Acquisition and retention of enhanced active avoidance are unaffected by interference with serotonergic activity

Pre-training administration of p-chloroamphetamine (PCA) produces reliable deficits of avoidance learning. When animals are trained in inhibitory avoidance with relatively high foot-shock intensities, other amnesic treatments have no effect. The present experiment was conducted to determine if this protective effect of high foot shock is also observed after administration of PCA (10 mg/kg, i.p., injected 7 days before training; this dose produces a lesion of central serotonin neurons). Rats were trained in active avoidance (a single 20-trial session), administering shocks of 0.6, 0.8, 1.0, or 1.4 mA to independent groups of rats. When compared to saline-injected groups trained with the same intensities, PCA produced a significant learning deficit in the low foot-shock groups, but not in the high foot-shock animals. These results indicate that the dose of PCA administered, which is known to deplete cerebral serotonin, does not interfere with acquisition and retention of enhanced active avoidance training.

There have been few pharmacological studies of serotonergic system dynamics in insects. A more precise knowledge of the response of serotonergic neurons to drugs will contribute to understanding of the role of this neurotransmitter in insect behaviour. The present work was carried out to study several aspects of serotonin (5-HT) metabolism and release in an insect, the butterfly Inachis io. The effects of a single intra-abdominal injection of reserpine (30 pg/insect) or p-chloroamphetamine (50 microg/insect) on cerebral ganglia 5-HT metabolism and release were studied. After reserpine injection a depletion of 5-HT stores concomitant with an increase in N-acetylserotonin levels was observed, but not significant alteration of extraneuronal 5-HT release was observed. Administration of p-chloroamphetamine (PCA) induced extraneuronal 5-HT release, together with inhibition of its reuptake. Finally, a single injection of p-chloroamphetamine in reserpine-treated insects was able to induce new release of 5-HT. Reserpine interferes with the vesicular storage of 5-HT, but does not affect the process of neuronal release, while PCA induces the synaptic release of 5-HT and inhibits its reuptake. These effects are similar to those observed in mammals.

Activation by p-chloroamphetamine of the spinal ejaculatory pattern generator in anaesthetized male rats

In urethane-anesthetized male rats, a branch of the hypogastric nerve was shown, anatomically and electrophysiologically, to supply the vas deferens. Recordings from this nerve revealed a low level of tonic activity, which was predominantly efferent motor activity. Administration of p-chloroamphetamine i.v., elicited a rhythmic burst of neuronal activity, coherent with rhythmic pressure increases in the vas deferens and contractions of the bulbospongiosus muscles, which together comprise ejaculation. This response to p-chloroamphetamine was still present after complete transection of the spinal cord at T8–T9. These data indicate that p-chloroamphetamine is capable of activating the spinal neuronal circuits that generate the pattern of autonomic and somatic responses similar to those of sexual climax. Furthermore based on the best documented action of p-chloroamphetamine, the results suggest that the excitability of the pattern generator is regulated by serotonergic, dopaminergic or noradrenergic receptors in the lumbosacral spinal cord. We conclude this animal model will enable robust studies of the pharmacology and physiology of central neural mechanisms involved in ejaculation and sexual climax.

3,4-methylenedioxymethamphetamine (MDMA) administration to rats decreases brain tissue serotonin but not serotonin transporter protein and glial fibrillary acidic protein.

Previous experiments conducted in this laboratory showed that administration of high-dose D-fenfluramine (D-FEN) and p-chloroamphetamine (PCA) decreased 5-HT transporter (SERT) binding and tissue 5-HT by 30-60% in caudate and whole brain tissue 2 days and 2 weeks after drug administration. However, protein expression as determined by Western blot analysis did not change in either tissue or time point, except for a 30% decrease in the caudate 2 days after PCA administration. In the present study, we studied the effect of MDMA and 5,7-dihydroxytryptamine (5,7-DHT) on tissue 5-HT levels and the protein expression level of SERT and glial fibrillary acidic protein (GFAP), a validated neurotoxicity marker. MDMA administration decreases SERT expression. Two weeks after MDMA administration (7.5 mg/kg i.p., q 2 h x 3 doses) or 2 weeks after i.c.v. administration of 5,7,-DHT (150 microg/rat), male Sprague-Dawley rats were sacrificed and the caudate, cortex, and hippocampal tissue collected. Western blots for SERT and GFAP were generated using published methods. Tissue 5-HT levels were determined by HPLC coupled to electrochemical detection. MDMA treatment decreased tissue 5-HT in cortex, hippocampus, and caudate by about 50%. However, MDMA treatment had no significant effect on expression level of SERT and GFAP in any brain region. In contrast, 5,7-DHT reduced tissue 5-HT by more than 90%, decreased SERT protein expression by 20-35%, and increased GFAP by 30-39%. These data suggest the MDMA treatment regimen used here does not cause degeneration of 5-HT nerve terminals. Viewed collectively with our previous results and other published data, these data indicate that MDMA-induced persistent 5-HT depletion may occur in the absence of axotomy.

Inhibition of neuronal nitric oxide synthase suppresses the maintenance but not the induction of psychomotor sensitization to MDMA (‘Ecstasy’) and p-chloroamphetamine in mice

Repeated exposure to psychostimulants such as cocaine and amphetamines results in behavioral sensitization, a paradigm thought to be relevant to drug craving and addiction in humans. We have previously shown that the induction, expression, and maintenance of psychomotor sensitization to cocaine, methamphetamine, and methylphenidate (indirect dopamine agonists) are blocked by co-administration of the neuronal nitric oxide synthase (nNOS) inhibitor 7-nitroindazole (7-NI). In the present study, we investigated the effects of 7-NI on the induction, expression, and maintenance of psychomotor sensitization to 3,4-methylenedioxymethamphetamine (MDMA; ‘Ecstasy’) and p-chloroamphetamine (PCA). The following observations are reported: (a) Repeated administration of MDMA (10 mg/kg) and PCA (5 mg/kg) to Swiss Webster mice for six consecutive days caused a 3-fold increase in the psychomotor stimulating effect of the drugs on day 6 compared to day 1. (b) Pretreatment with 7-NI (25 mg/kg) did not affect the induction and expression of sensitization to MDMA and PCA. (c) Pretreatment with 7-NI did, however, suppress the enduring sensitized response to challenge injections of MDMA and PCA which was observed in mice pretreated with vehicle instead of 7-NI. (d) Unlike other psychostimulants, MDMA and PCA treatment did not produce conditioned (context-dependent) hyperlocomotion. These findings, coupled with our previous studies, suggest the following: (a) The induction and expression of psychomotor sensitization to MDMA and PCA are independent of nNOS activity and involve primarily serotonergic transmission. (b) The maintenance of psychomotor sensitization is dependent on intact nNOS activity and involves primarily dopaminergic transmission.

Enhanced inhibitory avoidance training protects against the amnesic effect of p–chloroamphetamine

The contribution of acetylcholine (ACh) to memory processing is well documented, but it has been proposed that it is not necessary for memory consolidation after an enhanced learning experience. It has been suggested that serotonin (5-HT) interacts with ACh during memory consolidation, although the nature of this interaction is unknown in the case of strong learning. As an initial approach to the study of these interactions, we determined whether training of inhibitory avoidance using relatively high aversive stimulation protects against the typical retention deficits produced by pre-training administration of the 5-HT releaser p–chloroamphetamine (PCA). Rats were trained after intraperitoneal administration of PCA or isotonic saline, using 2.0, 2.5, 3.0 or 3.5 mA and retention of the task was measured 24 h later. A significant amnesic state was observed only in the PCA groups that had been trained with the two lower intensities. These results indicate that 5-HT systems behave similarly to ACh systems, in the sense that the amnesic effect produced by interference with their physiological activity may be cancelled when animals are submitted to an intense learning situation.

Characterization of p-chloroamphetamine-induced penile erection and ejaculation in anesthetized rats

Methodological shortcomings present in elicitation of male sexual reflexes in anesthetized animals. The present study has demonstrated, however, that intraperitoneal (i.p.) injection of p-chloroamphetamine (PCA), an indirect serotonin (5-HT) agonist, elicited simultaneously both penile erection and ejaculation in anesthetized rats. PCA (2.5–10.0 mg/kg, i.p.) caused an intermittent cluster of genital responses consisting of penile erection, glans erections, and penile cups, which closely resembles the response observed during the ex copula tests in unanesthetized rats. Measurements of intracavernous penile pressure showed that rhythmic changes in penile pressure were produced by PCA, together with glans erections and penile cups. PCA also caused a frequent ejaculations and the weighing of ejaculate accumulated over 0.5 hr was increased in a bell-shaped pattern, and the maximum effect was observed at 5.0 mg/kg. Pretreatment with p-chlorophenylalanine, a serotonin (5-HT) - synthesis inhibitor, significantly inhibited the expression of PCA-induced penile erection and ejaculation, while acute spinal transection at thoracic level did not affect the sexual responses. These results indicate that PCA-induced penile erection and ejaculation in anesthetized rats are mainly produced by the release of 5-HT, which is limited to the lower spinal cord and/or the peripheral sites. Furthermore, the sexual responses can be easily and reliably elicited by administration of PCA, which may be useful for the study of the mechanisms underlying male sexual functions.

Long-term modulation of presynaptic 5-HT-output: experimentally induced changes in cortical 5-HT-transporter density, tryptophan hydroxylase content and 5-HT innervation density.

Whereas experimentally induced long-term changes of postsynaptic mechanisms of 5-HT neurotransmission have been studied in great detail, much less is currently known about the effects of certain treatments on the presynaptic components governing the output of 5-HT in individual brain regions. This contribution summarizes the results of a series of experiments on the influence of different physiologic and pharmacologic manipulations on three different parameters of 5-HT presynapses, 5-HT transporter density, tryptophan hydroxylase content, and serotonin level in the rat frontal cortex. The combined measurement of several parameters of 5-HT presynapses allows to differentiate between treatments which exclusively affect the density of 5-HT transporters (long-term food restriction), which exclusively affect the level of tryptophan hydroxylase apoenzyme (imipramine treatment of olfactory bulbectomized rats) or which cause a parallel increase (bulbectomy, chronic administration of tranylcypramine to rats with chemical lesions of their cortical 5-HT innervation) or a parallel decrease (administration of p-chloroamphetamine) of both parameters, indicating treatment-induced changes in the density of 5-HT presynapses in the frontal cortex. Each of these changes may lead to an altered output of serotonergic afferences, and may therefore act to either potentiate or to attenuate the impact of serotonin-mediated effects on the activity of local networks located in a certain brain region.

P-Chloroamphetamine- and d-Fenfluramine-Induced Brain Serotonin Release in Experimental Portal-Systemic Encephalopathy

Portal-systemic encephalopathy (PSE) is associated with increased brain turnover of serotonin (5-HT) in vivo but the brain 5-HT output seems to be unaltered. Recent results suggest, however, that an augmented neocortical 5-HT release in experimental chronic PSE may prevail under certain conditions. In the present study, neocortical extracellular 5-HT and 5-hydroxyindoleacetic-3-acid (5-HIAA) levels were measured in portacaval shunted (PCS) rats and sham-operated controls following local administration of p-chloroamphetamine (pCA) and d-fenfluramine (dFEN), two specific 5-HT releasing agents. The basal neocortical extracellular 5-HT concentrations were unaltered and the 5-HIAA levels were elevated in experimental PSE, supporting an unchanged brain 5-HT output despite elevated brain 5-HT metabolism. Perfusion with pCA or dFEN (5 microM; one 20-min pulse) produced marked increases in brain 5-HT release both in PCS and sham-operated rats compared with corresponding basal values. While no difference in the 5-HT response to dFEN administration was seen between sham (5-HT levels increased by 330%) and PCS (500%) rats, a clear difference (p<0.05) in the brain 5-HT output was observed between the two experimental groups following pCA perfusion (sham, 1100% versus PCS, 1470%). These results support our previous contention of an enhanced neocortical 5-HT output in experimental chronic PSE under certain pharmacological conditions.

Effects of 5-HT 4 Receptor Agonists and Antagonists in Learning

In the present work, the effects of pre- or post-training (ip) injection of BIMU1 and BIMU8 (5-HT 4 agonists) were figured out in the autoshaping learning task. Furthermore, the post-training effects of these agonists after treatment with SDZ 205-557 and GR 125487D (5-HT 4 antagonists) or p-Chloroamphetamine (PCA) were also explored. Animals were individually trained in a lever-press response on the autoshaping task and 24 hours later were tested. The results showed that pre-training injection of BIMU1 (5–20 mg/Kg) or BIMU8 (20 mg/Kg) increased the CR; in contrast, the post-training administration of BIMU1 (10–20 mg/Kg) or BIMU8 (5 and 20 mg/Kg) decreased it. Further experiments revealed that the post-training injections of SDZ 205-557 (1.0–10.0 mg/Kg) or GR 125487D (0.39–1.56 mg/Kg) by themselves did not alter the CR. When BIMU1 or BIMU8 was administered to rats pretreated with SDZ 205-557 (10 mg/Kg) or GR 125487D (0.78 mg/Kg), the decrement induced by 5-HT 4 the agonists was reversed; in contrast, the administration of PCA failed to modify the CR or the agonist-induced responses. The findings showed that the pre-training stimulation of 5-HT 4 receptors enhanced the acquisition of CR, while, post-training activation of 5-HT 4 receptors, impaired the consolidation of learning. The latter effect was not altered by PCA pretreatment. The data show that 5-HT 4 receptors are involved in the acquisition and consolidation of learning. It seems that postsynaptic 5-HT 4 receptors are involved in the latter effect.

P-chloroamphetamine induces C-fos in rat brain: A study of serotonin2A/2C receptor function

Recent studies suggest that a class of substituted amphetamines, which includes p-chloroamphetamine, causes an acute release of serotonin (5-hydroxytryptamine) and appears to act preferentially on axons arising from the dorsal raphe nucleus. The postsynaptic targets of these axons are not well characterized, but they have been localized in close proximity to the distribution of serotonin2A receptor binding sites. In the present study, c-fos immunocytochemistry has been used to investigate this anatomical relationship further. Administration of p-chloroamphetamine or the serotonin2A/2C receptor agonist 1-(2,5-dimethoxy-4-iodophenyl)-2-aminopropane to rats resulted in similar patterns of Fos-like immunoreactivity in some, but not all, forebrain areas. Areas which expressed Fos following either treatment included cerebral cortex, claustrum, amygdala and nucleus accumbens. A particularly close match was seen in layer Va of the somatosensory cortex. No specificity of p-chloroamphetamine for dorsal raphe nucleus-innervated areas was noted. Prior treatment of animals with p-chloroamphetamine two weeks before a second challenge with the same drug, or with the serotonin2A/2C receptor antagonist ritanserin 30 min before p-chloroamphetamine challenge, resulted in an attenuation of p-chloroamphetamine-induced Fos-like immunoreactivity in the olfactory tubercle, the islands of Calleja and the caudate-putamen. The reduction was most noticeable in layer Va of the somatosensory cortex. The results of this study indicate that a close anatomical correlation may exist between the fine serotonin axon terminals that show vulnerability to the neurotoxic effects of p-chloroamphetamine and serotonin2A receptors in some brain regions. This association may prove to be important in explaining the actions of certain psychotropic drugs, for example in the control of affective states.

Lack of neuroprotective effect of sigma receptor ligands in the neurotoxicity of p-chloroamphetamine in rat brain.

We studied the mechanism of antagonism of p-chloroamphetamine-induced neurotoxicity by dextromethorphan. The pretreatment with potent and selective sigma receptor ligands, 4-phenyl-4-(1-phenylbutyl)piperidine (4-PPBP) and N,N-dipropyl-2-[4-methoxy-3-(2-phenylethoxy)phenyl]-ethylamine monohydrochloride (NE-100), did not alter the reduction of 5-hydroxytryptamine and 5-hydroxytryptamine and 5-hydroxyindoleacetic acid in the cerebral cortex by repeated administration of p-chloroamphetamine. These results suggest that sigma receptors might not play a significant role in the antagonism of p-chloroamphetamine-induced neurotoxicity by dextromethorphan.

Involvement of central serotonergic pathways in analgesia elicited by salmon calcitonin in the mouse

The contribution of central serotonergic pathways to the analgesic activity induced by salmon calcitonin in the writhing test was investigated. Salmon calcitonin was administered to mice after lesioning of the ascending and descending serotonergic pathways by means of i.p. administration of p-chloroamphetamine (40 mg/kg, for 2 days) or p-chlorophenylalanine (300 mg/kg, for 3 days). The analgesic effect induced by salmon calcitonin at the doses of 10 and 20 IU/kg was not evident in mice previously treated with p-chloroamphetamine or p-chlorophenylalanine. However, the analgesic effect of salmon calcitonin 40 IU/kg was not significantly modified by p-chloroamphetamine or p-chlorophenylalanine pretreatment. Salmon calcitonin did not alter the depletion of 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid after p-chloroamphetamine or p-chlorophenylalanine administration. Similarly, this hormone did not change the NSD 1015-induced accumulation of 5-hydroxytryptophan or the tranylcypromine-induced accumulation of 5-HT. These results indicate that although salmon calcitonin does not influence the synthesis and metabolism of 5-HT, it does require the integrity of the serotonergic system in order to cause analgesia.

3H]paroxetine binding and serotonin content of rat cortical areas, hippocampus, neostriatum, ventral mesencephalic tegmentum, and midbrain raphe nuclei region following p-chlorophenylalanine and p-chloroamphetamine treatment.

The agents p-chlorophenylalanine (PCPA) and p-chloroamphetamine (PCA) deplete brain serotonin (5-HT) levels by two different mechanisms; PCPA inhibits the enzyme tryptophan hydroxylase, whereas PCA has a neurotoxic action on certain 5-HT neurons. The parameters of [3H]paroxetine binding to homogenates prepared from the cerebral cortex of rats treated with PCPA, PCA, or saline; vehicle were investigated. The tissue concentrations of 5-HT and 5-hydroxyindole-3-acetic acid (5-HIAA) were also determined by HPLC in the same brain samples. After PCPA treatment, neither the maximum binding capacity (Bmax) nor the dissociation constant (KD) of [3H]paroxetine for the 5-HT uptake recognition site differed from controls despite a substantial reduction in the concentration of 5-HT and 5-HIAA. In contrast, significant changes in both the Bmax and KD values were observed in the cerebral cortex of rats treated with PCA. Furthermore, [3H]paroxetine binding and tissue concentrations of 5-HT and 5-HIAA were measured in the following different regions of the rat brain: cingulate, parietal, and visual cortical areas; dorsal and ventral hippocampus; rostral and caudal halves of neostriatum; ventral mesencephalic tegmentum; and midbrain raphe nuclei region after administration of PCPA, PCA, or saline vehicle. There was an excellent correlation between regional 5-HT levels and specific [3H]paroxetine binding in control and PCA-treated rats although this correlation was lost after PCPA treatment. Under these conditions, the 5-HT innervation remains unchanged whereas the concentration of 5-HT and 5-HIAA is greatly reduced. Thus, [3H]paroxetine binding appears to provide a reliable marker of 5-HT innervation density within the mammalian CNS.

Dual serotoninergic projections to forebrain in the rat: Morphologically distinct 5‐HT axon terminals exhibit differential vulnerability to neurotoxic amphetamine derivatives

The cerebral cortex of the rat and other mammals is innervated by two morphologically distinct classes of serotoninergic (5-HT) axon terminals: fine axons with minute varicosities and beaded axons characterized by large, spherical varicosities. Fine and beaded 5-HT axons exhibit different regional and laminar distributions in forebrain and arise from separate brainstem nuclei, the dorsal and median raphe nuclei, respectively. The present neuroanatomic study, based on immunocytochemical methods to visualize 5-HT axons, demonstrates that the two axon types differ markedly in their vulnerability to the neurotoxic amphetamine derivatives, methylenedioxyamphetamine (MDA), and p-chloroamphetamine (PCA). While both drugs cause extensive degeneration of fine 5-HT axons throughout forebrain, beaded 5-HT axons are consistently spared. Fine 5-HT axons, which richly innervate most regions of dorsal forebrain in control rats, are rarely seen 2 weeks after treatment with MDA or PCA; this loss of fine axons reflects a marked denervation that persists for months after drug administration. The serotoninergic axon terminals remaining after MDA or PCA administration are almost entirely of the beaded type and appear to be unaffected by both drugs. Over a wide range of doses (2.5-40 mg/kg PCA) and survival times (2 weeks to 2 months), these spared 5-HT axons with large, spherical varicosities cannot be distinguished from the normal, beaded 5-HT axons in control rats by morphologic criteria. Moreover, beaded 5-HT axons exhibit a highly characteristic regional distribution which is the same in control as in MDA- and PCA-treated rats: these axons innervate specific zones or layers within parietal and occipital cortex, hippocampus, cingulate cortex, entorhinal cortex, and the olfactory bulb, among other forebrain areas, and they form a dense plexus lining the ventricular system. Taken together, the results of this study demonstrate that fine 5-HT axons are highly vulnerable to the neurotoxic effects of the amphetamine derivatives MDA and PCA, while beaded 5-HT axons are markedly resistant. These findings are consistent with the hypothesis that there are two anatomically and functionally distinct sets of serotoninergic neurons projecting to forebrain. While both of these neuronal systems utilize 5-HT as a neurotransmitter, they differ in several features: 1) origin from separate nuclei in the brainstem (the dorsal and median raphe), 2) two types of morphologically distinct axon terminals, 3) markedly different distribution and innervation patterns in forebrain, and 4) dissimilar pharmacological properties. The results further suggest that psychotropic amphetamine derivatives have a selective action upon fine serotoninergic axons that arise from the dorsal raphe nucleus.

Chloroamphetamine did not prevent the effects of chronic antidepressants on 5-hydroxytryptamine inhibition of forskolin-stimulated adenylate cyclase in rat hippocampus

Administration of p-chloroamphetamine (PCA, 10 mg/kg i.p. on two occasions) to rats resulted in a severe depletion of [ 3H]paroxetine binding sites, a measure of presynaptic serotonergic terminals, in both cortex and hippocampus, but did not affect [ 3H]8-hydroxy-2-(di-n-propylamino)tetralin ([ 3H]8-OH-DPAT) binding or 5-hydroxytryptamine (5-HT)-induced inhibition of forskolin-stimulated adenylate cyclase in hippocampal membranes. Administration of either imipramine (15 mg/kg i.p. for 2 weeks) or lithium (0.2% for 2 weeks) to PCA-treated rats did not affect [ 3H]8-OH-DPAT binding but reduced the degree of inhibition of forskolin-stimulated adenylate cyclase by 5-HT in hippocampal membranes. It is concluded that the effects of imipramine and Li + on 5-HT 1A receptor-mediated responses in the hippocampus are exerted postsynaptically, possibly at a level distal to the receptor.

Effects ofp-chloroamphetamine and a side-chain monofluorinated analogue on levels of indoles in rat brain

Levels of tryptophan (Trp), 5-hydroxytryptamine (5-HT), and 5-hydroxyindole-3-acetic acid (5-HIAA) in rat brain were measured at 1, 2, 4, and 24 hr after intraperitoneal administration of p-chloroamphetamine (pCA) (0.03 mmol/kg) or of p-cloro-α-fluoromethylphenylethylamine (fluoro-p-chloroamphetamine; FpCA) (0.05 or 0.10 mmol/kg). FpCA is less potent than pCA at producing depletions of 5-HT and elevations of Trp, and the effects with FpCA are shorter-lasting. pCA has been reported to possess good antidepressant properties but was withdrawn from clinical studies because of reported neurotoxicity to 5-HT systems in rat brain. The less potent effects of FpCA on 5-HT and 5-HIAA reduction shown here suggest that further research on this analogue as a potential antidepressant is warranted.

Disruption of neocortical lamina V neuronal bursts by serotonin in urethane anaesthetized rats.

Time-shared high speed cyclic voltametry using carbon fibre multibarrelled microelectrodes was used to monitor the concentration of 5-Hydroxytryptamine administered by iontophoresis to locations in lamina V of Sm l neocortex and to record spontaneous neuronal spike activity. In the absence of 5-Hydroxytryptamine at any one recording location the firing of two or more individual units was seen to be synchronized so that the pattern of multi-unit activity consisted of synchronized clusters of spike activity interspersed with period of neuronal silence. The repetition rate of such clusters of neuronal activity was seen to be between 0.5 and 4Hz. Maximum concentrations of 2.7 x 10(-7) M 5-Hydroxytryptamine produced by iontophoresis disrupted synchronized neuronal cluster activity. 5-Hydroxytryptamine at a concentration of 6.2 x 10(-8) M resulted in a greater than 50% inhibition of activity for 47 single units but a change in firing pattern from cluster restricted high frequency activity to a continuous mode of firing for a separate population of 11 units. Intraperitoneal administration of P-Chloroamphetamine produced similar changes of neuronal firing and hence loss of synchrony.

Evidence for dual serotonergic projections to neocortex: Axons from the dorsal and median raphe nuclei are differentially vulnerable to the neurotoxin p-chloroamphetamine (PCA)

Previous studies have shown that there are morphologically dissimilar serotonergic (5-HT) axon types in rat cerebral cortex which are differentially sensitive to the neurotoxic effects of certain psychotropic drugs: methylenedioxyamphetamines (MDA and MDMA) and p-chloroamphetamine (PCA) cause degeneration of fine 5-HT axon terminals in cortex, while sparing beaded axons. Moreover, a recent anterograde transport study suggests that fine and beaded 5-HT axons arise from the dorsal raphe (DR) and median raphe (MR) nuclei, respectively. These data led us to propose that the DR projection to neocortex is selectively vulnerable to the neurotoxic effects of PCA, while the MR projection is resistant; this hypothesis was tested in the present study by comparing retrograde axonal transport of the fluorescent tracer Fluoro-Gold in PCA-treated and control rats. Using this method, only axons that survive PCA treatment can take up and transport the injected label back to the cell bodies of origin, thus allowing us to determine which raphe—cortical projections remain intact after PCA. The results show that PCA administration produces a loss of fine 5-HT axon terminals in neocortex and a concomitant reduction in the number of retrogradely labeled neurons in the DR (77% decrease), when compared to controls. In contrast, beaded 5-HT axon terminals are spared and the number of labeled neurons in the MR remains unchanged after PCA. These results demonstrate that DR and MR projections to cortex are differentially vulnerable to PCA: fine axon terminals arise from neurons in the Dr and are highly sensitive to the neurotoxic effects, whereas beaded axons from the MR are resistant. We therefore propose that there are two anatomically and functionally separate 5-HT projections to cortex having different (1) nuclei of origin, (2) axon morphology, (3) regional distributions, and (4) pharmacological properties. Since the mood-altering substances MDA, MDMA, and PCA act specifically upon 5-HT axon terminals from the dorsal raphe nucleus, DR neurons may be preferentially involved in the control of affective state.
