The Current Status of Viral Nervous Necrosis Disease in Türkiye

The agent of Viral Nervous Necrosis (VNN) disease is betanodavirus and is a viral fish disease and VNN disease has been reported in many fish species in Türkiye and around the world. It is known to cause high mortality rates in aquatic animals living in both marine and fresh water. It has been reported that the RGNNV and RGNNV/SJNNV genotypes of the virus, which has four genotypes, were detected in Turkey by the end of 2023. RGNNV genotype was detected in sea bass (Dicentrarchus labrax) for the first time in our country in 2011 and after that has since been found in other fish species such as sea bream (Sparus aurata), red mullet (Mullus barbatus), and garfish (Belone belone) in the Mediterranean region. RGNNV genotypes have also been reported in sea bass (D. labrax) in the Black Sea, and in sea bass (D. labrax) and RGNNV/SJNNV genotypes have been reported sea bream (S. aurata) in the Aegean Sea. In this study, studies on VNN in Turkey were reviewed and it was aimed to discuss the current status of the disease as a whole.

Genetic and morphological characterization of a new genotype of nervous necrosis virus circulating among Nile tilapia in the south of Egypt.

Nervous necrosis virus (NNV) is the causative agent of viral nervous necrosis in freshwater and marine fishes. In this study, NNV circulating among wild and farmed Nile tilapia (Oreochromis niloticus) was genetically and morphologically characterized using reverse transcription polymerase chain reaction (RT-PCR), sequencing analysis, and transmission electron microscopy (TEM). Brain, eye, and other organ (spleen, kidney, heart, and liver) specimens were collected from 87 wild (66) and farmed (21) Nile tilapia fish during their adult or juvenile stage at different localities in Qena and Sohag governorates in southern Egypt. Among them, 57/87 fish showed suspected NNV clinical signs, and 30/87 were healthy. The results revealed that NNV was detected in 66 out of 87 fish (58.62% in the wild and 17.24% in farmed Nile tilapia by RT-PCR), and the prevalence was higher among diseased (55.17%) than in healthy (20.69%) fish. NNV was detected in the brain, eye, and other organs. Using TEM, virion size variations based on the infected organs were observed. Nucleotide sequence similarity indicated that NNVs had a divergence of 75% from other fish nodaviruses sequenced in Egypt and worldwide. Phylogenetic analysis distinguished them from other NNV genotypes, revealing the emergence of a new NNV genotype in southern Egypt. In conclusion, NNV is circulating among diseased and healthy Nile tilapia, and a new NNV genotype has emerged in southern Egypt.

Isolation and identification of a new strain of nervous necrosis virus from the big-belly seahorse Hippocampus abdominalis

BackgroundBetanodaviruses, members of the Nodaviridae family, are the causative agents of viral nervous necrosis in fish, resulting in great economic losses worldwide.MethodsIn this study, we isolated a virus strain named seahorse nervous necrosis virus (SHNNV) from cultured big-belly seahorses Hippocampus abdominalis in Xiamen city, Fujian Province, China. Virus isolation, PCR detection, phylogenetic analysis, qRT-PCR, fluorescence in situ hybridization and histology were used for virus identification and analysis of virus histopathology. Furthermore, an artificial infection experiment was conducted for virulence testing.ResultsBrain and eye tissue homogenates of diseased big-belly seahorses were inoculated onto a grouper spleen (GS) cell monolayer at 28 °C. Tissue homogenates induced obvious cytopathic effects in GS cells. PCR and sequencing analyses revealed that the virus belonged to Betanodavirus and shared high sequence identity with red-spotted grouper nervous necrosis virus isolates. qRT-PCR and fluorescence in situ hybridization revealed that SHNNV mainly attacked the brain and eye. Histopathological examination revealed that the virus led to cytoplasmic vacuolation in the brain and retinal tissues. Infection experiments confirmed that SHNNV was highly infectious, causing massive death in big-belly seahorses.ConclusionA novel seahorse betanodavirus from the big-belly seahorse cultured in China was discovered. This finding will contribute to the development of efficient strategies for disease management in aquaculture.

Multiple isoforms of HSP70 and HSP90 required for betanodavirus multiplication in medaka cells.

Heat shock proteins (HSPs) are molecular chaperones that have recently been shown to function as host factors (HFs) for virus multiplication in fish as well as in mammals, plants, and insects. HSPs are classified into families, and each family has multiple isoforms. However, no comprehensive studies have been performed to clarify the biological importance of these multiple isoforms for fish virus multiplication. Betanodaviruses are the causative agents of viral nervous necrosis in cultured marine fish and cause very high mortality. Although the viral genome and encoded proteins have been characterized extensively, information on HFs for these viruses is limited. In this study, therefore, we focused on the HSP70 and HSP90 families to examine the importance of their isoforms for betanodavirus multiplication. We found that HSP inhibitors (17-AAG, radicicol, and quercetin) suppressed viral RNA replication and production of progeny virus in infected medaka (Oryzias latipes) cells. Thermal stress or virus infection resulted in increased expression of some isoform genes and facilitated virus multiplication. Furthermore, overexpression and knockdown of some isoform genes revealed that the isoforms HSP70-1, HSP70-2, HSP70-5, HSP90-α1, HSP90-α2, and HSP90-β play positive roles in virus multiplication in medaka. Collectively, these results suggest that multiple isoforms of fish HPSs serve as HFs for betanodavirus multiplication.

Mass Production of Virus-Like Particles Using Chloroplast Genetic Engineering for Highly Immunogenic Oral Vaccine Against Fish Disease.

Nervous necrosis virus (NNV) is the causative agent of viral nervous necrosis (VNN), which is one of the most serious fish diseases leading to mass mortality in a wide range of fish species worldwide. Although a few injectable inactivated vaccines are commercially available, there is a need for more labor-saving, cost-effective, and fish-friendly immunization methods. The use of transgenic plants expressing pathogen-derived recombinant antigens as edible vaccines is an ideal way to meet these requirements. In this study, chloroplast genetic engineering was successfully utilized to overexpress the red-spotted grouper NNV capsid protein (RGNNV-CP). The RGNNV-CP accumulated at high levels in all young, mature, and old senescent leaves of transplastomic tobacco plants (averaging approximately 3 mg/g leaf fresh weight). The RGNNV-CP efficiently self-assembled into virus-like particles (RGNNV-VLPs) in the chloroplast stroma of the transgenic lines, which could be readily observed by in situ transmission electron microscopy. Furthermore, intraperitoneal injection and oral administration of the crudely purified protein extract containing chloroplast-derived RGNNV-VLPs provided the sevenband grouper fish with sufficient protection against RGNNV challenge, and its immunogenicity was comparable to that of a commercial injectable vaccine. These findings indicate that chloroplast-derived VLP vaccines may play a promising role in the prevention of various diseases, not only in fish but also in other animals, including humans.

Autophagy Participates in Lysosomal Vacuolation-Mediated Cell Death in RGNNV-Infected Cells.

Nervous necrosis virus (NNV) is the etiological agent of viral nervous necrosis (VNN), also known as viral encephalopathy and retinopathy (VER), which results in heavy economic losses to the aquaculture industry worldwide. Dramatic cytoplasmic vacuoles were observed during NNV infection both in vitro and in vivo; however, the origin and mechanism of cytoplasmic vacuolization remains unknown. In this report, we found that the cytoplasmic vacuole morphology became fused and enlarged during infection with red spotted grouper nervous necrosis virus (RGNNV), which was accompanied by increased cell death. Notably, Lyso-Tracker, but not Mito-Tracker or ER-Tracker, was accumulated in the vacuoles, and abnormal lysosome swelling was observed in RGNNV-infected cells, suggesting that the cytoplasmic vacuoles originated from lysosomal organelles. Cytoplasmic vacuolization and cell death in RGNNV-infected cells was completely blocked by the vacuolar H+-ATPase inhibitor (bafilomycin A1), and was significantly weakened by chloroquine (CQ), a lysosomotropic agent that induces the acidification of the lysosomes. This suggests that lysosome acidification was essential for vacuole formation. Significant inhibitory effects on vacuolization and cell death were also observed in the RGNNV-infected cells following treatment with nigericin and monensin (ionophores that uncouple the proton gradient present in lysosomes). This indicated that lysosome function was tightly associated with RGNNV infection-induced cell death. In addition, vacuoles were found to be partially co-localized with GFP-LC3II punctate dots during RGNNV infection. Moreover, the severity of vacuolization and cell death were both significantly decreased after treatment with the autophagy inhibitor, 3-MA, suggesting that autophagy was involved in lysosomal vacuolization and cell death evoked by RGNNV infection. Thus, our results demonstrate that autophagy participates in lysosomal vacuolation-mediated cell death during RGNNV infection, and provides new insight into our understanding of the potential mechanisms underlying nodavirus pathogenesis in vitro.

Viral nervous necrosis (VNN) an emerging disease caused by Nodaviridae in aquatic hosts: Diagnosis, control and prevention: A review

Betanodavirus is one of the two genera making up the family Nodaviridae and is the etiological agent of viral nervous necrosis (VNN, also known as viral encephalopathy and retinopathy or VER). The virus infects a large range of host species in more than 50 species of marine and freshwater fish worldwide from different geographical areas and the known host range continues to expand as new species of fish are used for aquaculture. The disease is characterized by vacuolating necrosis of neural cells of the brain, retina and spinal cord and causes up to 100% mortality in larval and juvenile fish, and can cause significant losses in older fish. The lack of knowledge about control and prevention of the disease makes the problem serious and impedes development of management approaches. Therefore this review focuses on current knowledge and future perspectives of viral nervous necrosis in the aquaculture industry with special focus on the type of diagnosis, control and prevention of the disease.

Cell Surface Display of Red-Grouper Nervous Necrosis Virus Capsid Protein on <i>Pichia pastoris</i>

Nervous necrosis virus (NNV), the etiological agent of viral nervous necrosis, has a high mortality rate of 100% in hatchery-reared larvae and juveniles. At present, there are still no effective vaccines available for NNV. Pichia pastoris surface display of viral capsid proteins was generated in hopes of developing an oral vaccine against red-grouper-nervous-necrosis virus (RGNNV) in fish. Fingerlings or juveniles that showed clinical signs of NNV infection were proved by RT-PCR for the appearance of expected length of 198 bpcDNA and further analysis by DNA sequencing. The DNA fragment containing AGα1 linked to RG-NNVRNA2, 2100 bp in length, was inserted into pPIC9K vector. Linearlized plasmids were electroporated into P. pastoris GS115 (mut+His−) and yeast isolates that had Muts−His+ and resistance phenotype at 4 mg/mL geniticin were selected to determine the integration of the target gene by PCR reaction. The extracted cell walls from the yeasts cultured in buffered-methanol-complex medium (BMMY) through an induction of 0.5% methanol for 6 days, were investigated for the fusion proteins by western blot. A protein band of 73 kDa predicted to be the fusion protein and a non-specific one of 56 kDa were detected. Staining of the fusion proteins expressing cells with corresponding antibodies revealed their presence of NNVRNA2, but varied the intensity of detected signals from cell to cell by confocal laser scanning fluorescence microscopy. The predicted fusion proteins tertiary structure also confirmed exposed conformation of the fusion protein on the cell wall. In this study, the capsid proteins from the red-spotted grouper nervous necrosis virus were successfully expressed on the cell surface of P. pastoris but still low levels of fusion protein expression. Further studies are required to optimize fully surface protein expression prior to evaluate the possible use of the constructed recombinant yeast as an oral vaccine against RG-NNV infection.

Transcriptomic profiles of striped snakehead fish cells (SSN-1) infected with red-spotted grouper nervous necrosis virus (RGNNV) with an emphasis on apoptosis pathway

Nervous necrosis virus (NNV), the causative agent of viral nervous necrosis (VNN) disease, has caused mass mortality of cultured marine and freshwater fish worldwide, resulting in enormous economic losses in the aquaculture industry. However, the molecular mechanisms underlying the pathogenicity of NNV are still poorly understood. In this study, the transcriptomic profiles of striped snakehead fish (Channa striatus) cells (SSN-1) infected with red-spotted grouper NNV (RGNNV) were investigated using deep RNA sequencing technique. From 254,955,234 raw reads, a total of 253,338,544 clean reads were obtained and they were assembled into 93,372 unigenes. Differentially expressed genes (DEGs) were identified from RGNNV-infected or mock-infected SSN-1 cells, including 1184 up-regulated and 1456 down-regulated genes at 3 h (h) post of infection (poi), and 1138 up-regulated and 2073 down-regulated genes at 24 h poi, respectively. These DEGs were involved in many pathways related to viral pathogenesis, including retinoic acid-inducible gene I (RIG-I) like receptors pathway, apoptosis pathway, oxidative phosphorylation, PI3K-Akt signaling pathway, and MAPK signaling pathway. Subsequent analysis focusing on the apoptosis pathway showed that the expression of Endonuclease G (EndoG) was up-regulated upon RGNNV infection at both 3 and 24 h poi. Therefore, EndoG gene was cloned and its function was further characterized. The results showed that over-expression of EndoG could also induce cellular apoptosis in SSN-1 cells, indicating that RGNNV infection might induce apoptosis of SSN-1 cells via EndoG-associated mitochondrial pathway. These results will shed a new light on the pathogenesis of NNV.

Molecular Basis for Antigenic Diversity of Genus Betanodavirus.

Betanodaviruses are the causative agents of viral nervous necrosis (VNN), a devastating disease for the Mediterranean mariculture. Four different betanodavirus species are recognized, Striped jack-, Redspotted grouper-, Tiger puffer-, and Barfin flounder nervous necrosis virus (SJNNV, RGNNV, TPNNV and BFNNV), but there is little knowledge on their antigenic properties. In order to describe the serological relationships among different betanodavirus genotypes, serum neutralization assays were performed using rabbit polyclonal antisera against eight fish nodaviruses that cover a wide species-, temporal-, spatial- and genetic range. The results indicate that the SJNNV and RGNNV are antigenically distinct, constituting serotypes A and C, respectively. The TPNNV and BFNNV, the latter representing cold-water betanodaviruses, are antigenically related and cluster within serotype B. The reassortant viruses RGNNV/SJNNV and SJNNV/RGNNV group within serotypes A and C, respectively, indicating that the coat protein encoded by RNA2 acts as major immunoreactivity determinant. Immunostaining of in vitro expressed wild type and chimeric capsid proteins between the RGNNV and the SJNNV species indicated that the C-terminal part of the capsid protein retains the immunoreactive portion. The amino acid (aa) residues determining RGNNV and SJNNV antigenic diversity were mapped to aa residues 217–256 and aa 257–341, respectively. Neutralization of reverse genetics derived chimeric viruses indicated that these areas determine the neutralizing epitopes. The data obtained are crucial for the development of targeted serological tests for the diagnosis of VNN, and informative for development of cross-protective vaccines against various betanodavirus genotypes.

Cell Culture Isolation of Piscine Nodavirus (Betanodavirus) in Fish-Rearing Seawater.

Piscine nodavirus (betanodavirus) is the causative agent of viral nervous necrosis (VNN) in a variety of cultured fish species, particularly marine fish. In the present study, we developed a sensitive method for cell culture isolation of the virus from seawater and applied the method to a spontaneous fish-rearing environment. The virus in seawater was concentrated by an iron-based flocculation method and subjected to isolation with E-11 cells. A real-time reverse transcriptase PCR (RT-PCR) assay was used to quantify the virus in water. After spiking into seawater was performed, a betanodavirus strain (red spotted grouper nervous necrosis virus [RGNNV] genotype) was effectively recovered in the E-11 cells at a detection limit of approximately 10(5)copies (equivalent to 10(2)50% tissue culture infective doses [TCID50])/liter seawater. In an experimental infection of juvenile sevenband grouper (Epinephelus septemfasciatus) with the virus, the virus was isolated from the drainage of a fish-rearing tank when the virus level in water was at least approximately 10(5)copies/liter. The application of this method to seven band grouper-rearing floating net pens, where VNN prevailed, resulted in the successful isolation of the virus from seawater. No differences were found in the partial sequences of the coat protein gene (RNA2) between the clinical virus isolates of dead fish and the cell-cultured virus isolates from seawater, and the viruses were identified as RGNNV. The infection experiment showed that the virus isolates from seawater were virulent to seven band grouper. These results showed direct evidence of the horizontal transmission of betanodavirus via rearing water in marine aquaculture.

Prokaryotic Production of Virus-Like Particle Vaccine of Betanodavirus.

Betanodaviruses are the causative agents of viral nervous necrosis (VNN), a serious disease of cultured marine fish worldwide. To control this disease, vaccines of subunit capsid proteins (recombinant proteins or peptides), inactivated viruses, and virus-like particles (VLPs) were developed. VLP, which is highly similar to the wild-type virus in virion structure and contains no viral genome, was proved as one of the good and safe vaccines that can activate humoral immune response in the long term and induce cellular and innate immunities in the early stage post-immunization. The VLP vaccines can be expressed in vitro either by Baculovirus-based or yeast-based eukaryotic system or by bacterial expression system. In this chapter, the prokaryotic expression and the subsequent purification of VLP of betanodavirus orange-spotted grouper nervous necrosis virus (OGNNV) are presented.

Effect of the coexistence on the replication of striped jack nervous necrosis virus (SJNNV) and red-spotted grouper nervous necrosis virus (RGNNV) using an in vitro approach

Summary Viral nervous necrosis virus (VNNV) is the aetiological agent of viral nervous necrosis (VNN), a widespread disease affecting different marine and freshwater fish species. Striped jack nervous necrosis virus (SJNNV) and red-spotted grouper nervous necrosis virus (RGNNV) are the only genotypes of the Betanodavirus genus recorded in the Iberian Peninsula to date, but a high percentage of wild specimens simultaneously carrying both genotypes has been recently reported. The coexistence of the two viruses may affect the course of both viral infections. In the present study, viral genome quantification by two absolute real-time PCR protocols has been performed to characterise the effect of the RGNNV-SJNNV coexistence (coinfection and superinfection) on the replication of each genotype in E-11 cells. This is the first study showing the effect of the coexistence on the viral replication of two genotypes within the Betanodavirus genus. The results obtained in vitro showed the partial inhibition of SJNNV replication by the coexistence with RGNNV, whereas RGNNV replication was favoured in coinfection or superinfection with SJNNV.

In vitro study of the replication capacity of the RGNNV and the SJNNV betanodavirus genotypes and their natural reassortants in response to temperature.

Betanodaviruses are the causative agents of viral nervous necrosis and affect a broad range of fish species worldwide. Their bi-segmented genome is composed of the RNA1 and the RNA2 molecules encoding the viral polymerase and the coat protein, respectively. In southern Europe the presence of the RGNNV and the SJNNV genotypes, and the RGNNV/SJNNV and RGNNV/SJNNV reassortants has been documented. Several studies have reported a correlation between water temperature and disease onset. To explore the replication efficiency of betanodaviruses with different genomes in relation to temperature and to understand the role of genetic reassortment on viral phenotype, RGNNV, SJNNV, RGNNV/SJNNV and RGNNV/SJNNV field isolates were fully sequenced, and growth curves generated in vitro at four different temperatures (15, 20, 25, 30 °C) were developed for each isolate. The data obtained, corroborated by statistical analysis, demonstrated that viral titres of diverse betanodavirus genotypes varied significantly in relation to the incubation temperature of the culture. In particular, at 30 °C betanodaviruses under investigation presented different phenotypes, and viruses containing the RNA1 of the RGNNV genotype showed the best replication efficiency. Laboratory results demonstrated that viruses clustering within the same genotype based on the polymerase gene, possess similar growth kinetics in response to temperature, thus highlighting the key role of RNA1 in controlling viral replication at different environmental conditions. The results generated might have practical implications for the inference of viral phenotype according to genetic features and may contribute to a better understanding of betanodavirus ecology.

Immune responses of orange-spotted grouper, Epinephelus coioides, against virus-like particles of betanodavirus produced in Escherichia coli

Betanodaviruses are the causative agents of viral nervous necrosis (VNN), a serious disease of cultured marine fish worldwide. Virus-like particles (VLPs) are one of the good novel vaccine candidates to control this disease. Until now, betanodavirus vaccine studies mainly focused on the humoral immune response and mortality after virus challenge. However, little is known about the activation of genes responsible for cellular and innate immunity by vaccines. In the present study, VLPs of orange-spotted grouper nervous necrosis virus (OGNNV) were produced in prokaryotes and their ability to enter Asian sea bass cells was the same as native virus, suggesting that they possess a similar structure to OGNNV. VLPs immunogenicity was then determined by intramuscularly vaccinating Epinephelus coioides at different concentrations (1.5 or 15μgg−1 fish body weight, FBW) and immunizing frequencies (administration once, twice and thrice). A single vaccination with the dosage of 1.5μgg−1 FBW is enough to provoke high titer antibodies (average 3 fold higher than that of negative control) with strong neutralizing antibody titer as early as 1 week post immunization. Furthermore, quantitative PCR analysis revealed that eleven genes associated with humoral, cellular and innate immunities were up-regulated in the liver, spleen and head kidney at 12h post immunization, correlating with the early antibody response. In conclusion, we demonstrated that VLP vaccination induced humoral immune responses and activated genes associated with cellular and innate immunity against betanodavirus infection in orange-spotted grouper.

Evaluation of antiviral activity of Oligonol, an extract of Litchi chinensis, against betanodavirus

Betanodaviruses, members of the family Nodaviridae, are the causal agents of viral nervous necrosis (VNN) in many species of marine farmed fish. In the aquaculture industry, outbreaks of betanodavirus infection result in devastating damage and heavy economic losses. Although an urgent need exists to develop drugs against betanodavirus infection, there have been few reports about antibetanodavirus drugs. In this study, we examined the inhibitory effect of Oligonol, a purified phenolic extract from lychee fruit, on betanodavirus infection in fish cells. Oligonol significantly inhibited replication of betanodavirus (EC(50) = 0.9-1.8 μg/mL) as shown by the reduction of the virus-induced cytopathogenic effect (CPE) and the protection of cells in the crystal violet staining assay. The inhibition was dose dependent. A time-of-addition assay indicated that Oligonol's action takes place at an early stage of the viral infection. According to an attachment inhibition assay, it is possible that Oligonol partially inhibits attachment of the virion to the cell. Our data show that Oligonol could serve as an antiviral agent against betanodavirus.

Establishment and characterization of an epithelial cell line from thymus of Catla catla (Hamilton, 1822)

A cell line, CTE, derived from catla (Catla catla) thymus has been established by explant method and subcultured for more than 70 passages over a period of 400days. The cell line has been maintained in L-15 (Leibovitz) medium supplemented with 10% fetal bovine serum. CTE cell line consists of homogeneous population of epithelial-like cells and grows optimally at 28°C. Karyotype analysis revealed that the modal chromosome number of CTE cells was 50. Partial amplification, sequencing and alignment of fragments of two mitochondrial genes 16S rRNA and COI confirmed that CTE cell line originated from catla. Significant green fluorescent signals were observed when the cell line was transfected with phrGFP II-N mammalian expression vector, indicating its potential utility for transgenic and genetic manipulation studies. The CTE cells showed strong positivity for cytokeratin, indicating that cell line was epithelial in nature. The flow cytometric analysis of cell line revealed a higher number of cells in S-phase at 48h, suggesting a high growth rate. The extracellular products of Vibrio cholerae MTCC 3904 were toxic to the CTE cells. This cell line was not susceptible to fish betanodavirus, the causative agent of viral nervous necrosis in a large variety of marine fish.

Disease control efficacy of synthetic double-stranded RNA Poly(I:C) administration for viral nervous necrosis (VNN) in sevenband grouper (Epinephelus septemfasciatus)

Nervous necrosis virus (NNV), the causative agent of viral nervous necrosis (VNN), is an indigenous virus in Korean coastal areas and a threat to the farming industry. In the present study, we investigated the curative and preventive effects against VNN in sevenband grouper (Epinephelus septemfasciatus) using polyinosinic polycytidylic acid [Poly(I:C)]. Fish challenged with brain-filtrates from VNN-affected fish at 0, 2, and 4d after Poly(I:C) injection showed <10% cumulative mortality, whereas fish that were not administered Poly(I:C) but challenged 14d after the Poly(I:C) injection showed 100% and 80% cumulative mortality, respectively. The surviving fish from the primary challenge with brain-filtrates following Poly(I:C) injection were highly protected from re-challenge at 15 and 30 d post-primary challenge. Moreover, fish pre-immersed with red-spotted grouper nervous necrosis virus (RGNNV) survived following Poly(I:C) administration, even though their clinical signs did not recover. From these results, it was confirmed that sevenband grouper were highly protected from RGNNV challenge by Poly(I:C) administration.

Potentiality of a live vaccine with nervous necrosis virus (NNV) for sevenband grouper Epinephelus septemfasciatus at a low rearing temperature

Nervous necrosis virus (NNV) is the causative agent of viral nervous necrosis (VNN), one of the most serious diseases in over 30 species of cultured marine fishes worldwide. Although several kinds of NNV vaccines have been developed, none of these vaccines have been yet marketed. Here, we demonstrate the potentiality of a live NNV vaccine for sevenband grouper Epinephelus septemfasciatus at a low rearing temperature (17°C). Moreover, we investigated the kinetics of NNV infectivity titer in fish reared at low and optimum temperatures (17°C and 26°C) for VNN onset to determine why sevenband grouper reared at 17°C survive NNV infection. In pathogenicity tests of NNV, fish mortality was reduced by decreasing the fish rearing temperature, and no mortality was observed in fish reared at 17°C regardless of the infection method. During fish acclimation to the optimum temperature of VNN onset (26°C), increased mortalities were observed in the survivors from the 1st NNV-infection. Little or no mortality was observed in the 2nd NNV-infection. Thus, it was demonstrated that the survivors from the 1st NNV-infection mounted a specific protective immune response against NNV. Especially, in the fish infected with NNV by immersion at 17°C, only two out of 30 fish died until the end of the 2nd infection (total survival rate: 93.3%), suggesting a positive potentiality for a live NNV vaccine. In the analysis of NNV kinetics in the fish reared at 26°C, NNV rapidly multiplied up to ≥ 10(9)TCID(50)g(-1) before fish began to die, and the critical level of NNV was around 10(10)TCID(50)g(-1). Probability of NNV multiplication reduced by decreasing the inoculated NNV dose, but NNV multiplication rate was independent of the NNV dose. The threshold of NNV for fish mounting a protective immune response was around >10(4)TCID(50)g(-1). Against this, in the fish reared at 17°C, NNV slowly multiplied in comparison with that in fish at 26°C. NNV titer in the peak was at 10(7.1 ± 1.4)TCID(50)g(-1), which was far behind the critical level of NNV but still greatly above the threshold level (10(4)TCID(50)g(-1)). Thus, it was demonstrated that the multiplication rate of NNV in vivo was strongly correlated to NNV virulence and fish mortality, and down-regulation of NNV multiplication in fish reared at 17°C enabled control of VNN onset for development of a live NNV vaccine.

Required dose of fish nervous necrosis virus (NNV) for Poly(I:C) immunization of sevenband grouper Epinephelus septemfasciatus

It was recently reported that Poly(I:C) immunization of fish confers protection against viral infection. In this process, fish are immunized with a live pathogenic virus followed by administration of Poly(I:C), which induces a transient, non-specific antiviral state. As a result, fish survive the initial immunization with live virus, which would otherwise be lethal. In the present study, we tried to determine the exact dosage of red-spotted grouper nervous necrosis virus (RGNNV), the causative agent of viral nervous necrosis, required for Poly(I:C) immunization of sevenband grouper Epinephelus septemfasciatus. In Poly(I:C) immunization with more than 90% relative percentage survival (RPS) against RGNNV challenge with 104.3 TCID50/fish, approximately 104.0 TCID50/fish or greater of RGNNV was administered by intramuscular (IM) injection, whereas 105.3 TCID50/fish or greater of RGNNV was required for immersion methods. It was concluded that the degree of RGNNV infection must be similar to a fatal dose in order for the fish to become immune to RGNNV. Interestingly, antibody against RGNNV was not detected in the sera of fish immunized by immersion. Thus, Poly(I:C) immunization by immersion may have some interesting effects on the epidermal immune response in fish.
