Effects of recombinant bovine somatotropin administration at breeding on cow, conceptus, and subsequent offspring performance of beef cattle.

The effects of administration of recombinant bovine ST (bST) on plasma hormone concentrations of cows, conceptus development, and postnatal calf performance were examined. Lactating beef cows ( = 190) were exposed to a fixed-time AI (TAI) protocol from d -10 to 0 (TAI on d 0). Cows were blocked by breed and stratified by days postpartum and then randomly assigned to receive, subcutaneously 1) 2 injections of saline (1 mL of 0.9% saline), 1 on d 0 at TAI and a second injection on d 14 (CTRL; = 53); 2) an injection of 325 mg of bST on d 0 and a saline injection on d 14 (bST0; = 48); 3) a saline injection on d 0 and an injection of 325 mg of bST on d 14 (bST14; = 49); or 4) 2 injections of 325 mg of bST, 1 on d 0 and a second injection on d 14 (bST0+14; = 40). Pregnancy status, crown-to-rump length (CRL) on Day 35, and crown-to-nose length (CNL) on Day 65 were determined via transrectal ultrasonography. Blood samples were collected on d 0, 7, 14, 21, 35, and 65, relative to TAI, to determine plasma concentrations of progesterone (P4), IGF-1, and pregnancy-specific protein B (PSPB) and also on d 18 and 21 for isolation of peripheral blood leukocytes for RNA extraction and measurement of interferon-stimulated genes transcript abundance. Individual calf BW was determined at birth and every 30 d until weaning. A subset of 24 calves was randomly selected for liver biopsies at birth to determine mRNA expression of target genes. Administration of bST to cows increased ( < 0.0001) concentrations of plasma IGF-1 for 14 d after injection compared with CTRL but did not affect fetal CRL and CNL ( = 0.23). Cows receiving bST only on d 0 had a greater ( = 0.05) transcript abundance in myxovirus resistance 2 on d 21 compared with 2bST cows (2.0- and 0.8-fold for bST0 and 2bST, respectively), whereas cows receiving bST14 and CTRL were intermediate (1.2- and 0.9-fold, respectively). Calf BW did not differ ( ≥ 0.100) among treatments on d 0, 30, 60, 90, 120, and 150 relative to birth. Injection of bST only on d 0 tended ( = 0.062) to increase calf liver mRNA expression of at birth compared with the calves born to cows in other treatments. Therefore, during a TAI protocol, the administration of 1 or 2 injections of 325 mg of bST to lactating beef cows enhanced their plasma concentrations of IGF-1 but failed to improve fetal size and plasma concentrations of maternal PSPB and P4 and had no effect on postnatal calf growth performance.

Effects of bovine somatotropin injection on serum concentrations of progesterone in non-lactating dairy cows

The objective of this experiment was to evaluate the effects of bovine somatotropin administration on serum concentrations of glucose, insulin, NEFA, IGF-I, and progesterone (P4) in ovariectomized non-lactating dairy cows receiving exogenous P4, as a model to estimate treatment effects on hepatic P4 degradation. Ten non-lactating, non-pregnant, and ovariectomized Gir×Holstein cows were assigned to the experiment (d −14 to 27). On d 0, cows were ranked by BW and BCS, and randomly assigned to one of two treatments: (1) bovine somatotropin (BST; n=5) or (2) saline control (control; n=5). Cows assigned to the BST treatment were administered s.c. injections containing 500mg of sometribove zinc on d 0, 9, and 18 of the experiment, whereas control cows concurrently received a 10-mL s.c. injection of 0.9% saline. On d −2, cows were inserted with an intravaginal releasing device containing 1.9g of P4, which remained in the cows until the end the experiment (d 27). Cow BW and BCS were assessed on d −14, 0, and 27. Blood samples were collected daily from d 0 to d 27, at 0 (immediately before), 1, and 2h relative to concentrate feeding for determination of serum glucose, insulin, NEFA, P4, and IGF-I concentrations. Concentrations of glucose, NEFA, and insulin obtained prior to feeding (0h) were used to determine pre-prandial revised quantitative insulin sensitivity check index (RQUICKI). No treatment effects were detected for BW (P=0.72) and BCS change (P=0.79) during the experiment. Beginning on d 2 of the experiment, BST cows had greater (P≤0.01) serum IGF-I concentrations compared with control cohorts (treatment×day interaction; P<0.01). Cows receiving BST had greater (P≤0.05) insulin concentrations compared with control cohorts from d 8 to d 11, d 16 and 17, as well as from d 19 to d 21 of the experiment (treatment×day interaction; P<0.01). Cows receiving BST had greater (P≤0.01) mean glucose and NEFA concentrations, as well as reduced (P<0.01) mean RQUICKI during the experiment compared with control cohorts. No treatment effects, however, were detected (P=0.73) for serum P4 concentrations. In conclusion, results from this experiment indicate that hepatic P4 catabolism is not directly regulated by circulating IGF-I, whereas BST administration decreases insulin sensitivity in non-lactating dairy cows in adequate nutritional status.

Effects of bovine somatotropin administration on growth, physiological, and reproductive responses of replacement beef heifers1

This experiment compared growth, body composition, plasma IGF-I and leptin, and reproductive development of beef heifers receiving or not recombinant bovine ST (BST) beginning after weaning until the first breeding season. Fifty Angus × Hereford heifers (initial BW = 219 ± 2 kg; initial age = 208 ± 2 d), weaned at approximately 6 mo of age, were assigned to the experiment (d 0 to 210). On d 0, heifers were ranked by initial BW and age and assigned to 1) treatment with BST or 2) saline control. Heifers assigned to the BST treatment received subcutaneous (s.c.) injections containing 250 mg of sometribove zinc whereas control heifers received a 5-mL s.c. injection of 0.9% saline every 14 d. Treatments were initiated on d 14 and last administered on d 196. Heifers were maintained on separate pastures harvested for hay the previous summer according to treatment and received grass and alfalfa hay at a rate to provide a daily amount of 7.0 and 1.0 kg of DM per heifer, respectively. Heifer shrunk BW was collected on d 1 and 211 for heifer ADG calculation. Blood samples were collected weekly from d 0 to 210 for determination of plasma progesterone to estimate puberty attainment as well as plasma concentrations of IGF-I and leptin in selected samples. On d 0, 63, 133, and 189, heifers were evaluated for intramuscular marbling, LM depth, and backfat thickness via real-time ultrasonography. No treatment effects were detected (P = 0.27) for heifer ADG (0.49 vs. 0.51 kg/d for control and BST heifers, respectively; SEM = 0.02). Mean backfat thickness was lesser (P < 0.01) in BST heifers compared with control cohorts (3.56 vs. 3.92 mm, respectively; SEM = 0.08). Heifers receiving BST had greater plasma IGF-I concentrations compared with control cohorts 7 d after treatment administration (treatment × day interaction; P < 0.01). Mean plasma leptin concentrations were lesser (P = 0.05) in BST heifers compared with control cohorts (1.82 vs. 2.03 ng/mL, respectively; SEM = 0.07). Onset of puberty was hastened in BST heifers compared with control cohorts (treatment × day interaction; P = 0.04). In summary, a greater proportion of BST heifers reached puberty during the experiment compared with control cohorts, despite lesser plasma leptin concentrations, backfat thickness, and similar ADG. Hence, circulating IGF-I was positively associated with hastened puberty attainment independently of growth rate, circulating leptin concentrations, and body fat content of replacement beef heifers.

Effect of bovine somatotropin administration during induction of lactation in 15-month-old heifers on production and health

Lactation can be induced successfully in 15-mo-old dairy heifers. Treatment of heifers induced into lactation with bovine somatotropin (bST) during an established lactation improved milk production; however, milk yields were still variable. The objective of the present study was to evaluate whether starting bST treatment during induction of lactation, rather than after lactation was established, would improve milk production beyond that of heifers induced into lactation but not treated with bST. Healthy Holstein heifers (n=32, 15 mo of age, 420±28kg of body weight) were induced into lactation with subcutaneous injections of estradiol (0.075mg/kg of body weight per d) and progesterone (0.25mg/kg of body weight per d) for 7 d. Bovine somatotropin (500mg) was administered to heifers (n=16) beginning on experimental d 1 along with the estrogen/progesterone treatment. Heifers continued to receive bST every 2 wk for 10 wk. Control animals (n=16) received no bST during this time. Milking began on experimental d 18, and milk production was compared through 53 d in milk (experimental d 70). Mean daily milk yield was 36% higher for bST-treated heifers than for control animals. A 15.5% difference in milk production between the groups was sustained through 305 d of lactation, even after control animals began bST treatment at 54 d in milk. Milk fat percentage was similar in bST and control heifers. Milk protein percentage was lower in bST-treated heifers (3.58%) compared with controls (3.99%) during the treatment comparison period and for the remainder of lactation (bST 3.25%, control 3.39%). Heifers treated with bST produced more total milk fat and protein compared with controls during the treatment comparison period. Throughout the induced lactation, heifers gained 0.87kg/d and averaged 2.4 services/pregnancy; 30 became pregnant. Four heifers were culled during the induced lactation, and 28 heifers calved at 27.6±2.0 mo of age for a second lactation. Addition of bST to the lactation induction protocol was advantageous because it stimulated greater milk production.

Short communication: Alterations in expression of gluconeogenic genes during heat stress and exogenous bovine somatotropin administration

Study objectives were to evaluate hepatic gluconeogenic enzyme gene expression in recombinant bovine somatotropin (rbST)-treated lactating dairy cattle during heat stress (HS) or in thermal-neutral, pair-fed (PF) animals. Twenty-two multiparous (99 d in milk, 656kg of BW) Holstein cows were subjected to 3 consecutive experimental periods (7 d each): (1) thermal neutral, (2) HS or PF, and (3) HS or PF with rbST (Posilac, administered on d 1 of period 3). Liver biopsies were obtained on the final day of each period. Heat stress conditions progressively decreased dry matter intake for the first 5 to 6 d during period 2 before stabilizing (a decrease of 6.15kg; 30%) on d 6 and 7, and feed intake remained stable and not different from period 2 during period 3. Cytosolic phosphoenolpyruvate carboxykinase mRNA abundance increased during PF, but was unaffected by HS or bST. Pyruvate carboxylase gene expression increased during HS and PF, and administrating bST decreased pyruvate carboxylase mRNA abundance during both HS and PF. Insulin-like growth factor-I gene expression increased following bST administration during HS and PF, confirming hepatic bST responsiveness. Exposure to HS leads to a change in hepatic gluconeogenic enzyme profile that appears to be dependent on plane of nutrition.

Profitability of bovine somatotropin administration to increase first insemination conception rate in seasonal dairy herds with heat stress

A dairy farm simulation program was used to estimate the economic value of increased conception rate at first insemination (FCR) as a result of one administration of bovine somatotropin (BST) per lactation. Further, the economically optimal BST administration period in a year was investigated. Typical conditions in Florida were assumed, including reduced milk production and conception rates in the summer and early fall due to heat stress. Administration of a single dose of BST ($6 per dose) per lactation was incorporated into a timed AI protocol for first insemination at 61 days after calving. Increases in FCR of 0, 4, 8, and 12 percentage points (PP) were compared including changes in prices, fertility, and seasonality of cow performance. No direct effect of BST on milk production was assumed. With default inputs and an increase in FCR of 4 PP, the optimal administration period was February through June which resulted in a small gain in profitability of $0.80/slot/year. Typically, one administration of BST per lactation to increase FCR was profitable in the cooler winter and spring, but not the hotter season. The use of BST in the cooler winter and spring resulted in increased seasonality of herd performance and demographics. When no seasonality in milk production was assumed, administration of BST was profitable throughout the year when FCR increased at least 8 PP. In conclusion, increases in FCR were most profitable during the cooler season of the year when basic conception rates were already greater than in the hotter season.

Microarray data analysis of gene expression levels in lactating cows treated with bovine somatotropin

Administration of bovine somatotropin (bST) to lactating cows results in an increase in milk production from 10 to 15%. While physiological mechanisms involved in bST administration are well known, there is limited knowledge about the mechanisms that regulate the bST action at genetic level. For this reason, a microarray experiment was conducted to identify differentially expressed genes when bST is given to milking cows. Sixteen high-density microarrays for cattle, each containing 18,263 gene spots, were used. RNA was extracted from the mammary tissue of four lactating Holstein cows, five and two days before, and one and six days after bST administration. A total of 1,251 and 1,167 differentially expressed genes were detected for mean and median expression intxsensities, respectively. Only the 115 genes which were identified by both mean and median intensities were taken into account. These genes were grouped into 8 clusters according to changes in expression through time points.

Administration of Bovine Somatotropin in Early Lactation: A Meta-Analysis of Production Responses by Multiparous Holstein Cows

A meta-analysis was conducted to assess production responses before 90 d in milk (DIM) when bovine somatotropin (bST) administration was initiated between 5 and 35 DIM. The database was developed from 13 studies of multiparous cows that were published between 1985 and 2006 and from an unpublished study that complied with the study selection criteria. The database included results from 842 cows and provided 50 treatment means for the effect of bST on 3.5% fat-corrected milk (FCM) in early lactation. Effects of bST were investigated using mixed model procedures that included fixed (intercept and slope) and random (intercept and slope) effects for independent variables. Yields of milk (38.6±1.3kg/d) and FCM (37.6±1.6kg/d) by control cows before 90 DIM were increased by 2.6±0.8 and 3.2±0.6kg/d by bST administration. Fat content in milk from bST-treated cows was 0.31±0.10 percentage units greater than that from control cows (3.46±0.13%) but milk protein content (2.95±0.03%) was not altered by bST. Milk fat (1.39±0.10kg/d) and protein (1.15±0.04kg/d) yields by controls were increased 0.16±0.03 and 0.07±0.03kg/d by bST, respectively. Dry matter intake and body weight loss were not altered by bST before 90 DIM, but duration of negative energy balance was prolonged and overall energy balance during this interval reduced when cows were treated with bST. Results are consistent with the premise that bST-treated cows partition nutrients and energy toward milk synthesis for a longer duration and thus likely need a longer interval to replenish their body reserves than cows not treated with bST. Production responses to bST were not altered when cows consumed typical early-lactation diets supplemented with fat except that supplemental fat tended to decrease the magnitude of the effect of bST on milk fat content and decreased the effect of bST on fat and protein yield. Yield of FCM increased curvilinearly with the amount of bST administered. Results indicate that initiation of bST administration to cows before 35 DIM increased FCM yield but the response was at the low end of that typically observed when bST administration is initiated in wk 9 of lactation.

The effects of pre-delivery administration of bovine somatotropin (bST) on first lactation performance were evaluated in this study, which involved eighteen Holstein pregnant heifers. Nine animals were supplemented with subcutaneous injections of 500 mg of bST, on days 21 and 9 prior expected delivery, and 9 were not supplemented (control group). Animals were fed total mixed ration, twice daily. Data evaluated in this experiment were related to sanitary, reproductive and productive parameters during first lactation. Administration of bST did not affect parameters related to sanity (retained placenta, metritis, displacement abomasum, milk fever, ketosis), reproduction (follicular cysts). However, heifers supplemented with bST showed changes of body condition score 65% greater, anticipated milk production peak in 9.7 days and decreased number of days in lactation in 21,8 days, compared to the non-supplemented ones (p < 0.05). Milk production peak and milk production corrected for 305 days were not affected by bST administration.

Use of computer simulation to teach a systems approach to metabolism

Use of a systems approach, as embodied in the computer simulation model of metabolism of a dairy cow, Molly (Baldwin, 2005), is ideal for teaching nutrition. This approach allows the overall complexity of the comprehensive system to be broken down into smaller manageable subunits that are easier to visualize. Quantitative interactions among nutrients supplied and metabolic production processes can be observed over extended time periods. Using Molly, undergraduate animal science students are able to observe detailed effects of changing dietary inputs, altering genetic milk production potential, and exogenously manipulating metabolism on metabolism of the whole cow. This paper demonstrates how Molly is used in the classroom to teach a systems approach to nutrition using example simulations. Three simulation examples demonstrate exercises examining effects of recombinant bovine somatotropin administration, dietary protein, and amino acid supplementation and nitrogen efficiency on milk production and cow metabolism. These and similar examples have been used to teach nutrition, metabolism, and lactation to undergraduate students for the past 20 yr.

Relationship of Thermal Status to Productivity in Heat-Stressed Dairy Cows Given Recombinant Bovine Somatotropin

The responses of lactating Holstein cows to daily administration of bovine somatotropin (bST) were measured at thermoneutrality (Tn) and under both constant and cycled heat-stress conditions to determine the relationship between thermal status and bST-induced shifts in milk production. All tests included a 5-d acclimation period at Tn (18°C), followed by a 2-d increase in ambient temperature to 28.5°C. After d 3, ambient temperature was cycled between 28.5 (day) and 25.5°C (night) for 4 d. Daily injections with either 31mg of bST or saline began on d 1 of the experiment. Milk production, feed intake, and respiratory rate (RR) were measured daily. Intraperitoneal, telemetric temperature transmitters were used for a continuous measure of core body temperature (Tcore). Blood samples were collected during each phase to evaluate the changes in serum chemistry in response to bST and heat stress. Following a 15-d recovery, cows were switched across injection treatments and the study was repeated. Milk production decreased by ∼18.4% below the initial yield at Tn by the end of 7 d of heat challenge. Although a reduction in milk production occurred during heat stress in both groups, milk production was higher in bST-treated cows compared with control cows during periods of constant and cyclic heat. Likewise, bST treatment during the entire period increased the milk-to-feed ratio over the control level by ∼11.3%. Plasma insulin-like growth factor 1 and serum nonesterified fatty acids accompanied the increased growth hormone level with bST treatment (∼122.0 and 88.8%, respectively), whereas plasma urea nitrogen was reduced by ∼13.3% to reflect the shift to lipid metabolism. There was no difference in Tcore of the treatment and control groups at Tn. Both bST and control cows increased RR and Tcore above the Tn level by ∼94.8 and 2.9%, respectively, during constant heat, with a greater increase in Tcore of bST-treated compared with control cows (∼0.6%). The increase in RR during heat stress preceded Tcore by 1 d for both groups. During cyclic heat, Tcore decreased by ∼0.4% compared with constant heat in both the control and bST-treated groups. Bovine somatotropin treatment increased milk production similarly during the Tn and heat-stress periods, ∼8.3% over the control; however, the bST-induced increase in milk-to-feed ratio was greatest during the continuous and cyclic heat-stress phases, ∼16.2%. This increase occurred together with the elevation in Tcore.

Risk factors associated with hemorrhagic bowel syndrome in dairy cattle

To determine risk factors associated with hemorrhagic bowel syndrome (HBS) among dairy cattle in the United States and identify characteristics of HBS in individual cows. Cross-sectional, population-based survey. A stratified random sample of 1,013 dairy operations with > or = 30 cows located in 21 states. Information on management and animal health-related topics was collected with a questionnaire. HBS was estimated to have been observed on 9.1% of operations during the preceding 5 years and on 5.1% of operations during the preceding 12 months. Factors found in multivariable analysis to be associated with the occurrence of HBS during the preceding 12 months were large herd size, administration of bovine somatotropin, and routine use of milk urea nitrogen concentration to determine ration composition. Use of pasture as part of the lactating cow ration during the growing season was associated with decreased odds of HBS in operations with rolling herd average milk production < or = 20,000 lb, whereas in operations with higher milk production, use of pasture was not associated with occurrence of HBS. For individual cows with signs consistent with HBS, the third lactation was the median of the parity distribution and the median time between parturition and the onset of clinical signs was 104 days. Results suggest that management practices implemented to achieve high milk production may increase the risk of developing HBS in dairy cattle. Increased consumption of a high-energy diet seems to be the most plausible common pathway for all of the risk factors that have been described.

Lactation persistency: insights from mammary cell proliferation studies.

A persistent lactation is dependent on maintaining the number and activity of milk secreting cells with advancing lactation. When dairy cows are milked twice daily, the increase in milk yield from parturition to peak lactation is due to increased secretory activity per cell rather than to accretion of additional epithelial cells. After peak lactation, declining milk yield is due to loss of mammary epithelial cells by apoptosis. During lactation, only 0.3% of mammary cells proliferate in a 24-h period. Yet this proliferative rate is sufficient to replace most mammary epithelial cells by the end of lactation. Management practices can influence lactation persistency. Administration of bovine somatotropin may enhance persistency by increasing cell proliferation and turnover, or by reducing the rate of apoptosis. Increased photoperiod may also increase persistency of lactation by mechanisms that are as yet undefined. Increased milking frequency during the first weeks of lactation increases milk yield, even after return to less frequent milking, with increases of approximately 8% over the entire lactation. A mammary cell proliferation response to frequent milking during early lactation appears to be involved. Conversely, advanced pregnancy, infrequent milking, and mastitis increase death of epithelial cells by apoptosis. Regulation of mammary cell renewal provides a key to increasing persistency. Investigations to characterize epithelial cells that serve as the proliferative population in the bovine mammary gland have been initiated. Epithelial cells that stain lightly in histological sections are evident through all phases of mammary development and secretion and account for nearly all proliferation in the prepubertal gland. Characterization of these cells may provide a means to regulate mammary cell proliferation and thus to enhance persistency, reduce the effects of mastitis, and decrease the necessity for a dry period.

Effects of Whole Cottonseed Diet and Recombinant Bovine Somatotropin on Ovarian Follicles in Lactating Dairy Cows

The effects of whole cottonseed (WCS) in the diet and the administration of bovine somatotropin (bST) on ovarian follicular dynamics and plasma progesterone (P4) concentrations were examined in cows during a period of synchronized follicular growth. Lactating Holstein cows (n = 28) were randomly assigned to treatments in a 2×2 factorial arrangement. Diets consisted of WCS (15% of dry matter) or no WCS, and bST at a dose of 0 or 208mg/14 d. Dietary treatments began within 24h of calving and bST treatments began within 7 d postpartum. Cows received GnRH at 65±3 d postpartum (d 0), PGF2α (d 7), a second dose of GnRH (d 9), and were inseminated 16h later (d 10). Ovarian changes were monitored daily by ultrasonography from d 0 to 9. On d 9, 93% of cows had a preovulatory follicle and 86% ovulated. For Class 2 (6 to 9mm) follicles, a diet×bST interaction was detected, with bST stimulating Class 2 follicles in cows fed WCS, but not in cows on the control diet. Neither diet nor bST affected numbers of Class 1 (2 to 5mm) or Class 3 (≥10mm) follicles or sizes of the subordinate and dominant follicles. During the luteal phase of the cycle, lactating cows fed WCS tended to have elevated concentrations of plasma P4, whereas bST was without effect. Plasma concentrations of high-density lipoprotein cholesterol were increased in cows fed WCS. Number and diameter of corpora lutea did not differ among treatments.

Dairy Herd Management Practices that Impact Nitrogen Utilization Efficiency

Improving the efficiency of feed N utilization by dairy cattle is the most effective means to reduce nutrient losses from dairy farms. The objectives of this study were to quantify the impact of different management strategies on the efficiency of feed N utilization for dairy farms in the Chesapeake Bay Drainage Basin. A confidential mail survey was completed in December 1998 by 454 dairy farmers in PA, MD, VA, WV, and DE. Nitrogen intake, urinary and fecal N, and efficiency of feed N utilization was estimated from survey data and milk analysis for each herd. Average efficiency of feed N utilization for milk production by lactating dairy cows (N in milk/N in feed × 100) was 28.4% (SD = 3.9). On average, farmers fed 6.6% more N than recommended by the National Research Council, resulting in a 16% increase in urinary N and a 2.7% increase in fecal N. Use of monthly milk yield and component testing, administration of bovine somatotropin (bST), and extending photoperiod with artificial light each increased efficiency of feed N utilization by 4.2 to 6.9%, while use of a complete feed decreased efficiency by 5.6%. Increased frequency of ration balancing and more frequent forage nutrient testing were associated with higher milk production, but not increased N utilization efficiency. Feeding protein closer to recommendations and increasing production per cow both contributed to improving efficiency of feed N utilization.

Bovine Somatotropin Administration to Dairy Goats in Late Lactation: Effects on Mammary Gland Function, Composition and Morphology

We investigated the effects of bovine somatotropin (bST) on mammary gland function and composition in the declining phase of lactation in goats. Sixteen Saanen goats, 180±11 days in milk (DIM), were divided equally into control and treated groups. The treated group received 120 mg/2 wk of slow-release bST for three cycles. Milk yield, milk composition, milk clotting measures, and plasmin-plasminogen activator activities were recorded weekly. Milk Na and K were determined in individual milk samples collected weekly during the third cycle. Blood samples were collected weekly during the second cycle and the plasma analyzed for nonesterified fatty acids (NEFA), glucose, and urea. At the end of the 6 wk, three goats from each group were slaughtered, and the udders were removed. Mammary gland weight, composition, and total DNA content were determined. The histological effects of bST on mammary tissue were investigated. The analyzed parameters included numbers of alveoli, corpora amylacea, apoptotic cells, and laminin fibronectin distribution and localization. An extensive morphological analysis on the epithelial and stromal components was performed. Milk yield was significantly higher in the treated group, fat content was not affected, but protein and nonprotein nitrogen were lower in treated goats milk. Treatment with bST did not influence milk pH but reduced coagulation time. Plasmin and plasminogen activator activities were not affected. Milk K levels were higher and the Na/K ratio was lower in treated animals. Plasma glucose, NEFA, and urea were unaffected. Mammary gland weight and total DNA were higher in treated than control animals, suggesting that with advancing lactation bST treatment maintains cells. Fat, protein, and collagen content of the mammary tissue did not differ between the groups. Treatment with bST significantly increased the number of lactating alveoli (LA) and significantly reduced the number of regressing alveoli (RA) and corpora amylacea, both within and outside the alveolar lumen. Laminin and fibronectin localization were not affected, and very few apoptotic cells were found in both treated and control samples. Our findings suggest that bST administration to dairy goats in late lactation can modulate mammary gland activity and improve lactation persistency; this is associated with maintained total mammary parenchyma weight and lactating alveoli.

The effect of interval from day of administration of bovine somatotropin (bST) to synchronization of ovulation and timed-insemination on conception rate of dairy cows with and without ovarian cysts

The objective of this study is to evaluate the effect of the interval from the day of administration of bovine somatotropin (bST) to the day of initiation of synchronization of ovulation (Day 0 and 7) and timed-insemination (TAI) on conception rate (CR) of dairy cows with and without ovarian cysts, respectively. Lactating dairy cows ( n=359) were divided into two groups. Cows in Group 1 ( n=238, without ovarian cysts) were treated with 100 ug, i.m. GnRH on Day 0; 25 mg, i.m. PGF2α on Day 8, 100 ug, i.m. GnRH on Day 10; and inseminated 16 h later without detection of estrus. Cows in Group 2 ( n=121, with ovarian cysts) were treated with 100 ug, i.m. GnRH on Day 0; 100 ug, i.m. GnRH on Day 7; 25 mg, i.m. PGF2α on Day 14, 100 ug, i.m. GnRH on Day 16; and inseminated 16 h later without detection of estrus. Between 60 and 63 days postpartum, all cows in the herd were given bST every 14 days for the duration of the study. Logistic regression was used to assess the risk of nonpregnancy associated with interval from bST treatment to Day 0 for cows without ovarian cysts, and both Day 0 and 7 for cows with ovarian cysts adjusting for parity and days in milk. The CR for cows in Group 1 was significantly higher when the interval from last treatment with bST to Day 0 was between 1 and 3 days (28%) compared to 4–6 days (14%). In addition, the risk of nonpregnancy was 2.19 times greater in cows 4–6 days after bST treatment compared to 1–3 days after adjusting for parity and days in milk. The CR for cows in Group 2 was not significantly different when the interval from last treatment with bST to both Day 0 and 7 was between 1 and 3, 4 and 6 , and 7 and 14 days. In conclusion, the results of this study suggested bST treatment closer to Day 0 had a positive effect on CR of cows without ovarian cysts, but bST treatment closer to both Day 0 and 7 had no effect on CR of cows with ovarian cysts. This was interpreted to mean that bST had a beneficial effect on either, or both, the preovulatory follicle and the oocyte in dairy cows without ovarian cysts, but not in dairy cows with ovarian cysts.

Induction of ovarian cystic follicles in sheep

Cystic follicles are a significant cause of infertility in women, dairy cattle and sheep. Sheep were used as a model to identify factors that may elicit formation of cystic follicles. Insulin resistance and elevated LH activity were tested in overweight ewes because of associations among these factors and the formation of cystic follicles. Sheep were synchronized using a progesterone-releasing pessary and insulin resistance was induced during the synchronization period through administration of bovine somatotropin. Following removal of pessaries follicular growth was stimulated by treatment with eCG or eCG and hCG (PG-600). Follicular growth was monitored via daily transrectal ultrasonography and blood samples were collected for hormonal analyses. Six of 18 ewes had a subnormal or absent preovulatory gonadotropin surge and developed cystic follicles. Neither insulin resistance nor elevated LH activity were associated with formation of cystic follicles. Ewes that developed cystic follicles were heavier (93 ± 4 kg) than ewes that ovulated (81 ± 3 kg; P = 0.02). Furthermore, following pessary removal and initiation of daily ultrasonography, ewes that developed cystic follicles lost body weight (−3 ± 1%), while ovulatory ewes continued to gain body weight (1 ± 1%; P = 0.005). It is speculated that in heavy ewes metabolic factors associated with acute body weight loss inhibit the positive feedback of estradiol and thereby suppress the preovulatory gonadotropin surge leading to formation of cystic follicles.

Effects of recombinant bovine somatotropin administration to lactating goats

Eight multiparus lactating goats, matched for parity and days in milk were used to evaluate the effects of recombinant bovine somatotropin (rbST) administration on milk yield and composition, as well as on certain metabolic parameters. The experiment was a switch-back design, with three periods lasting 28 days each. Animals were divided into two groups ( n = 4) and treatments consisted of subcutaneous injection of 160 mg rbST at 14-day intervals. Controls remained uninjected. Supplementation with rbST increased milk yield significantly over the entire experimental period (1506 ± 80.8 vs. 1337 ± 81.8 ml/day, p < 0.05). Percentage of fat in milk was greater for goats treated with rbST than controls, (3.26 ± 0.09% vs. 2.9 ± 0.08%, p < 0.05), while protein percentage did not differ ( p > 0.05) between treated and control animals. Lactose content also increased after rbST administration (3.88 ± 0.06% vs. 3.55 ± 0.06%, p < 0.01). Plasma NEFA and β-hydroxybutyrate levels were not affected by treatment. Short-chain fatty acids in milk were similar in both groups. In conclusion, rbST administration increased milk yield and the percentage of fat and lactose in milk, while no effect was recorded in milk protein content.
