-time tracking of the infected site distribution in vivo and guiding photodynamic therapy (PDT) for eliminating bacteria in inflammation region. That significantly improves the accuracy and sterilization rate of bacterial-infected wounds with negligible side effects. The investigation developed a potential antibacterial agent and also provides an instructive way for targeting treatment based on bioorthogonal reaction.

Organic Nanoparticles with Aggregation-Induced Emission and Two-Photon Excitation for Fluorescence Imaging of Living Cells/Tissues.

Aggregation-induced emission (AIE) nanoparticles (NPs) have been applied in bioimaging for cancer diagnosis due to high fluorescence efficiency. However, the poor cell permeability as well as autofluorescence of biological cells/tissues caused by ultraviolet (UV) irradiation is still the key problem of AIE luminophores for biological imaging. Here, we report green-emitting organic AIE luminophores for fluorescence imaging of living cells/tissues, which possess high fluorescence quantum yields and strong AIE under two-photon excitation with near-infrared light beyond 800 nm. These AIE luminophores can bind with bovine serum albumin (BSA) to form biocompatible BSA/AIE-NPs due to their terminal aldehyde groups providing specific anchor sites with the receptor groups in BSA. Furthermore, one/two-photon fluorescence bioimaging for Hela cancer cells has been successfully carried out with BSA/AIE-NPs as the fluorescent probe, and BSA/AIE-NPs show excellent stain properties with a fast permeability of only 5 min, high cellular uptakes, and strong fluorescence. The results demonstrate the great advantages of BSA/AIE-NPs in fast fluorescence biological imaging as well as further cancer diagnosis and therapy.

Lysosome-Targeting Aggregation-Induced Emission Nanoparticle Enables Adoptive Macrophage Transfer-Based Precise Therapy of Bacterial Infections.

Traditional antibacterial procedures are getting inefficient due to the emergence of antimicrobial resistance, which makes alternative treatments in urgent demand. However, the selectivity toward infectious bacteria is still challenging. Herein, by taking advantage of the self-directed capture of infectious bacteria by macrophages, we developed a strategy to realize precise in vivo antibacterial photodynamic therapy (APDT) through adoptive photosensitizer-loaded macrophage transfer. TTD with strong reactive oxygen species (ROS) production and bright fluorescence was first synthesized and was subsequently formulated into TTD nanoparticles for lysosome targeting. TTD-loaded macrophages (TLMs) were constructed by direct incubation of TTD nanoparticles with macrophages, in which the TTD was localized in the lysosomes to meet the captured bacteria in the phagolysosomes. The TLMs could precisely capture and eradicate bacteria while being activated toward the proinflammatory and antibacterial M1 phenotype upon light illumination. More importantly, after subcutaneous injection, TLMs could effectively inhibit bacteria in the infected tissue through APDT, leading to good tissue recovery from severe bacterial infection. Overall, the engineered cell-based therapeutic approach shows great potential in the treatment of severe bacterial infectious diseases.

Neutrophil-like Biomimic AIE Nanoparticles with High-Efficiency Inflammatory Cytokine Targeting Enable Precise Photothermal Therapy and Alleviation of Inflammation.

Although photothermal therapy (PTT) has thrived as a promising treatment for drug-resistant bacterial infections by avoiding the abuse of antibiotics, the remaining challenges that limit the treatment efficiency are the poor targeting properties of infected lesions and low penetration to the cell membrane of Gram-negative bacteria. Herein, we developed a biomimetic neutrophil-like aggregation-induced emission (AIE) nanorobot (CM@AIE NPs) for precise inflammatory site homing and efficient PTT effects. Due to their surface-loaded neutrophil membranes, CM@AIE NPs can mimic the source cell and thus interact with immunomodulatory molecules that would otherwise target endogenous neutrophils. Coupled with the secondary near-infrared region absorption and excellent photothermal properties of AIE luminogens (AIEgens), precise localization, and treatment in inflammatory sites can be achieved, thereby minimizing damage to surrounding normal tissues. Moreover, CM@AIE NP-mediated PTT was stimulated in vivo by a 980 nm laser irradiation, which contributed to the extent of the therapeutic depth and limited the damage to skin tissues. The good biocompatibility and excellent in vitro and in vivo antibacterial effects prove that CM@AIE NPs can provide a strategy for broad-spectrum antibacterial applications.

Revealing the In Situ Behavior of Aggregation-Induced Emission Nanoparticles and Their Biometabolic Effects via Mass Spectrometry Imaging.

Simultaneous imaging of exogenous nanomaterials and endogenous metabolites in situ remains challenging and is beneficial for a systemic understanding of the biological behavior of nanomaterials at the molecular level. Here, combined with label-free mass spectrometry imaging, visualization and quantification of the aggregation-induced emission nanoparticles (NPs) in tissue were realized as well as related endogenous spatial metabolic changes simultaneously. Our approach enables us to identify the heterogeneous deposition and clearance behavior of nanoparticles in organs. The accumulation of nanoparticles in normal tissues results in distinct endogenous metabolic changes such as oxidative stress as indicated by glutathione depletion. The low passive delivery efficiency of nanoparticles to tumor foci suggested that the enrichment of NPs in tumors did not benefit from the abundant tumor vessels. Moreover, spatial-selective metabolic changes upon NPs mediated photodynamic therapy was identified, which enables understanding of the NPs induced apoptosis in the process of cancer therapy. This strategy allows us to simultaneously detect exogenous nanomaterials and endogenous metabolites in situ, hence to decipher spatial selective metabolic changes in drug delivery and cancer therapy processes.

Aggregation-Induced Red Emission Nanoparticle-Based Lateral Flow Immunoassay for Highly Sensitive Detection of Staphylococcal Enterotoxin A.

Staphylococcal enterotoxin A (SEA) has presented enormous difficulties in dairy food safety and the sensitive detection of SEA provides opportunities for effective food safety controls and staphylococcal food poisoning tracebacks. Herein, a novel aggregation-induced emission (AIE)-based sandwich lateral flow immunoassay (LFIA) was introduced to detect SEA by using red-emissive AIE nanoparticles (AIENPs) as the fluorescent nanoprobe. The nanoprobe was constructed by directly immobilising antibodies on boronate-tagged AIENPs (PBA-AIENPs) via a boronate affinity reaction, which exhibited a high SEA-specific affinity and remarkable fluorescent performance. Under optimal conditions, the ultrasensitive detection of SEA in pasteurised milk was achieved within 20 min with a limit of detection of 0.04 ng mL-1. The average recoveries of the PBA-AIENP-LFIA ranged from 91.3% to 117.6% and the coefficient of variation was below 15%. It was also demonstrated that the PBA-AIENP-LFIA had an excellent selectivity against other SE serotypes. Taking advantage of the excellent sensitivity of this approach, real chicken and salad samples were further analysed, with a high versatility and accuracy. The proposed PBA-AIENP-LFIA platform shows promise as a potent tool for the identification of additional compounds in food samples as well as an ideal test method for on-site detections.

Ultrabright orange-yellow aggregation-induced emission nanoparticles for highly sensitive immunochromatographic quantification of ochratoxin A in corn

Herein, we report a novel aggregation-induced emission nanoparticles (AIENPs)-based immunochromatography assay (ICA) platform to detect ochratoxin A (OTA) using orange-yellow-emitting AIENPs as fluorescent nanoprobes. Immunochromatographic strip is used for the quantitative detection of OTA in crop matrix using AIENPs coupled with anti-OTA ascites. Under optimal conditions, AIENPs-ICA exhibits stronger signal output capacity and higher sensitivity than traditional gold nanoparticles-based ICA. The half-maximal inhibitory concentration is as low as 0.149 ng mL−1, and the limit detection is 0.042 ng mL−1 at 10 % competitive inhibition concentration. The average recovery of AIENPs-ICA ranges from 82.60 % to 113.14 % with the coefficient of variation ranging from 1.26 % to 11.57 %, proving the proposed method possesses good reliability and reproducibility. Moreover, the developed AIENPs-ICA exhibits negligible cross-reactions with other mycotoxins. We believe the presented AIENPs-ICA platform holds promising potential as a powerful tool for on-site detection of OTA and other molecules detection in food samples.

Enrichment of Aggregation-Induced Emission Aggregates Using Acoustic Streaming Tweezers in Microfluidics for Trace Human Serum Albumin Detection.

Aggregation-dependent brightness (ADB) indirectly limits the in vitro performance of a pure aggregation-induced emission (AIE) probe in many ways; thus, controlling the aggregation state of the AIE probe is helpful for detecting an object of interest. Many studies are focused on the molecule design of the AIE probes, while less efforts have been made for the control of the aggregation of the AIEs. Here, an acoustic streaming tweezer (AST) generated using a gigahertz bulk acoustic wave resonator was applied to manipulate the aggregation status of the AIE probe and further enhance their performance for human serum albumin (HSA) detection. As the trapping size of the AST matches the working size of the AIE probe, the streaming can enrich and accumulate AIE nanoparticles, which then further trigger larger aggregates. Due to the ADB effect, the fluorescence intensity strongly increased, and thus, the detection limit of HSA was reduced to 0.5 μg/mL, which is low enough for kidney disease detection. Such an AST-assisted ADB strategy is potentially applicable to other AIE probes and can work as a portable choice for the biomedical detection.

Brightness of fluorescent organic nanomaterials.

Brightness is a fundamental property of fluorescent nanomaterials reflecting their capacity to absorb and emit light. In sensing materials, brightness is crucial for high-sensitivity (bio)molecular detection, while in optical bioimaging it ensures high spatial and temporal resolution. Fluorescent organic nanoparticles (NPs) are particularly attractive because of their superior brightness compared to organic dyes. With the ever-growing diversity of organic nanomaterials, it is important to establish universal principles for measuring and estimating their brightness. This tutorial review provides definitions of brightness and describes the major approaches to its analysis based on ensemble and single-particle techniques. We present the current chemical approaches to fight Aggregation-Caused Quenching (ACQ) of fluorophores, which is a major challenge in the design of bright organic nanomaterials. The main classes of fluorescent organic NPs are described, including conjugated polymer NPs, aggregation-induced emission NPs, and NPs based on neutral and ionic dyes. Their brightness and other properties are systematically compared. Some brightest examples of bulk solid-state emissive organic materials are also mentioned. Finally, we analyse the importance of brightness and other particle properties in biological applications, such as bioimaging and biosensing. This tutorial will provide guidelines for chemists on the design of fluorescent organic NPs with improved performance and help them to estimate and compare the brightness of new nanomaterials with literature reports. Moreover, it will help biologists to select appropriate materials for sensing and imaging applications.

Genetically edited T-cell membrane coated AIEgen nanoparticles effectively prevents glioblastoma recurrence

Glioblastoma stem cells (GSCs) are subpopulations of tumor-initiating cells responsible for glioblastoma (GBM) tumorigenesis and recurrence. Dual inhibition of vascular endothelium and GSCs is still a challenge due to their different pathological features. Here we present a combined all-in-control strategy to realize a local photothermal therapy (PTT). We designed T-cell-mimic nanoparticles with aggregation-induced emission (AIE) characteristics by coating the genetically engineered T cell membrane (CM) onto AIE nanoparticles (CM@AIE NPs). The CM shell was designed against CD133 and epidermal growth factor receptor (EGFR) which provides the possibility to target both GBM cells and GSCs for cancer therapy. CM@AIE NPs can serve as the tight junction (TJ) modulators to trigger an intracellular signaling cascade, causing TJ disruption and actin cytoskeleton reorganization to allow CM@AIE NPs to cross the blood-brain barrier (BBB) silently. The 980 nm excitation-triggered PTT can completely inhibit tumorigenesis and recurrence. The combination of CM-coating nanotechnology and genetic editing technique can inspire further development of synergetic techniques for preventing GBM recurrence.

Erythrocyte Membrane-Camouflaged Aggregation-Induced Emission Nanoparticles for Fetal Intestinal Maturation Assessment.

Assessment of fetal maturity is essential for timely termination of pregnancy, especially in pregnant women with pregnancy complications. However, there is a lack of methods to assess the maturity of fetal intestinal function. Here, we constructed erythrocyte membrane-camouflaged aggregation-induced emission (AIE) nanoparticles. Nanocore is formed using a hollow mesoporous silicon nanobox (HMSN) of different particle sizes loaded with AIE luminogens -PyTPA (P), which are then co-extruded with erythrocyte membranes (M) to construct M@HMSN@P. The 100 nm M@HMSN@P has a more effective cellular uptake efficiency in vitro and in vivo. Swallowing and intestinal function in fetal mice mature with the increase in gestational age. After intrauterine injection of M@HMSN@P, they were swallowed and absorbed by fetal mice, and their swallowed and absorbed amount was positively correlated with the gestational age with a correlation coefficient of 0.9625. Using the M@HMSN@P (fluorescence intensity) in fetal mice, the gestational age can be imputed, and the difference between this imputed gestational age and the actual gestational age is less than 1 day. Importantly, M@HMSN@P has no side effect on the health status of pregnant and fetal mice, showing good biocompatibility. In conclusion, we constructed M@HMSN@P nanoparticles with different particle sizes and confirmed that the smaller size M@HMSN@P has more efficient absorption efficiency and it can assess fetal intestinal maturity by the intensity of the fluorescence signal.

Short-Wavelength Aggregation-Induced Emission Photosensitizers for Solid Tumor Therapy: Enhanced with White-Light Fiber Optic.

White-light photodynamic therapy (wPDT) has been used in the treatment of cancer due to its convenience, effectiveness and less painful. However, the limited penetration of white-light into the tissues leads to a reduced effectiveness of solid tumor treatment. Two short-wavelength aggregation-induced emission (AIE) nanoparticles were prepared, PyTPA@PEG and TB@PEG, which have excitation wavelengths of 440 nm and 524 nm, respectively. They were characterized by UV, fluorescence, particle size and TEM. The ability of nanoparticles to produce reactive oxygen species (ROS) and kill cancer cells under different conditions was investigated in vitro, including white-light, after white-light penetrating the skin, laser. A white-light fiber for intra-tumor irradiation was customized. Finally, induced tumor elimination with fiber-mediated wPDT was confirmed in vivo. In vitro, both PyTPA@PEG and TB@PEG are more efficient in the production ROS when exposed to white-light compared to laser. However, wPDT also has a fatal flaw in that its level of ROS production after penetrating the skin is reduced to 20-40% of the original level. To this end, we have customized a white-light fiber for intra-tumor irradiation. In vivo, the fiber-mediated wPDT significantly induces tumor elimination with maximized therapeutic outcomes by irradiating the interior of the tumor. In addition, wPDT also has the advantage that its light source can be adapted to a wide range of photosensitizers (wavelength range 400-700 nm), whereas a laser of single wavelength can only target a specific photosensitizer. This method of using optical fiber to increase the tissue penetration of white light can greatly improve the therapeutic effect of AIE photosensitizers, which is needed for the treatment of large/deep tumors and holds great promise in cancer treatment.

All-in-One Theranostic Platforms: Deep-Red AIE Nanocrystals to Target Dual-Organelles for Efficient Photodynamic Therapy.

Aggregation-induced emission (AIE) nanoparticles have been widely applied in photodynamic therapy (PDT) over the past few years. However, amorphous nanoaggregates usually occur in their preparation, resulting in loose packing with disordered molecular structures. This still allows free intramolecular motions, thus leading to limited brightness and PDT efficiency. Herein, we report deep-red AIE nanocrystals (NCs) of DTPA-BS-F by following the facile method of nanoprecipitation. It is observed that DTPA-BS-F NCs possess not only a high photoluminescence quantum yield value of 8% in the deep-red region (600-850 nm) but also an impressive reactive oxygen species (ROS) generation efficiency of up to 69%. Moreover, DTPA-BS-F NCs targeting dual-organelles of lysosomes and nucleus to generate ROS are also achieved, thus boosting the PDT effect in cancer therapy both in vitro and in vivo. This work provides high-performance AIE NCs to simultaneously target two organelles for efficient photodynamic therapy, indicating their promising application in all-in-one theranostic platforms.
