Photosynthetically-powered phototactic active nematic liquid crystal fluids and gels

One of the most ancient forms of life dating to ~3.5 billion years ago, cyanobacteria are highly abundant organisms that convert light into energy and motion, often within conjoined filaments and larger colonies that attract a great deal of interest but their active nematic behavior remained unexplored. Here we demonstrate how light causes a spontaneous self-assembly of two- and three-dimensional active nematic states of cyanobacterial filaments, with a plethora of topological defects. We quantify light-controlled evolutions of orientational and velocity order parameters during the transition between disordered and orientationally ordered states of photosynthetic active matter, as well as the subsequent active nematic’s fluid-gel transformation. Patterned illumination and foreign inclusions with different shapes interact with cyanobacterial active nematics in nontrivial ways while inducing interfacial boundary conditions and fractional boojum defects. Our phototactic model system promises opportunities to systematically explore fundamental properties and technological utility of the liquid crystalline active matter.

Flow patterns and defect dynamics of active nematic liquid crystals under an electric field.

The effects of an electric field on the flow patterns and defect dynamics of two-dimensional active nematic liquid crystals are numerically investigated. We found that field-induced director reorientation causes anisotropic active turbulence characterized by enhanced flow perpendicular to the electric field. The average flow speed and its anisotropy are maximized at an intermediate field strength. Topological defects in the anisotropic active turbulence are localized and show characteristic dynamics with simultaneous creation of two pairs of defects. A laning state characterized by stripe domains with alternating flow directions is found at a larger field strength near the transition to the uniformly aligned state. We obtained periodic oscillations between the laning state and active turbulence, which resembles an experimental observation of active nematics subject to anisotropic friction.

Active nematic liquid crystals simulated by particle-based mesoscopic methods.

Two Multi-particle collision dynamics algorithms that simulate nematic liquid crystals are generalised to reproduce active behaviour. One of the algorithms is due to Shendruk and Yeomans and is based on particles that carry an orientation vector ordered by a mean-field energy [T. N. Shendruk and J. M. Yeomans, Soft Matter, 2015, 11, 5101]. In the other algorithm, due to Mandal and Mazza, particles possess an order parameter tensor which evolves according to the Qian-Sheng model of nematohydrodynamics [S. Mandal and M. G. Mazza, Phys. Rev. E, 2019, 99, 063319]. For both methods activity is incorporated through a force proportional to the divergence of the local average order parameter tensor. Both implementations produce disclination curves in the nematic fluid that undergo nucleation and self-annihilation dynamics. Topological defects are found to be consistent with those observed in recent experiments of three-dimensional active nematics. Results permit to compare the length-scales over which the different nematic Multi-particle collision dynamics methods operate. The structure and dynamics of the orientation and flow fields agree with those obtained recently in numerical studies of continuum three-dimensional active nematics. Overall, our results open the opportunity to use mesoscopic particle-based approaches to study active liquid crystals in situations such as nonequilibrium states driven by flow or colloidal particles in active anisotropic solvents.

Topological defect-mediated morphodynamics of active–active interfaces

Physical interfaces widely exist in nature and engineering. Although the formation of passive interfaces is well elucidated, the physical principles governing active interfaces remain largely unknown. Here, we combine simulation, theory, and cell-based experiment to investigate the evolution of an active-active interface. We adopt a biphasic framework of active nematic liquid crystals. We find that long-lived topological defects mechanically energized by activity display unanticipated dynamics nearby the interface, where defects perform "U-turns" to keep away from the interface, push the interface to develop local fingers, or penetrate the interface to enter the opposite phase, driving interfacial morphogenesis and cross-interface defect transport. We identify that the emergent interfacial morphodynamics stems from the instability of the interface and is further driven by the activity-dependent defect-interface interactions. Experiments of interacting multicellular monolayers with extensile and contractile differences in cell activity have confirmed our predictions. These findings reveal a crucial role of topological defects in active-active interfaces during, for example, boundary formation and tissue competition that underlie organogenesis and clinically relevant disorders.

Coexistence of Active and Hydrodynamic Turbulence in Two-Dimensional Active Nematics.

In active nematic liquid crystals, activity is able to drive chaotic spatiotemporal flows referred to as active turbulence. Active turbulence has been characterized through theoretical and experimental work as a low Reynolds number phenomenon. We show that, in two dimensions, the active forcing alone is able to trigger hydrodynamic turbulence leading to the coexistence of active and inertial turbulence. This type of flow develops for sufficiently active and extensile flow-aligning nematics. We observe that the combined effect of an extensile nematic and large values of the flow-aligning parameter leads to a broadening of the elastic energy spectrum that promotes a growth of kinetic energy able to trigger an inverse energy cascade.

Vector formalism for active nematic liquid crystals in two dimensions.

Specific features of two-dimensional nematodynamics give rise to shortfalls of the tensor representation of the nematic order parameter commonly used in computations, especially in theory of active matter. The alternative representation in terms of the vector order parameter follows with small adjustments the classical director-based theory, but is applicable to 2D problems where both nematic alignment and deviation from the isotropic state are variable. Stability analysis of nematic alignment and flow is used as a testing ground. A director-based analysis demonstrates a shortfall of the standard theory, which does not ensure relaxation to equilibrium in a passive system. It also demonstrates the inadequacy of the director-based description, which misses a stabilizing effect of perturbations of the modulus ensuring stability of a passive system on scales far exceeding the healing length.

Assembling Microtubule-Based Active Matter.

Studied for more than a century, equilibrium liquid crystals provided insight into the properties of ordered materials, and led to commonplace applications such as display technology. Active nematics are a new class of liquid crystal materials that are driven out of equilibrium by continuous motion of the constituent anisotropic units. A versatile experimental realization of active nematic liquid crystals is based on rod-like cytoskeletal filaments that are driven out of equilibrium by molecular motors. We describe protocols for assembling microtubule-kinesin based active nematic liquid crystals and associated isotropic fluids. We describe the purification of each protein and the assembly process of a two-dimensional active nematic on a water-oil interface. Finally, we show examples of nematic formation and describe methods for quantifying their non-equilibrium dynamics.

Catapulting of topological defects through elasticity bands in active nematics.

Active materials are those in which individual, uncoordinated local stresses drive the material out of equilibrium on a global scale. Examples of such assemblies can be seen across scales from schools of fish to the cellular cytoskeleton and underpin many important biological processes. Synthetic experiments that recapitulate the essential features of such active systems have been the object of study for decades as their simple rules allow us to elucidate the physical underpinnings of collective motion. One system of particular interest has been active nematic liquid crystals (LCs). Because of their well understood passive physics, LCs provide a rich platform to interrogate the effects of active stress. The flows and steady state structures that emerge in an active LCs have been understood to result from a competition between nematic elasticity and the local activity. However most investigations of such phenomena consider only the magnitude of the elastic resistance and not its peculiarities. Here we investigate a nematic liquid crystal and selectively change the ratio of the material's splay and bend elasticities. We show that increases in the nematic's bend elasticity specifically drives the material into an exotic steady state where elongated regions of acute bend distortion or "elasticity bands" dominate the structure and dynamics. We show that these bands strongly influence defect dynamics, including the rapid motion or "catapulting" along the disintegration of one of these bands thus converting bend distortion into defect transport. Thus, we report a novel dynamical state resultant from the competition between nematic elasticity and active stress.

Orientational Correlations in Active and Passive Nematic Defects.

We investigate the emergence of orientational order among +1/2 disclinations in active nematic liquid crystals. Using a combination of theoretical and experimental methods, we show that +1/2 disclinations have short-range antiferromagnetic alignment, as a consequence of the elastic torques originating from their polar structure. The presence of intermediate -1/2 disclinations, however, turns this interaction from antialigning to aligning at scales that are smaller than the typical distance between like-sign defects. No long-range orientational order is observed. Strikingly, these effects are insensitive to material properties and qualitatively similar to what is found for defects in passive nematic liquid crystals.

Interplay of Active Stress and Driven Flow in Self-Assembled, Tumbling Active Nematics

Lyotropic chromonic liquid crystals (LCLCs) are a special type of hierarchical material in which self-assembled molecular aggregates are responsible for the formation of liquid crystal phases. Thanks to its unusual material properties and bio compatibility, it has found wide applications including the formation of active nematic liquid crystals. Recent experiments have uncovered tumbling character of certain LCLCs. However, how tumbling behavior modifies structure and flow in driven and active nematics is poorly understood. Here, we rely on continuum simulation to study the interplay of extensile active stress and externally driven flow in a flow-tumbling nematic with a low twist modulus to mimic nematic LCLCs. We find that a spontaneous transverse flow can be developed in a flow-tumbling active nematic confined to a hybrid alignment cell when it is in log-rolling mode at sufficiently high activities. The orientation of the total spontaneous flow is tunable by tuning the active stress. We further show that activity can suppress pressure-driven flow of a flow-tumbling nematic in a planar-anchoring cell but can also promote a transition of the director field under a pressure gradient in a homeotropic-anchoring cell. Remarkably, we demonstrate that the frequency of unsteady director dynamics in a tumbling nematic under Couette flow is invariant against active stress when below a threshold activity but exhibits a discontinuous increase when above the threshold at which a complex, periodic spatiotemporal director pattern emerges. Taken together, our simulations reveal qualitative differences between flow-tumbling and flow-aligning active nematics and suggest potential applications of tumbling nematics in microfluidics.

Active flows on curved surfaces

We consider a numerical approach for a covariant generalized Navier–Stokes equation on general surfaces and study the influence of varying Gaussian curvature on anomalous vortex-network active turbulence. This regime is characterized by self-assembly of finite-size vortices into linked chains of anti-ferromagnet order, which percolate through the entire surface. The simulation results reveal an alignment of these chains with minimal curvature lines of the surface and indicate a dependency of this turbulence regime on the sign and the gradient in local Gaussian curvature. While these results remain qualitative and their explanations are still incomplete, several of the observed phenomena are in qualitative agreement with experiments on active nematic liquid crystals on toroidal surfaces and contribute to an understanding of the delicate interplay between geometrical properties of the surface and characteristics of the flow field, which has the potential to control active flows on surfaces via gradients in the spatial curvature of the surface.

Chaotic dynamics of active topological defects

ABSTRACT Topological defects are interesting phenomena which can be observed in ordered phases such as oriented active fluids or nematic liquid crystals. Topological defects are determined by the overall structure of the director field in an active fluid in nematic phase and by exerting force to the units of the active particles they can interact or cause motion in the environment. Studying them as particles with dynamical equations, instead of studying the director field of the nematic environment, would provide us the power to study the characteristics of their motion. The equations of motion for multi-defect systems have been previously studied and in this work we focus on the chaotic properties of the dynamics and analyze the effect of activity on the transition of such systems to chaos by numerically estimating their Lyapunov exponents. We show the importance of the defects for this transition (as they are active in the sense of having self-propulsion) and numerically derive critical activity values for multi-defect systems in which this transition occurs.

Spatiotemporal control of liquid crystal structure and dynamics through activity patterning.

Active materials are capable of converting free energy into mechanical work to produce autonomous motion, and exhibit striking collective dynamics that biology relies on for essential functions. Controlling those dynamics and transport in synthetic systems has been particularly challenging. Here, we introduce the concept of spatially structured activity as a means of controlling and manipulating transport in active nematic liquid crystals consisting of actin filaments and light-sensitive myosin motors. Simulations and experiments are used to demonstrate that topological defects can be generated at will and then constrained to move along specified trajectories by inducing local stresses in an otherwise passive material. These results provide a foundation for the design of autonomous and reconfigurable microfluidic systems where transport is controlled by modulating activity with light.

Living proof of effective defects

A class of biological matter including elongated cells and filaments can be understood in the framework of active nematic liquid crystals. Within these systems, topological defects emerge and give rise to remarkable collective behaviours.

Topological defects promote layer formation in Myxococcus xanthus colonies

The soil bacterium Myxococcus xanthus lives in densely packed groups that form dynamic three-dimensional patterns in response to environmental changes, such as droplet-like fruiting bodies during starvation. The development of these multicellular structures begins with the sequential formation of cell layers in a process that is poorly understood. Using confocal three-dimensional imaging, we find that motile rod-shaped M. xanthus cells are densely packed and aligned in each layer, forming an active nematic liquid crystal. Cell alignment is nearly perfect throughout the population except at point defects that carry half-integer topological charge. We observe that new cell layers preferentially form at the position of +1/2 defects, whereas holes preferentially open at -1/2 defects. To explain these findings, we model the bacterial colony as an extensile active nematic fluid with anisotropic friction. In agreement with our experimental measurements, this model predicts an influx of cells toward +1/2 defects, and an outflux of cells from -1/2 defects. Our results suggest that cell motility and mechanical cell-cell interactions are sufficient to promote the formation of cell layers at topological defects, thereby seeding fruiting bodies in colonies of M. xanthus.

Pressure-driven changes to spontaneous flow in active nematic liquid crystals

.We consider the effects of a pressure gradient on the spontaneous flow of an active nematic liquid crystal in a channel, subject to planar anchoring and no-slip conditions on the boundaries of the channel. We employ a model based on the Ericksen-Leslie theory of nematics, with an additional active stress accounting for the activity of the fluid. By directly solving the flow equation, we consider an asymptotic solution for the director angle equation for large activity parameter values and predict the possible values of the director angle in the bulk of the channel. Through a numerical solution of the full nonlinear equations, we examine the effects of pressure on the branches of stable and unstable equilibria, some of which are disconnected from the no-flow state. In the absence of a pressure gradient, solutions are either symmetric or antisymmetric about the channel midpoint; these symmetries are changed by the pressure gradient. Considering the activity-pressure state space allows us to predict qualitatively the extent of each solution type and to show, for large enough pressure gradients, that a branch of non-trivial director angle solutions exists for all activity values.Graphical abstract

The lattice Hamiltonian modeling for active nematic liquid crystals with disclinations and oriented surfaces

We study the system of colloid active nematic liquid crystals evolving with transformation of the structure of surface linear topological defects. We estimate numerically the SO(2) Hamiltonian of the BKT model for disclinations on the surfaces connected with flows, which can participate in the defect ordering transitions.

Geometrical Control of Active Turbulence in Curved Topographies.

We investigate the turbulent dynamics of a two-dimensional active nematic liquid crystal constrained to a curved surface. Using a combination of hydrodynamic and particle-based simulations, we demonstrate that the fundamental structural features of the fluid, such as the topological charge density, the defect number density, the nematic order parameter, and defect creation and annihilation rates, are approximately linear functions of the substrate Gaussian curvature, which then acts as a control parameter for the chaotic flow. Our theoretical predictions are then compared with experiments on microtubule-kinesin suspensions confined on toroidal droplets, finding excellent qualitative agreement.

Theory of defect motion in 2D passive and active nematic liquid crystals.

The motion of topological defects is an important feature of the dynamics of all liquid crystals, and is especially conspicuous in active liquid crystals. Understanding defect motion is a challenging theoretical problem, because the dynamics of orientational order is coupled with backflow of the fluid, and because a liquid crystal has several distinct viscosity coefficients. Here, we suggest a coarse-grained, variational approach, which describes the motion of defects as effective "particles". For passive liquid crystals, the theory shows how the drag depends on defect orientation, and shows the coupling between translational and rotational motion. For active liquid crystals, the theory provides an alternative way to describe motion induced by the activity coefficient.
