Dietary phosphorus requirement of bullfrog Aquaranacatesbeiana

This experiment investigated the dietary phosphorus (P) requirement of bullfrogs Aquaranacatesbeiana. Six diets containing available P (AP) levels of 0.29% (the control group), 0.47%, 0.65%, 0.88%, 1.06% and 1.24% were formulated and fed to bullfrogs for 56 days. As dietary AP level increased from 0.29% to 1.24%, weight gain rate increased and then decreased, reaching the highest at the AP level of 0.88% (P < 0.05). Compared to the control group, content of crude protein in the whole-body was significantly increased in bullfrogs fed the 0.88% AP diet (P < 0.05). Content of crude lipid in the whole-body was significantly decreased in bullfrogs fed the 0.65%, 0.88%, 1.06% and 1.24% AP diets compared to those fed the 0.29% and 0.47% AP diets (P < 0.05). Bullfrogs fed the 0.65%, 0.88%, 1.06%, and 1.24% AP diets had higher content of serum P than those fed the 0.29% and 0.47% AP diets (P < 0.05). Activity of glutamic pyruvic transaminase in the serum was significantly decreased in bullfrogs fed the 0.88%, 1.06% and 1.24% AP diets compared to those fed the 0.29% and 0.47% AP diets (P < 0.05). Bullfrogs fed the 0.65%, 0.88%, 1.06% and 1.24% AP diets had lower contents of total cholesterol in the liver and triglycerides in the serum than the control group (P < 0.05). Meanwhile, bullfrogs fed the 0.47%, 0.65%, 0.88% and 1.06% AP diets had higher total antioxidant capacity in the serum than the control group (P < 0.05). Compared to the control group, contents of P and calcium in the whole-body were significantly increased in bullfrogs fed the 0.65%, 0.88%, 1.06% and 1.24% AP diets (P < 0.05). Compared to the control group, bullfrogs fed the 0.88%, 1.06% and 1.24% AP diets had lower expression of sodium-dependent phosphate transporter in the intestine and kidney, and higher expression of hormone-sensitive triglyceride lipase in the liver (P < 0.05). Bullfrogs fed the 0.88% and 1.06% AP diets had higher the mucosal fold height than those fed the 0.29% and 1.24% AP diets (P < 0.05). Compared to those fed the 0.29% and 1.24% AP diets, the abundance of potentially pathogenic bacteria Serratia and Acinetobacter in the intestine was decreased in bullfrogs fed the 0.88% AP diet. Using the weight gain rate, feed efficiency, whole-body and serum P content as evaluation indicators, the AP requirements of bullfrogs was 0.82% to 0.89%.

Exploring the relationship between dietary rapeseed meal, condensed tannin and growth, nutrive metabolism in largemouth bass (Micropterus salmoides)

This study evaluated the effects of rapeseed meal (RM) and condensed tannin (CT) on the growth and nutritional metabolism of largemouth bass (Micropterus salmoides), and speculated whether there was interaction between anti-nutritional factors in RM. Eight diets were formulated for 55-day feeding trial, six of which were practical diet groups, including control group (CG), five RM inclusive groups with graded RM levels of 5% (RM5), 10% (RM10), 15% (RM15), 20% (RM20) and 25% (RM25) partially replacing fishmeal, and the other two were semi-purified diets with 0.062% CT (CT5) ≈ the CT content in the RM5 and 0.31% CT (CT25) ≈ the CT content in the RM25. The results of the experiments on replacing fishmeal with RM demonstrated that with the increase of RM substitution level, the weight gain rate (WGR), survival rate (SR) and specific growth rate (SGR) of largemouth bass showed a downward trend. High levels of RM led to the inhibition of digestive enzyme activity in largemouth bass, as well as the manifestation of abnormal markers related to protein, fat, and carbohydrate metabolism in both plasma and liver. RM reduced muscle protein and fat content, although the effect was not statistically significant at 5% RM level. The results of CT concentration experiments showed that the addition of 0.31% CT significantly reduced WGR, SGR, SR, protein efficiency ratio (PER) and gastrointestinal digestive enzyme activity, and significantly increased the feed coefficient ratio (FCR), plasma ammonia (PA), plasma glucose (PG), and hepatic glycogen (HG) and liver triglyceride (TG). The addition of 0.062% CT significantly reduced PG and HG. Supplementation of CT significantly reduced muscle fat. Among the groups of CT supplementation and RM, CT5 exhibited superior WGR, PER and FCR compared to RM5, along with enhanced intestinal trypsin, amylase activity, liver glutamic pyruvic transaminase (GPT) activity and decreased content of HG and liver TG; CT25 exhibited decreased activities of gastric lipase (LPS), enteric trypsin and pepsin, and increased content of total cholesterol (T-CHO) and glucose in plasma in compariosn to RM25, accompanied with no significant change in growth performance. Therefore, largemouth bass exhibited tolerance to a 5% inclusion of RM in the feed without compromising their growth, digestion and metabolic functions. The incorporation of 5% RM, equivalent to a supplemental 0.062% CT, had regulatory effects on the digestion and metabolism of largemouth bass while improving feed utilization. There was an interaction among anti-nutritional factors in RM, with synergistic effect being evident at low RM levels, whereas antagonism more promient at high levels of RM.

Growth performance, health status, gut microbiome, and expression of immune and growth-related genes of rainbow trout (Oncorhynchus mykiss) fed diets with pea protein replacement of fish meal

The need for fish meal constrains fish farming and significantly impacts sustainability of the aquaculture industry. Hence, it is important to investigate the use of plant-based protein sources in fish diets. The present study was conducted to determine the effects of different levels of fish meal (FM) replacement by pea protein (PP) in a 60-day feeding experiment in rainbow trout, Oncorhynchus mykiss. Effects on growth performance, body composition, hematology, serum biochemistry and immunology, and immune (TNF-α, IL1-ß and Il-8) and growth-related (GH and IGFI) gene expression were investigated. Five experimental diets (45% protein and 20% lipid) differed in replacement level of FM by PP at rates of 0% (control (PP0)), 25% (PP25), 50%(PP50), 75%(PP75) and 100%(PP100). Fish were fed with experimental diets in triplicate twice daily. The best growth performance was obtained in PP0 and PP25 groups. While fat ratios of fish fillets significantly differed (p < 0.05), there was no significant effects on protein ratios (p < 0.05). There was no significant change in the hematological values of fish, except those fed the PP100 diets, which displayed a reduction in eyrthocyte counts, hemoglobin content and hematocrit. As PP supplementation increased fish showed elevated serum glucose, total protein, cholesterol and myeloperoxidase activity and decreased glutamic pyruvic transaminase and alkaline phosphatase activity. Fish fed diets with between 25 and 75% replacement showed a decline in lactic acid bacteria in the gut. Significant increases in expression were observed in the liver of the PP25 fish relative to the 0% control for all immune and growth-related genes except for IL1-ß. These data suggest that up to 25% of FM can be replaced by PP without any adverse effects on rainbow trout.

Bifidobacterium Relieved Fluoride-Induced Hepatic and Ileal Toxicity via Inflammatory Response and Bile Acid Transporters in Mice.

Fluoride is a pervasive environmental contaminant. Prolonged excessive fluoride intake can inflict severe damage on the liver and intestines. Previous 16S rDNA sequencing revealed a decrease in ileal Bifidobacterium abundance during fluoride-induced hepatointestinal injury. Hence, this work aimed to investigate the possible mitigating function of Bifidobacterium on hepatointestinal injury caused by fluoride. Thirty-six 6-week-old C57BL/6J mice (equally divided between males and females) were allotted randomly to three groups: Ctrl group (distilled water), NaF group, and NaF + Ba group (100 mg/L NaF distilled water). After 10 weeks, the mice were given 1 × 109 CFU/mL Bifidobacterium solution (0.2 mL/day) intragastrically in the NaF + Ba group for 8 weeks, and the mice in other groups were given the same amount of distilled water. Dental damage, bone fluoride content, blood routine, liver and intestinal microstructure and function, inflammatory factors, and regulatory cholic acid transporters were examined. Our results showed that fluoride increased glutamic-oxalacetic transaminase (GOT), glutamic-pyruvic transaminase (GPT) activities, and the levels of lipopolysaccharide (LPS), IL-1β, IL-6, TNF-α, and IL-10 levels in serum, liver, and ileum. However, Bifidobacterium intervention alleviated fluoride-induced changes in the above indicators. In addition, Bifidobacterium reduced the mRNA expression levels of bile acid transporters ASBT, IBABP, OST-α, and OST-β in the ileum. In summary, Bifidobacterium supplementation relieved fluoride-induced hepatic and ileal toxicity via an inflammatory response and bile acid transporters in the liver and ileum of mice.

Redefining the hepatoprotective potential of Javanese turmeric (Curcuma xanthorrhiza) Kombucha towards the diethylnitrosamine-induced hepatotoxicity of mice

PurposeThis research intended the utilization of Javanese turmeric (0.4% w/v) as a kombucha substrate and analysis of its hepatoprotective activity, in comparison against nonfermented Javanese turmeric beverage (JTB) and black tea kombucha.Design/methodology/approachForty-two healthy male Balb/c mice (two- to three-week-old, 20–30 g) were divided into six groups with seven replicates each. The treatments were normal diet, normal diet + Javanese turmeric kombucha (JTK), normal diet + diethylnitrosamine (DEN), DEN + JTB, DEN + JTK, DEN + black tea kombucha. Kombuchas and JTB were given at 0.3 mL/20 g BW/d. DEN was induced intraperitoneally at a dose of 100 mg/kg. Observed biomarkers were blood serum glutamate pyruvate transaminase (SGPT) and serum glutamate oxaloacetate transaminase (SGOT) activity, serum malonaldehyde (MDA), as well as liver histology. Data were analyzed using analysis of variance.FindingsAmong DEN-induced groups, JTK significantly (p < 0.05) diminished the level of blood SGPT, SGOT and serum MDA. JTK also had lower blood SGPT (8.604 ± 2.195 U/L) and serum MDA levels (2.884 ± 0.083 nM/mL) compared to the normal group (8.604 ± 2.195 U/L and 5.050 ± 0.998 nM/mL, respectively). JTK also produced the least damaged liver-cell numbers.Originality/valueJTK demonstrated better hepatoprotective activity compared to JTB.

Dietary supplementation of prebiotic yeast Saccharomyces cerevisiae cell wall promotes growth performance and intestinal health in broiler chickens challenged with Clostridium perfringens

ABSTRACT 1. This study evaluated the effectiveness of yeast (S accharomyces cerevisiae) cell wall (YCW) supplementation on the growth performance, carcase characteristics, serum biomarkers, liver function, ileal histology and microbiota of broiler chickens challenged with Clostridium perfringens (C. perfringens). 2. In a 35-d trial, 240 chicks aged 1-d-old were randomly assigned to one of four treatment groups, each with 10 replicates: control (CON) with no challenge or additives, challenged with C. perfringens (CHAL), CHAL and supplemented with YCW at either 0.25 g/kg (YCW0.25) or 0.5 g/kg (YCW0.5). 3. In comparison to CON, the CHAL birds had reduced growth performance, survival rate, dressing percentage, breast meat yield, levels of total protein (TP), globulin (GLO), glucose (GLU), total antioxidant capacity (T-AOC) and total superoxide dismutase (T-SOD), as well as a decreased Lactobacillus population (P < 0.01). Additionally, this group showed elevated levels of glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and C. perfringens count (P < 0.01). Compared to CHAL, the YCW0.25 or YCW0.5 groups had improved growth performance, survival rate, dressing percentage, breast meat yield, levels of TP, GLO, GLU, and T-AOC, as well as the activities of T-SOD, GOT, and GPT, villus height, villus surface area, villus height to crypt depth ratio, and the populations of both Lactobacillus and C. perfringens; (P < 0.01). 4. The data suggested that YCW supplementation at either 0.25 or 0.50 g/kg can restore the growth performance of broiler chickens during a C. perfringens challenge.

Effects of Varying Dietary Protein Levels on Growth Performance, Survival, Body Composition, Haemato-Biochemical Profile, and Metabolic Responses of Hypselobarbus jerdoni (Day, 1870) Juveniles

A 60-day feeding trial has been carried out to access the optimal dietary crude protein (DCP) requirement of juvenile Jerdon’s carp, Hypselobarbus jerdoni. Six isoenergetic (around 400 Kcal DE/100 g), isolipidic (60.80 g/kg), and heteronitrogenous diets were prepared with graded levels of protein, viz., 200, 250, 300, 350, 400, and 450 g/kg (TCP 20-TCP 45 with 50 g/kg incremental level). A completely randomised design (CRD) was used to distribute 270 Jerdon’s carp juveniles (6.12–6.18 g) in six experimental groups in triplicates (15 fish/tank, 200 l water capacity). After 60 days, the fishes of the TCP30 group showed significantly higher weight gain, final body weight, and percentage weight gain (WG%). The fish of the TCP30 group exhibited a significantly higher feed efficiency ratio and specific growth rate. On the other hand, a significantly lower feed conversion ratio is recorded in the TCP30 and TCP35 groups. The protein efficiency ratio of fish was reduced significantly with the elevated DCP level. Whole-body moisture, lipid, and total ash contents of H. jerdoni were remain unaffected by DCP levels. However, DCP levels significantly influenced the whole-body protein of H. jerdoni juveniles, with significantly higher values noticed in the TCP30 and TCP35 groups. The TCP35, TCP40, and TCP45 groups exhibited significantly higher (p&lt;0.05) protease activity, and the amylase activity showed a decreasing trend in response to dietary protein levels. A significantly (p&lt;0.05) higher SOD and CAT activity were observed in the TCP20 and TCP25 groups. However, lower hepatic glutamate pyruvate transaminase and glutamic-oxaloacetic transaminase activity were observed in the TCP30 and TCP35 groups, respectively. Furthermore, based on broken-line linear and second-order polynomial regression with respect to WG%, the optimal dietary crude protein requirement of H. jerdoni cultured for 60 days was found to be 309.72 and 316.40 g/kg.

Effect of biosal®, deltamethrin and lambda-cyhalothrin on the activity of GOT, GPT and total protein contents in two fodder pests Hermolaus modestus and Hermolaus ocimumi.

The assessment of the comparative effect of biosal (phytopesticide), deltamethrin, and lambda-cyhalothrin (pyrethroids) were made against two fodder pests, Hermolaus modestus and Hermolaus ocimumi by filter paper impregnation method. The activity of total protein contents, GPT (glutamic-pyruvic transaminase) and GOT (glutamic oxaloacetate transaminase) were affected in Hermolaus modestus and Hermolaus ocimumi against biosal, deltamethrin, and lambda cyhalothrin. The activity of total protein contents in H. modestus was 31.053%, 4.607%, and 24.575%, against biosal, deltamethrin, and lambda-cyhalothrin, respectively. The activity of total protein contents was observed as 24.202%, 15.25%, and 56.036% against deltamethrin, lambda-cyhalothrin, and biosal, respectively in H. ocimumi. The activity of GOT was observed as 98.675% for biosal 33.95% for deltamethrin and 83.619% for lambda-cyhalothrin in H. modestus. The GOT activity was estimated in H. ocimumi as 78.831%, 47.645%, and 71.287% against biosal, deltamethrin, and lambda-cyhalothrin, respectively. The efficacy of GPT enzyme against biosal, deltamethrin, and lambda-cyhalothrin was calculated as 89.26%, 73.07%, and 47.58%, respectively in H. modestus. The H. ocimumi showed GPT activity as 77.58% for biosal, 68.84% for deltamethrin, and 52.67% for lambda-cyhalothrin, respectively.

Optimizing planting density and nitrogen application to mitigate yield loss and improve grain quality of late-sown wheat under rice-wheat rotation

Late sowing is a critical factor that hinders achieving high-yield, good-quality wheat under rice-wheat rotation. Understanding the physiological basis and regulatory pathways that lead to late-sown wheat of high yield and good quality is crucial for developing effective cultivation strategies. A 2-year field experiment was conducted to investigate the effects of sowing date, nitrogen (N) application rate, and planting density on wheat yield, grain quality, population characteristics, and the underlying physiological factors. The results revealed significant interactions among the sowing date, planting density, and N application in regulating both yield and quality. Late sowing reduced grain yield primarily by reducing the number of spikes and kernels. However, the latter was improved by increasing N application and the planting density, thus mitigating the yield losses caused by late sowing. Moreover, the grain protein content (GPC) and wet gluten content (WGC) increased with delayed sowing dates and higher N rates, but decreased with increased planting densities. For wheat yields over 9,000 or 7,500 kg ha–1, the latest sowing date should not be later than Nov. 4 or 15, respectively. In addition, specific criteria should be met, including a maximum of 1.5 and 1.0 million stems and tillers ha–1, a maximum leaf area index of 6.7 and 5.5, and a dry matter accumulation (DMA) at anthesis of 14,000 and 12,000 kg ha–1, respectively. For high-yield, good-quality late-sown wheat, the optimal combination is a 25% increase in the N rate (300 kg N ha–1) and a planting density of 2.25 million (N300D225) or 3.75 million (N300D375) plants ha–1 for 10- or 20-day delays in sowing, respectively. These combinations result in higher leaf net photosynthetic rate; higher activities of leaf nitrate reductase, glutamine synthetase, and grain glutamic pyruvic transaminase; and a lower sugar-N ratio during post-anthesis.

Effects of Synthetic Astaxanthin on the Growth Performance, Pigmentation, Antioxidant Capacity, and Immune Response in Black Tiger Prawn (Penaeus monodon).

Synthetic astaxanthin is an effective nutritional strategy for improving shrimp body color and promoting growth. However, the optimal amount of astaxanthin in feed also varies with the synthetic technology and purity. In the present study, five diets containing different doses of synthetic astaxanthin (0% (CON), 0.02% (AX0.02), 0.04% (AX0.04), 0.08% (AX0.08), and 0.16% (AX0.16)) were administered to Penaeus monodon (initial body weight: 0.3 ± 0.03 g) for 8 weeks. With an increase in astaxanthin content in feed, weight gain and specific growth rate increased initially and subsequently decreased, with the highest value appearing at AX0.08. Dietary astaxanthin supplementation obviously improved the carapace and muscle color by enhancing astaxanthin pigmentation. Meanwhile, the fatty acid profile was altered by dietary astaxanthin, as evidenced by a decline in palmitic acid proportion, along with an increase in n-3 polyunsaturated fatty acids (n-3 PUFA) contents in muscle. In addition, dietary astaxanthin supplementation regulated prawn's antioxidant capacity. In the hemolymph, the activities of glutamic pyruvic transaminase (GPT) showed a significantly decrease trend with linear effect. The activities of glutamic oxaloacetic transaminase (GOT) and the contents of malondialdehyde (MDA) were first downregulated and then upregulated with significantly quadratic pattern. In the hepatopancreas, the activities of superoxide dismutase (SOD) and the contents of MDA were significantly downregulated with the increase of dietary astaxanthin levels. Reduced glutathione (GSH) contents and catalase (CAT) activities were also significantly decreased in group AX0.08. Correspondingly, astaxanthin decreased GSH and MDA contents under transportation stress. Moreover, the mRNA expression of immune genes (traf6, relish, and myd88) were inhibited by dietary astaxanthin supplementation. Based on the results of polynomial contrasts analysis and Duncan's test, dietary synthetic astaxanthin is a suitable feed additive to improve the growth, body color, antioxidant capacity, and nonspecific immunity of P. monodon. According to the second-order polynomial regression analysis based on the weight gain, the optimal supplementation level of dietary astaxanthin was 90 mg kg-1 in P. monodon.

Improvement of sea buckthorn (Hippophae rhamnoides L.) flavonoids on the antioxidant and immune performance of Yellow River carp (Cyprinus carpio L.) fed high-carbohydrate diet

High-carbohydrate (HC) diets may lead to the deterioration of the antioxidant and immune properties of Yellow River carp and the healthy development of the industry. Studies in mammals have found that sea buckthorn flavonoids (SF) improve antioxidant and immune performance. Therefore, this study comprehensively evaluated the effects of SF on Yellow River carp using in vitro and feeding trials with an HC diet. Control (C, 27.23 %), high-carbohydrate (HC, 42.99 %), and HC + SF (0.1 %, 0.2 %, and 0.4 %) groups were studied in a 10-week aquaculture experiment. The main findings were as follows: (1) SF scavenged O2·‐, ·OH, and DPPH free radicals in vitro, which gradually increased with the SF concentration. (2) The antioxidant and immune performance of Yellow River carp was enhanced by dietary supplementation with SF, which involved the regulation of activities of antioxidant and immune enzymes, as well as their changes at the transcription and protein levels. In terms of antioxidant properties, compared to the HC group, HC + SF significantly decreased the activities of glutamic-oxaloacetic transaminase and glutamic-pyruvic transaminase and the contents of H2O2 and malondialdehyde in the serum and hepatopancreas. The activities of glutathione, glutathione-Px, superoxide dismutase, catalase, and total antioxidant activity in the HC-diet group. In contrast, the addition of SF increased antioxidant enzyme activity. In the hepatopancreas and muscles, SF regulated and activated Nrf2-Keap1, a key signaling pathway for oxidative stress. SF significantly increased the mRNA expression levels of downstream genes (gr, ho-1, cat, and sod) regulated by nrf2. In terms of immune performance, 0.4 % SF markedly increased the activity of immune-related enzymes. SF inhibited the gene expression of pro-inflammatory factors induced by the HC diet and promoted the gene expression of anti-inflammatory factors. In addition, the resistance of Yellow River carp to Aeromonas hydrophila was enhanced by SF. In summary, SF supplementation can reduce oxidative stress and inflammatory harm caused by the HC diet and improve the antioxidant and immune performance of Yellow River carp to varying degrees.

Effects of Replacing Fish Meal with Stickwater Hydrolysate and Meal on the Growth, Serum Biochemical Indexes, and Muscle Quality of Yellow Catfish (Tachysurus fulvidraco)

An eight-week feeding experiment was conducted to evaluate the effects of replacing fish meal with stickwater hydrolysate (SWH) or stickwater hydrolysate meal (SWM) on the growth, serum biochemical parameters, intestinal digestive enzyme activity, and muscle quality of yellow catfish (Tachysurus fulvidraco). The control diet (CON) contained 30% fish meal and the remaining five diets were substituted for fish meal with 2.5% (SWM2.5), 5% (SWM5) SWM, and 5% (SWH5), 10% (SWH10), and 15% (SWH15) SWH, respectively. The results showed that there were no significant differences in weight gain rate, feed conversion rate, survival rate, hepatosomatic index, and viscerosomatic index among the groups. The substitution of fish meal with SWH significantly augmented the serum triglyceride and total cholesterol levels, whereas urea nitrogen content exhibited a reduction proportional to the replacement ratio. The incorporation of SWH led to a notable rise in glutamate-pyruvate transaminase activity, albeit with a gradual decline as the substitution ratio escalated. Relative to the CON group, the SWH5 group displayed a significant reduction in serum superoxide dismutase activity and a significant elevation in serum catalase activity. The substitution of fish meal with SWM yielded noticeable increments in the activities of complement 3, immunoglobulin M, and alkaline phosphatase. Neither SWH nor SWM exerted a substantial influence on intestinal amylase activity. Regarding muscle characteristics, neither SWH nor SWM showed a marked effect on hardness and springiness; however, adhesiveness, cohesiveness, gumminess, and chewiness properties exhibited enhancement as the proportion of fish meal replacement increased. In conclusion, within this experimental context, substituting fish meal with SWH and SWM did not adversely impact the growth and meat quality of yellow catfish. Specifically, replacing 15% fish meal with stickwater hydrolysate and 5% fish meal with stickwater hydrolysate meal contributed to an enhanced immune capacity in yellow catfish to a certain extent.

Synthetic astaxanthin has better effects than natural astaxanthins on growth performance, body color and n-3 PUFA deposition in black tiger prawn (Penaeus monodon)

Astaxanthin is an efficient nutrition strategy to relieve oxidative stress and enhance crustacean body color. Because of technological and economic benefits, synthetic astaxanthin is favored by the aquatic feed industry. However, whether synthetic astaxanthin can achieve the effects of natural astaxanthin needs further investigation. Here, we evaluated the effects of synthetic astaxanthin and natural astaxanthin (dietary supplementation with Haematococcus pluvialis and Phaffia rhodozyma) on the growth performance, body color, fatty acid composition and antioxidant capacity of black tiger prawn (Penaeus monodon). Compared with the control group (no astaxanthin supplementation), only synthetic astaxanthin significantly increased the weight gain rate (WG) and specific growth rate (SGR), together with the elevation of feed efficiency ratio (FER). All three types of astaxanthin enhanced body color by increasing astaxanthin deposition. The highest value of (a*) in the carapace and muscle occurred in the synthetic astaxanthin group. Synthetic astaxanthin significantly increased the n-3 polyunsaturated fatty acid (n-3 PUFA) contents in muscle. As expected, different sources of astaxanthin inhibited glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) activities in the hemolymph and decreased the contents of reduced glutathione (GSH) and malondialdehyde (MDA) in the hepatopancreas. Correspondingly, the structure of hepatopancreas was enhanced by astaxanthin. In addition, three types of astaxanthin almost indistinguishably enhanced the ability of shrimps to resist stress caused by transportation simulation. In conclusion, these results indicate that dietary synthetic astaxanthin has a better effect than natural astaxanthin on growth promotion, body color enhancement and n-3 PUFA deposition in muscle of black tiger prawn. Hence, synthetic astaxanthin is a suitable feed additive to promote the healthy and sustainable development of the shrimp industry.

Gambaran Aktivitas Serum Glutamic Pyruvic Transaminase (SGPT) pada Petugas Batubara di Talang Duku Kabupaten Muaro Jambi

Dust generated from the coal production process is known to have various cytotoxic and carcinogenic contents. Prolonged exposure to coal dust can trigger health problems and affect the function of organs, one of which is the liver. Liver damage is characterized by increased activity of the enzyme Serum Glutamic Pyruvic Transaminase (SGPT), which is an indicator of liver function tests. Groups at risk of exposure to coal dust include people who are in the coal transport traffic area and the coal workers themselves. The study was conducted to describe the activity of Serum Glutamic Pyruvic Transaminase (SGPT) in coal workers at Talang Duku, Muaro Jambi. The research was conducted descriptively with a cross sectional approach. The research was conducted on 30 coal workers at Talang Duku, Muaro Jambi. The Automatic method is used to check SGPT activity using a Chemistry Analyzer. Based on the results of the study most of respondents had normal SGPT activities (73.33%), both in respondents with characteristics aged &lt;40 years (46.67%), working period &lt;5 years (36.66%) and field work division (60%). The results of examining SGPT activity in coal workers at Talang Duku, Muaro Jambi, showed that most of respondents had normal SGPT activity (73.33%) as well as respondents with spesific characteristics.

Effects of White Fish Meal Replaced by Low-Quality Brown Fish Meal with Compound Additives on Growth Performance and Intestinal Health of Juvenile American Eel (Anguilla rostrata).

With a reduced supply and increased price of white fish meal (WFM), the exploration of a practical strategy to replace WFM is urgent for sustainable eel culture. A 70-day feeding trial was conducted to evaluate the effects of replacing WFM with low-quality brown fish meal (LQBFM) with compound additives (CAs) on the growth performance and intestinal health of juvenile American eels (Anguilla rostrata). The 300 fish (11.02 ± 0.02 g/fish) were randomly distributed in triplicate to four groups (control group, LQBFM20+CAs group, LQBFM30+CAs group and LQBFM40+CAs group). They were fed the diets with LQBFM replacing WFM at 0, 20%, 30% and 40%, respectively. The CAs were a mixture of Macleaya cordata extract, grape seed proanthocyanidins and compound acidifiers; its level in the diets of the trial groups was 0.50%. No significant differences were found in the growth performance between the control and LQBFM20+CAs groups (p > 0.05), whereas those values were significantly decreased in LQBFM30+CAs and LQBFM40+CAs groups (p < 0.05). Compared to the control group, the activity of glutamic-pyruvic transaminase was significantly increased in LQBFM30+CAs and LQBFM40+CAs groups, while lysozyme activity and complement 3 level were significantly decreased in those two groups (p < 0.05). There were decreased antioxidant potential and intestinal morphological indexes in the LQBFM30+CAs and LQBFM40+CAs groups, and no significant differences in those parameters were observed between the control group and LQBFM20+CAs group (p > 0.05). The intestinal microbiota at the phylum level or genus level was beneficially regulated in the LQBFM20+CAs group; similar results were not shown in the LQBFM40+CAs group. In conclusion, with 0.50% CA supplementation in the diet, LQBFM could replace 20% of WFM without detrimental effects on the growth and intestinal health of juvenile American eels and replacing 30% and 40%WFM with LQBFM might exert negative effects on this fish species.

In Vitro Study on Spermicidal Action of Hydro-methanol Extract of Tinospora cordifolia (Willd.) Stem in Rat and Human Sperm: a Comparative Analysis.

Human fertility regulation is a major way to control overpopulation. In this perspective, this study emphasized the in vitro effect of hydro-methanol extract of Tinospora cordifolia (TCHME) stem for spermicidal and reproductive hypo-functions using human and rat samples. Control, 0.5-, 1-, and 2-mg TCHME-charged groups were considered to assess the relevant parameters. Levels of spermiological parameters like sperm motility, viability, the integrity of plasma and acrosomal membrane, and nuclear chromatin decondensation were significantly reduced (p < 0.05) in the dose- and duration-dependent TCHME-charged groups compared to the control. The inhibitory concentration 50 (IC50) of TCHME on motile human and rat sperms were 0.8 and 0.4 mg/ml, respectively. Testicular androgenic key enzymes and antioxidant enzymes (human sperm pellet, testes, and epididymis of rat)' activities were significantly diminished (p < 0.05), while antioxidant enzymes' activities were significantly elevated (p < 0.05) in renal and insignificantly (p > 0.05) elevated in hepatic tissues of rat in TCHME-charged groups compared to the control. Significant elevation (p < 0.05) of thiobarbituric acid reactive substances (TBARS)' level in human sperm pellet, testes, and epididymis of rats and significant diminution (p < 0.05) in TBARS levels of liver and kidney were observed in TCHME-charged groups. It focused that TCHME is more potent for stress imposition on reproductive tissues and sperm compared to the other tested tissues. Non-significant alterations (p > 0.05) in glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) activities in the said organs of rat indicated its non-toxic effect. It highlighted that TCHME possesses spermicidal and reproductive tissue-specific effects which strengthen the possibilities of male contraceptive development from it.

The Protective Effect of a Dietary Extract of Mulberry (Morus alba L.) Leaves against a High Stocking Density, Copper and Trichlorfon in Crucian Carp (Carassius auratus).

This study was designed to examine the protective effects of the extract of mulberry (Morus alba L.) leaves (EML) on crucian carp (Carassius auratus) against a high stocking density, Cu exposure and trichlorfon exposure, which adversely impact fish growth performance, feed intake and fish locomotion. High stocking densities decreased the activities of amylase, lipase, trypsin, Na+/K+-ATPase and alkaline phosphatase (AKP), and increased the content of malonaldehyde (MDA) in fish digestive organs, indicating an impairment of the digestive function and a disturbance of the antioxidant status. Cu exposure increased the activities of glutamate-oxaloacetate transaminase (GOT) and glutamate-pyruvate transaminase (GPT) in fish digestive organs, suggesting the activation of amino acid metabolism. Furthermore, trichlorfon exposure reduced the activities of lactate dehydrogenase (LDH), glutathione reductase (GR), GOT and GPT, and the capacities of the anti-superoxide anion (ASA) and anti-hydroxyl radical (AHR) in fish muscles, indicating a disruption of the bioenergetic homeostasis and antioxidant status. Our present study indicates that dietary EML supplementation relieved the detrimental effects induced by these stressors.

Investigations of hemato-biochemical and histopathological parameters, and growth performance of walking catfish (Clarias batrachus) exposed to PET and LDPE microplastics

Fish inhabiting various trophic levels are affected differently as the presence of microplastic (MP) in the water column and their ingestion by fish varies. Walking catfish (Clarias batrachus) inhabits the bottom of the water bodies. To understand the effects of MP, we exposed C. batrachus to two types of MP – polyethylene terephthalate (PET) and low-density polyethylene (LDPE) for 60 days. After exposure, hematological indices, mainly red blood cells and hemoglobin levels decreased, and white blood cells increased significantly compared to the control group (p < 0.05). A significant increase in the levels of blood urea and glucose was observed, and serum glutamic pyruvate transaminase and serum glutamyl oxaloacetic transaminase activity remained elevated (p < 0.05). Histopathological examination of the liver, kidney, intestine, and gills showed morphological alterations. Moreover, MP exposure caused growth retardation (p < 0.05) in C. batrachus. Widespread pollution of water bodies by MP may impose serious ecological risks to bottom-feeding fish in Bangladesh.

Scoparia dulcis L. Extract Relieved High Stocking Density-Induced Stress in Crucian Carp (Carassius auratus).

The objective of this study was to investigate the effects of Scoparia dulcis extract (SDE) on stress induced by high stocking density and Cu and trichlorfon exposure in crucian carp (Carassius auratus). The results showed that these stressors exerted detrimental effects in fish, such as inhibition of growth performance, reduced feed intake, and interruption of fish locomotion. Under high stocking density, dietary SDE supplementation increased the content of reduced glutathione (GSH) and the activities of amylase, catalase (CAT), and glutathione reductase (GR) and decreased the content of malonaldehyde (MDA) in the intestine of crucian carp. A similar trend was presented in the hepatopancreas under Cu exposure. Dietary SDE supplementation enhanced the activities of CAT, superoxide dismutase (SOD), glutathione peroxidase (GPx), lactate dehydrogenase, glutamate-oxaloacetate transaminase, and glutamate-pyruvate transaminase in the muscle of crucian carp under trichlorfon exposure. The optimum dietary SDE supplementation levels were 4.07, 4.33, and 3.95 g kg-1 diet based on the recovery rate of weight gain (RWG), feed intake (FI), and inhibitory rate of rollover (IR) for crucian carp under high stocking density and Cu and trichlorfon exposure, respectively. Overall, dietary supplementation with SDE may be a useful nutritional strategy for relieving these stresses in aquatic animals.

Dinoflagellate Karenia mikimotoi on the growth performance, antioxidative responses, and physiological activities of the rotifer Brachionus plicatilis.

The harmful dinoflagellate Karenia mikimotoi is responsible for the mortality of aquatic animals. However, the mechanism behind these toxic effects has not been fully determined. Herein, the toxic effects of K. mikimotoi on the growth performance, antioxidative responses, physiological activities, and energetic substance contents of rotifer Brachionus plicatilis were assessed. Rotifers were exposed to Nannochloropsis salina (Eustigmatophyceae), K. mikimotoi, and a mixture of N. salina and K. mikimotoi with biomass ratio proportions of 3:1, 1:1, and 1:3, respectively. Results indicated that K. mikimotoi negatively affected the population growth, survival, and specific growth rates of rotifers within 24 h. The level of reactive oxygen species (ROS), the content of malondialdehyde, and the activity of amylase increased. However, the total antioxidant capacity level, pepsase, cellulase, alkaline phosphatase, xanthine oxidase, and lactate dehydrogenase activities, and glycogen and protein contents decreased with increasing proportions of K. mikimotoi. The mixture of 50% N. salina and 50% K. mikimotoi promoted the increase in glutamic-pyruvic transaminase activity and triglyceride content. These findings underscore ROS-mediated antioxidative responses, physiological responses, and energetic substance content changes in B. plicatilis work together to affect population dynamics inhibition of rotifers by K. mikimotoi.
