Persistent activity of aerobic methane-oxidizing bacteria in anoxic lake waters due to metabolic versatility

Lacustrine methane emissions are strongly mitigated by aerobic methane-oxidizing bacteria (MOB) that are typically most active at the oxic-anoxic interface. Although oxygen is required by the MOB for the first step of methane oxidation, their occurrence in anoxic lake waters has raised the possibility that they are capable of oxidizing methane further anaerobically. Here, we investigate the activity and growth of MOB in Lake Zug, a permanently stratified freshwater lake. The rates of anaerobic methane oxidation in the anoxic hypolimnion reached up to 0.2 µM d−1. Single-cell nanoSIMS measurements, together with metagenomic and metatranscriptomic analyses, linked the measured rates to MOB of the order Methylococcales. Interestingly, their methane assimilation activity was similar under hypoxic and anoxic conditions. Our data suggest that these MOB use fermentation-based methanotrophy as well as denitrification under anoxic conditions, thus offering an explanation for their widespread presence in anoxic habitats such as stratified water columns. Thus, the methane sink capacity of anoxic basins may have been underestimated by not accounting for the anaerobic MOB activity.

Soil warming-induced reduction in water content enhanced methane uptake at different soil depths in a subtropical forest

Global warming can significantly impact soil CH4 uptake in subtropical forests due to changes in soil moisture, temperature sensitivity of methane-oxidizing bacteria (MOB), and shifts in microbial communities. However, the specific effects of climate warming and the underlying mechanisms on soil CH4 uptake at different soil depths remain poorly understood. To address this knowledge gap, we conducted a soil warming experiment (+4 °C) in a natural forest. From August 2020 to October 2021, we measured soil temperature, soil moisture, and CH4 uptake rates at four different soil depths: 0–10 cm, 10–20 cm, 20–40 cm, and 40–60 cm. Additionally, we assessed the soil MOB community structure and pmoA gene (with qPCR) at the 0–10 and 10–20 cm depths. Our findings revealed that warming significantly enhanced soil net CH4 uptake rate by 12.28 %, 29.51 %, and 61.05 % in the 0–10, 20–40, and 40–60 cm soil layers, respectively. The warming also led to reduced soil moisture levels, with more pronounced reductions observed at the 20–40 cm depth compared to the 0–20 cm depth. At the 0–10 cm depth, warming increased the relative abundance of upland soil cluster α (a type of MOB) and decreased the relative abundance of Methylocystis, but it did not significantly increase the pmoA gene copies. Our structural equation model analysis indicated that warming directly regulated soil CH4 uptake rate through the decrease in soil moisture, rather than through changes in the pmoA gene and MOB community structure at the 0–20 cm depth. In summary, our results demonstrate that warming enhances soil CH4 uptake at different depths, with soil moisture playing a crucial role in this process. Under warming conditions, the drier soil pores allow for better CH4 penetration, thereby promoting more efficient activity of MOB.

Enhanced methane oxidation efficiency by digestate biochar in landfill cover soil: Microbial shifts and carbon metabolites insights

The ability of biochar to enhance the oxidation of methane (CH4) in landfill cover soil by promoting the growth and activity of methane-oxidizing bacteria (MOB) has attracted significant attention. However, the optimal characteristics of digestate-derived biochar (DBC) for promoting the MOB community and CH4 removal performance remain unclear. This study examined how the CH4 oxidation capacity and respiratory metabolism of MOB life process are affected by the application of DBC compared with the most commonly used woody-derived biochar (WBC). The addition of both WBC and DBC enhanced CH4 oxidation, with DBC exhibiting a nearly twofold increase in cumulative CH4 oxidation mass (7.14 mg CH4 g−1) compared to WBC. The high ion-exchange capacity of DBC was found to be more favorable for the growth of Type I MOB, which have more efficient metabolic pathways for CH4 oxidation. Type I MOB which are abundant in DBC may prefer monovalent positive ions, while the charge-rich nature of DBC may also have hindered extracellular protein aggregation. The superiority of DBC in terms of CH4 oxidation thus highlights the underlying mechanisms of biochar-MOB interactions, offering potential biochar options for landfill cover soil.

The Influence of Abiotic Factors on the Activity of Methane-Oxidizing Bacteria

Methanotrophs are widespread bacteria and utilize a number of hydrocarbons under normal conditions. In this study, a model reaction of propylene oxidation by methane-oxidizing bacteria Methylococcus capsulatus (M) was considered. The influence of abiotic factors (the flow rate, exogenous metals) on methane-oxidizing bacteria has been studied. It was found that increasing the flow velocity slightly effect on the growth of bacterial biomass. Studies have shown that under the action of salts of the studied metals a significant decrease the propylene oxidation by bacteria is observed. Methanotrophs exhibit resistance to stress factors, which ensures their wide distribution in nature and provides biotechnological potential.

Activity and abundance of methane-oxidizing bacteria on plants in experimental lakes subjected to different nutrient and warming treatments

Shallow lakes produce and emit substantial amounts of methane (CH4). Part of the CH4 produced in lakes is consumed by methane-oxidizing bacteria (MOB) present in the sediment and water column, thus reducing the overall CH4 emissions. However, the role of aquatic plants as habitat for CH4 oxidation by MOB is poorly understood. In this study, we compared CH4 oxidation rates and MOB abundance associated with different types of aquatic plants (periphyton, filamentous algae, and both above-ground macrophytes and their rhizosphere). The plants were collected from shallow lake mesocosms exposed to experimental nutrient enrichment and warming treatments for 17 years prior to this study. Incubations of all sampled plants showed CH4 oxidation, with above-ground macrophyte tissue and filamentous algae having the highest rates of up to 0.25 µmol CH4 h−1 g−1 dw. Oxidation rates associated with macrophytes were species dependent, with consumption rates on rhizospheres of Potamogeton crispus higher than those on Elodea canadensis. The increase in nutrients and dissolved CH4 in the water tended to increase MOB abundance and activity for all plant types, while no effect of long term warming was detectable. Our results showed that MOB associated with periphyton, filamentous algae and macrophytes oxidize CH4 in shallow lakes at different rates across species or plant types. We also found that high macrophyte biomass is associated with reduced CH4 concentration in the water. This study shows that CH4 oxidation occurs on many plant surfaces but that oxidation rates alone cannot explain the reduced CH4 emissions at higher plant biomass.

Ecological Aerobic Ammonia and Methane Oxidation Involved Key Metal Compounds, Fe and Cu.

Interactions between metals and microbes are critical in geomicrobiology and vital in microbial ecophysiological processes. Methane-oxidizing bacteria (MOB) and ammonia-oxidizing microorganisms (AOM) are key members in aerobic environments to start the C and N cycles. Ammonia and methane are firstly oxidized by copper-binding metalloproteins, monooxygenases, and diverse iron and copper-containing enzymes that contribute to electron transportation in the energy gain pathway, which is evolutionally connected between MOB and AOM. In this review, we summarized recently updated insight into the diverse physiological pathway of aerobic ammonia and methane oxidation of different MOB and AOM groups and compared the metabolic diversity mediated by different metalloenzymes. The elevation of iron and copper concentrations in ecosystems would be critical in the activity and growth of MOB and AOM, the outcome of which can eventually influence the global C and N cycles. Therefore, we also described the impact of various concentrations of metal compounds on the physiology of MOB and AOM. This review study could give a fundamental strategy to control MOB and AOM in diverse ecosystems because they are significantly related to climate change, eutrophication, and the remediation of contaminated sites for detoxifying pollutants.

The role of methanotrophy in the microbial carbon metabolism of temperate lakes

Previous stable isotope and biomarker evidence has indicated that methanotrophy is an important pathway in the microbial loop of freshwater ecosystems, despite the low cell abundance of methane-oxidizing bacteria (MOB) and the low methane concentrations relative to the more abundant dissolved organic carbon (DOC). However, quantitative estimations of the relative contribution of methanotrophy to the microbial carbon metabolism of lakes are scarce, and the mechanism allowing methanotrophy to be of comparable importance to DOC-consuming heterotrophy remained elusive. Using incubation experiments, microscopy, and multiple water column profiles in six temperate lakes, we show that MOB play a much larger role than their abundances alone suggest because of their larger cell size and higher specific activity. MOB activity is tightly constrained by the local methane:oxygen ratio, with DOC-rich lakes with large hypolimnetic volume fraction showing a higher carbon consumption through methanotrophy than heterotrophy at the whole water column level. Our findings suggest that methanotrophy could be a critical microbial carbon consumption pathway in many temperate lakes, challenging the prevailing view of a DOC-centric microbial metabolism in these ecosystems.

Methane Emission Reduction Enhanced by Hydrophobic Biochar-Modified Soil Cover

The microbial oxidation of CH4 in biochar-modified soil cover is considered a potent option for the mitigation of emissions from old landfills or sites containing wastes full of low CH4 generation rates. The mechanism of methane oxidizing bacteria (MOB) can be enhanced by amending the landfill cover soil with biochar, which is recalcitrant to biological degradation and can adsorb CH4 while facilitating the growth and activity of MOB within its porous structure. However, the increase in the permeability coefficient and water content of the cover due to the addition of biochar also affects the methane removal efficiency. A hydrophobic biochar modified by KH-570 was employed to reduce the water content and to promote the diffusion and oxidation of CH4 in the cover. Several series of small-scale column tests were conducted to quantify the CH4 oxidation properties of the landfill cover soil amended with biochar and hydrophobic biochar under different levels of exposed CH4 concentrations (5% and 15%), heights (10–66 cm), and temperatures (15–40 °C). After 30 days of domestication, the removal rate of the hydrophobic biochar-modified soil cover reached 98.8%. The water holding capacity of the cover and the CH4 oxidation efficiency under different moisture contents were investigated in different columns. The hydrophobic biochar-modified soil cover has a weak water holding capacity, low saturated water content, and optimal CH4 oxidation efficiency at this time.

Interactions of sulfur and methane-oxidizing bacteria in tropical estuarine sediments

The bacterial oxidation of sulfur and methane is central to the biogeochemical processes in sediments such as the tropical mangrove sediments. However, there is a lacuna of information on the seasonal interactions including the influence of monsoons which is a major driver of seasonal change, on sulfur-oxidizing bacteria (SOB) and methane-oxidizing bacteria (MOB), their activity and the environmental variables. To understand these interactions, the analysis was carried out on sediment samples that were sampled monthly for a year from Chorao mangrove, Goa, southwest coast of India. SOB (3.8×105CFU g-1) and MOB (0.90×105CFU g-1) had maximum average abundance in the surface sediments in the post-monsoon and monsoon season, respectively. The mean sulfur-oxidation activity (SOA) of 2.63mMday-1 and methane-oxidation activity (MOA) of 110.94mMday-1 were highest in surface sediments during the post-monsoon season. Generally, the activity of SOB and MOB in surface sediments of post-monsoon was 2.2 times(×) and 2.8× respectively higher than that in the monsoon season. Among the environmental parameters analyzed, protein and sulfide concentrations significantly (p < 0.001) influenced SOA and MOA, respectively. There was a significant difference in SOA (p < 0.003) and MOA (p < 0.036) in surface sediments between the monsoon and the post-monsoon season. During the monsoon season, when the system is a sink of terrestrial/anthropogenic material, the interrelationship of SOB with MOA (r = 0.617, p < 0.001) and SOB with SOA (r = 0.489, p < 0.05) aids in maintaining the homeostasis of the system.

High Temporal and Spatial Variability of Atmospheric-Methane Oxidation in Alpine Glacier Forefield Soils.

Glacier forefield soils can provide a substantial sink for atmospheric CH4, facilitated by aerobic methane-oxidizing bacteria (MOB). However, MOB activity, abundance, and community structure may be affected by soil age, MOB location in different forefield landforms, and temporal fluctuations in soil physical parameters. We assessed the spatial and temporal variability of atmospheric-CH4 oxidation in an Alpine glacier forefield during the snow-free season of 2013. We quantified CH4 flux in soils of increasing age and in different landforms (sandhill, terrace, and floodplain forms) by using soil gas profile and static flux chamber methods. To determine MOB abundance and community structure, we employed pmoA gene-based quantitative PCR and targeted amplicon sequencing. Uptake of CH4 increased in magnitude and decreased in variability with increasing soil age. Sandhill soils exhibited CH4 uptake rates ranging from -3.7 to -0.03 mg CH4 m-2 day-1 Floodplain and terrace soils exhibited lower uptake rates and even intermittent CH4 emissions. Linear mixed-effects models indicated that soil age and landform were the dominating factors shaping CH4 flux, followed by cumulative rainfall (weighted sum ≤4 days prior to sampling). Of 31 MOB operational taxonomic units retrieved, ∼30% were potentially novel, and ∼50% were affiliated with upland soil clusters gamma and alpha. The MOB community structures in floodplain and terrace soils were nearly identical but differed significantly from the highly variable sandhill soil communities. We concluded that soil age and landform modulate the soil CH4 sink strength in glacier forefields and that recent rainfall affects its short-term variability. This should be taken into account when including this environment in future CH4 inventories.IMPORTANCE Oxidation of methane (CH4) in well-drained, "upland" soils is an important mechanism for the removal of this potent greenhouse gas from the atmosphere. It is largely mediated by aerobic, methane-oxidizing bacteria (MOB). Whereas there is abundant information on atmospheric-CH4 oxidation in mature upland soils, little is known about this important function in young, developing soils, such as those found in glacier forefields, where new sediments are continuously exposed to the atmosphere as a result of glacial retreat. In this field-based study, we investigated the spatial and temporal variability of atmospheric-CH4 oxidation and associated MOB communities in Alpine glacier forefield soils, aiming at better understanding the factors that shape the sink for atmospheric CH4 in this young soil ecosystem. This study contributes to the knowledge on the dynamics of atmospheric-CH4 oxidation in developing upland soils and represents a further step toward the inclusion of Alpine glacier forefield soils in global CH4 inventories.

Does acidification of a soil biofilter compromise its methane-oxidising capacity?

A biofilter made using volcanic pumice soil from a landfill in Taupo, New Zealand has been found to mitigate CH4 emissions from New Zealand dairy effluent ponds. However, the biofilter after drying out following almost 5 years of use removed little or no CH4. Furthermore, H2S present in the biogas (from the dairy effluent ponds) had increased the acidity (pH) in the soil biofilter from 5.2 to 3.72 during this 5-year period. In this study, we adjusted the soil moisture to 60 % water-holding capacity (WHC) and investigated the CH4-oxidising capacity of a reconstituted acidic soil biofilter operating at low pH (3.72) and characterised the abundance and diversity of methane-oxidising bacteria (MOB) using quantitative polymerase chain reaction (qPCR) and terminal-restriction fragment length polymorphism (T-RFLP). The acidic soil biofilter achieved a maximum CH4 removal rate of 30.3 g m–3 h–1. Both types I and II MOB communities, along with some uncultured novel MOB strains or species in the biofilter column, were present. Among these, Methylocapsa-like type II methanotrophs were significantly more prominent than the other MOB. Other MOB, Methylococcus (type I), Methylobacter/Methylomonas/Methylosarcina (type I) genera, Methylosinus and Methylocystis (type II), were least abundant. During the 90-day study, the population of Methylocapsa-like MOB increased 4-fold, demonstrating the ability of these soil microorganisms to grow under acidic pH conditions in the biofilter, whereas the populations of type I MOB remained stable, and the populations of type II MOB (except Methylocapsa) decreased. Our results indicated that (i) a soil biofilter can effectively regain efficiency if sufficient moisture levels are maintained, regardless of the soil acidity; (ii) changes in the MOB population did not compromise the capacity of the volcanic pumice soil to oxidise CH4; (iii) the more acidic environment (pH 3.72) tends to favour the growth and activity of acid loving Methylocapsa-like MOB while being detrimental to the growth of Methylobacter/Methylocystis/Methylococcus group of MOB; and (iv) novel species or strains of uncultured Methylomicrobium/Methylosarcina/Methylobacter (type I MOB) could be present in the soil biofilter. This study has revealed the MOB population changes in the biofilter with acidification did not compromise its capacity to oxidise CH4 demonstrating that soil biofilter can operate effectively under acidic conditions.

Adsorption and transport of methane in landfill cover soil amended with waste-wood biochars

The natural presence of methane oxidizing bacteria (MOB) in landfill soils can stimulate the bio-chemical oxidation of CH4 to CO2 and H2O under suitable environmental conditions. This mechanism can be enhanced by amending the landfill cover soil with organic materials such as biochars that are recalcitrant to biological degradation and are capable of adsorbing CH4 while facilitating the growth and activity of MOB within their porous structure. Several series of batch and small-scale column tests were conducted to quantify the CH4 sorption and transport properties of landfill cover soil amended with four types of waste hardwood biochars under different levels of amendment percentages (2, 5 and 10% by weight), exposed CH4 concentrations (0–1 kPa), moisture content (dry, 25% and 75% water holding capacity), and temperature (25, 35 and 45 °C). The linear forms of the pseudo second-order kinetic model and the Langmuir isotherm model were used to determine the kinetics and the maximum CH4 adsorption capacity of cover materials. The maximum CH4 sorption capacity of dry biochar-amended soils ranged from 1.03 × 10−2 to 7.97 × 10−2 mol kg−1 and exhibited a ten-fold increase compared to that of soil with 1.9 × 10−3 mol kg−1. The isosteric heat of adsorption for soil was negative and ranged from −30 to −118 kJ/mol, while that of the biochar-amended soils was positive and ranged from 24 to 440 kJ/mol. The CH4 dispersion coefficients for biochar-amended soils obtained through predictive transport modeling indicated that amending the soil with biochar enhanced the methane transport rates by two orders of magnitude, thereby increasing their potential for enhanced exchange of gases within the cover system. Overall, the use of hardwood biochars as a cover soil amendment to reduce methane emissions from landfills appears to be a promising alternative to conventional soil covers.

Conversion of cropland to forest increases soil CH4 oxidation and abundance of CH4 oxidizing bacteria with stand age

We investigated CH4 oxidation in afforested soils over a 200-year chronosequence in Denmark including different tree species (Norway spruce, oak and larch) and ages. Samples of the top mineral soil (0–5cm and 5–15cm depth) were incubated and analyzed for the abundance of the soil methane-oxidizing bacteria (MOB) and ammonia-oxidizing bacteria (AOB) and archaea (AOA) based on quantitative PCR (qPCR) on pmoA and amoA genes. Our study showed that CH4 oxidation rates and the abundance of MOB increased simultaneously with time since afforestation, suggesting that the methanotrophic activity is reflected in the abundance of this functional group.The development of forest soils resulted in increased soil organic carbon and reduced bulk density, and these were the two variables that most strongly related to CH4 oxidation rates in the forest soils. For the top mineral soil layer (0–5cm) CH4 oxidation rates did not differ between even aged stands from oak and larch, and were significantly smaller under Norway spruce. Compared to the other tree species Norway spruce caused a decrease in the abundance of MOB over time that could explain the decreased oxidation rates. However, the cause for the lower abundance remains unclear. The abundance of ammonia-oxidizers along the chronosequence decreased over time, oppositely to the MOB. However, our study did not indicate a direct link between CH4 oxidation rates and ammonia-oxidizers. Here, we provide evidence for a positive impact of afforestation of former cropland on CH4 oxidation capacity in soils most likely caused by an increased population size and activity of MOB.

Remarkable Recovery and Colonization Behaviour of Methane Oxidizing Bacteria in Soil After Disturbance Is Controlled by Methane Source Only

Little is understood about the relationship between microbial assemblage history, the composition and function of specific functional guilds and the ecosystem functions they provide. To learn more about this relationship we used methane oxidizing bacteria (MOB) as model organisms and performed soil microcosm experiments comprised of identical soil substrates, hosting distinct overall microbial diversities(i.e., full, reduced and zero total microbial and MOB diversities). After inoculation with undisturbed soil, the recovery of MOB activity, MOB diversity and total bacterial diversity were followed over 3 months by methane oxidation potential measurements and analyses targeting pmoA and 16S rRNA genes. Measurement of methane oxidation potential demonstrated different recovery rates across the different treatments. Despite different starting microbial diversities, the recovery and succession of the MOB communities followed a similar pattern across the different treatment microcosms. In this study we found that edaphic parameters were the dominant factor shaping microbial communities over time and that the starting microbial community played only a minor role in shaping MOB microbial community.

Atmospheric methane oxidizers are present and active in Canadian high Arctic soils

The melting of permafrost and the associated potential for methane emissions to the atmosphere are major concerns in the context of global warming. However, soils can also represent a significant sink for methane through the activity of methane-oxidizing bacteria (MOB). In this study, we looked at the activity, diversity, and community structure of MOB at two sampling depths within the active layer in three soils from the Canadian high Arctic. These soils had the capacity to oxidize methane at low (15 ppm) and high (1000 ppm) methane concentrations, but rates differed greatly depending on the sampling date, depth, and site. The pmoA gene sequences related to two genotypes of uncultured MOB involved in atmospheric methane oxidation, the 'upland soil cluster gamma' and the 'upland soil cluster alpha', were detected in soils with near neutral and acidic pH, respectively. Other groups of MOB, including Type I methanotrophs and the 'Cluster 1' genotype, were also detected, indicating a broader diversity of MOB than previously reported for Arctic soils. Overall, the results reported here showed that methane oxidation at both low and high methane concentrations occurs in high Arctic soils and revealed that different groups of atmospheric MOB inhabit these soils.

Field-scale labelling and activity quantification of methane-oxidizing bacteria in a landfill-cover soil

Aerobic methane-oxidizing bacteria (MOB) play an important role in soils, mitigating emissions of the greenhouse gas methane (CH(4)) to the atmosphere. Here, we combined stable isotope probing on MOB-specific phospholipid fatty acids (PLFA-SIP) with field-based gas push-pull tests (GPPTs). This novel approach (SIP-GPPT) was tested in a landfill-cover soil at four locations with different MOB activity. Potential oxidation rates derived from regular- and SIP-GPPTs agreed well and ranged from 0.2 to 52.8 mmol CH(4) (L soil air)(-1) day(-1). PLFA profiles of soil extracts mainly contained C(14) to C(18) fatty acids (FAs), with a dominance of C(16) FAs. Uptake of (13) C into MOB biomass during SIP-GPPTs was clearly indicated by increased δ(13)C values (up to c. 1500‰) of MOB-characteristic FAs. In addition, (13)C incorporation increased with CH(4) oxidation rates. In general, FAs C(14:0) , C(16:1ω8), C(16:1ω7) and C(16:1ω6) (type I MOB) showed highest (13)C incorporation, while substantial (13)C incorporation into FAs C(18:1ω8) and C(18:1ω7) (type II MOB) was only observed at high-activity locations. Our findings demonstrate the applicability of the SIP-GPPT approach for in situ quantification of potential CH(4) oxidation rates and simultaneous labelling of active MOB, suggesting a dominance of type I MOB over type II MOB in the CH(4)-oxidizing community in this landfill-cover soil.

Structure and function of methanotrophic communities in a landfill-cover soil

In landfill-cover soils, aerobic methane-oxidizing bacteria (MOB) convert CH(4) to CO(2), mitigating emissions of the greenhouse gas CH(4) to the atmosphere. We investigated overall MOB community structure and assessed spatial differences in MOB diversity, abundance and activity in a Swiss landfill-cover soil. Molecular cloning, terminal restriction-fragment length polymorphism (T-RFLP) and quantitative PCR of pmoA genes were applied to soil collected from 16 locations at three different depths to study MOB community structure, diversity and abundance; MOB activity was measured in the field using gas push-pull tests. The MOB community was highly diverse but dominated by Type Ia MOB, with novel pmoA sequences present. Type II MOB were detected mainly in deeper soil with lower nutrient and higher CH(4) concentrations. Substantial differences in MOB community structure were observed between one high- and one low-activity location. MOB abundance was highly variable across the site [4.0 × 10(4) to 1.1 × 10(7) (g soil dry weight)(-1)]. Potential CH(4) oxidation rates were high [1.8-58.2 mmol CH(4) (L soil air)(-1) day(-1) ] but showed significant lateral variation and were positively correlated with mean CH(4) concentrations (P < 0.01), MOB abundance (P < 0.05) and MOB diversity (weak correlation, P < 0.17). Our findings indicate that Methylosarcina and closely related MOB are key players and that MOB abundance and community structure are driving factors in CH(4) oxidation at this landfill.

Seasonal increase of methane in sediment decreases δ13C of larval chironomids in a eutrophic shallow lake

Recent studies have shown that larval chironomids assimilate 13C-depleted carbon derived from biogenic methane by feeding on methane-oxidizing bacteria (MOB). The dietary contribution of MOB is known to be maximized in the autumn overturn period or winter in eutrophic dimictic lakes due to the increase of MOB biomass following the supply of oxygenated water, but in polymictic lakes, such seasonal variability has not been revealed. We investigated the seasonal patterns of larval δ13C and methane concentrations in the sediment of a eutrophic polymictic lake, Izunuma, Japan. Larval δ13C decreased in late summer and autumn. Methane concentrations above a 6 cm depth peaked in late summer or autumn, while those in the 10–11- and 20–21-cm layers peaked in October. Negative correlations between methane concentrations in the 5–6/10–11-cm layers and larval δ13C were found. This suggests that an increase in the supply of methane stimulated the activity of MOB in a polymictic lake, where water above the lake bottom rarely became anoxic because of frequent overturn, thus increasing the dietary contribution of MOB to larval chironomids.

The active methanotrophic community in a wetland from the High Arctic

The dominant terminal process of carbon mineralization in most freshwater wetlands is methanogenesis. With methane being an important greenhouse gas, the predicted warming of the Arctic may provide a positive feedback. However, the amount of methane released to the atmosphere may be controlled by the activity of methane-oxidizing bacteria (methanotrophs) living in the oxic surface layer of wetlands. Previously, methanotrophs have been isolated and identified by genetic profiling in High Arctic wetlands showing the presence of only a few genotypes. Two isolates from Solvatnet (Ny-Ålesund, Spitsbergen; 79°N) are available: Methylobacter tundripaludum (type I) and Methylocystis rosea (type II), raising the question whether the low diversity is a cultivation effect. We have revisited Solvatnet applying stable isotope probing (SIP) with (13) C-labelled methane. 16S rRNA profiling revealed active type I methanotrophs including M. tundripaludum, while no active type II methanotrophs were identified. These results indicate that the extant M. tundripaludum is an active methane oxidizer at its locus typicus; furthermore, Methylobacter seems to be the dominant active genus. Diversity of methanotrophs was low as compared, e.g. to wetland rice fields in the Mediterranean. This low diversity suggests a high vulnerability of Arctic methanotroph communities, which deserves more attention.

Because methane-oxidizing bacteria (MOB) are the only biological sink for the greenhouse gas methane, knowledge of the functioning of these bacteria in various ecosystems is needed to understand the dynamics observed in global methane emission. The activity of MOB is commonly assessed by methane oxidation assays. The resulting methane depletion curves often follow a biphasic pattern of initial and induced methane oxidation activity, often interpreted as representing the in situ active and total MOB community, respectively. The application of quantitative-PCR on soil incubations, which were stopped before, at and after the transition point in the methane-depletion curve, demonstrated that both pmoA-mRNA was produced as well as substantial cell growth took place already in the initial phase. In addition, type Ia and II MOB displayed markedly different behaviour, which can be interpreted as ecologically different strategies. For the correct interpretation of methane oxidation assays, the use of small time windows is recommended to calculate methane oxidation activities to avoid substantial cell growth.
