A ripple effect? The impact of obesity on sperm quality and function.

Infertility affects approximately 15% of couples trying to conceive. Male-related causes account for roughly 50% of cases, with obesity emerging as a possible significant factor. Obesity, defined as a body mass index of 30.0 or higher, has become a widespread epidemic associated with numerous health issues, including a decrease of fertility. This review discusses the relationship between obesity and male infertility, particularly focusing on sperm quality and function. An overview of the literature suggests that obesity may influence the male reproductive system via disruptions in hormonal profiles, oxidative stress, and inflammation, leading to changes in sperm parameters. Several studies have discussed if obesity causes a decrease in sperm concentration, motility, and normal morphology, so far without a consensus being reached. However, available evidence suggests an impairment of sperm function in obese men, due to an increase in DNA damage and oxidative stress, impaired mitochondrial function and acrosome reaction in response to progesterone. Finally, the relationship between obesity and assisted reproductive technologies outcomes remains debatable, with conflicting evidence regarding the influence on fertilisation, pregnancy, and live birth rates. Therefore, the actual impact of obesity on human spermatozoa still needs to be clarified, due to the multiple factors potentially in play.

Use of a mitochondrial-specific live sperm stain to confirm penetration of equine oocytes after in vitro fertilization

Intracytoplasmic sperm injection (ICSI) is an important research and clinical tool. However, work with standard in vitro fertilization (IVF) is needed to explore sperm capacitation and the early events of fertilization. Repeatable methods for equine IVF have not yet been reported. Definitive evidence of fertilization, such as presence of the sperm tail within the cytoplasm, is critical when assessing potential methods for IVF. We have previously stained live sperm with the mitochondrial-specific stain, MitoTracker, and successfully visualized the sperm tail in the oocyte cytoplasm after ICSI. In the present study, we evaluated whether staining sperm with MitoTracker would A) be compatible with maintenance of sperm motility in culture and B) allow visualization of sperm tails within the COC or oocyte after co-culture with sperm. Semen was collected from one fertile stallion, and 200 µl of extended semen was layered under 1 mL of MOPS-buffered medium containing 10 or 100 nM Mitotracker Red CMXRos, in a 5-mL tube. The tube was incubated at a 45 o angle at 38 °C for 20 min. The uppermost 750 µL of the medium was collected, washed, centrifuged, and the pellet resuspended in modified TALP. Sperm were added to 50-µL droplets of modified TALP to a concentration of 1 million sperm/mL and these were incubated in 5% CO 2 in air. Sperm motility was assessed visually after 1, 6 and 22 h in culture. At 22 h, sperm were removed, counterstained with DAPI, and evaluated under fluorescence microscopy. In some replicates, in vitro-maturedequine cumulus-oocyte complexes (COCs) were added to the sperm droplets and co-incubated for 1 to 9 h. To assess sperm progress toward oocyte penetration, oocytes were denuded, fixed, and counterstained with DAPI. Some COCs were stained intact for confocal assessment of chromatin, sperm mitochondria, and acrosome status. These were fixed, permeabilized with 0.1% Triton-X 100, labeled with fluorescein-conjugated peanut agglutinin, then stained with Hoechst 33342. A minimum of three replicates were performed with each MitoTracker concentration. MitoTracker-stained sperm showed progressive motility after 1, 6 and 22 h incubation (∼80%, 50%, and 15% respectively) but at 22 h appeared to have a higher proportion of immotile sperm, and less vigorous motility, than did unstained sperm (∼25% progressive at 22 h). Staining with 10 nM MitoTracker supported more vigorous motility than did 100 nM. Both concentrations enabled clear visualization of the sperm tail on fluorescence microscopy. In penetrated oocytes, the sperm tail was clearly visualized in the oocyte cytoplasm, associated with sperm chromatin. On confocal microscopy, acrosomal ghosts were visualized at the periphery of the zona pellucida, indicating that sperm underwent the acrosome reaction in response to the zona. We conclude that staining of live sperm with 10 nM MitoTracker during swim-up provides a simple method to allow confirmation of sperm penetration of oocytes after sperm-oocyte coincubation.

ACRBP (Sp32) is involved in priming sperm for the acrosome reaction and the binding of sperm to the zona pellucida in a porcine model.

In boar sperm, we have previously shown that capacitation is associated with the appearance of the p32 tyrosine phosphoprotein complex. The principal tyrosine phosphoprotein involved in this complex is the acrosin-binding protein (ACRBP), which regulates the autoconversion of proacrosin to intermediate forms of acrosin in both boar and mouse sperm. However, the complete biological role of ACRBP has not yet been elucidated. In this study, we tested the hypothesis that tyrosine phophorylation and the presence of the ACRBP in the sperm head are largely necessary to induce capacitation, the acrosome reaction (AR) and sperm-zona pellucida (ZP) binding, all of which are necessary steps for fertilization. In vitro fertilization (IVF) was performed using matured porcine oocytes and pre-capacitated boar sperm cultured with anti-phosphotyrosine antibodies or antibodies against ACRBP. Anti-ACRBP antibodies reduced capacitation and spontaneous AR (P<0.05). Sperm-ZP binding declined in the presence of anti-phosphotyrosine or anti-ACRBP antibodies. The localisation of anti-ACRBP antibodies on the sperm head, reduced the ability of the sperm to undergo the AR in response to solubilized ZP or by inhibiting the sarco/endoplasmic reticulum Ca2+-ATPase. These results support our hypothesis that tyrosine phosphorylated proteins and ACRBP are present upon the sperm surface in order to participate in sperm-ZP binding, and that ACRBP upon the surface of the sperm head facilitates capacitation and the AR in the porcine.

The Calcium-Sensing Receptor Is Essential for Calcium and Bicarbonate Sensitivity in Human Spermatozoa.

The calcium-sensing receptor (CaSR) is essential to maintain a stable calcium concentration in serum. Spermatozoa are exposed to immense changes in concentrations of CaSR ligands such as calcium, magnesium, and spermine during epididymal maturation, in the ejaculate, and in the female reproductive environment. However, the role of CaSR in human spermatozoa is unknown. This work aimed to investigate the role of CaSR in human spermatozoa. We identified CaSR in human spermatozoa and characterized the response to CaSR agonists on intracellular calcium, acrosome reaction, and 3',5'-cyclic adenosine 5'-monophosphate (cAMP) in spermatozoa from men with either loss-of-function or gain-of-function mutations in CASR and healthy donors. CaSR is expressed in human spermatozoa and is essential for sensing extracellular free ionized calcium (Ca2+) and Mg2+. Activators of CaSR augmented the effect of sperm-activating signals such as the response to HCO3- and the acrosome reaction, whereas spermatozoa from men with a loss-of-function mutation in CASR had a diminished response to HCO3-, lower progesterone-mediated calcium influx, and were less likely to undergo the acrosome reaction in response to progesterone or Ca2+. CaSR activation increased cAMP through soluble adenylyl cyclase (sAC) activity and increased calcium influx through CatSper. Moreover, external Ca2+ or Mg2+ was indispensable for HCO3- activation of sAC. Two male patients with a CASR loss-of-function mutation in exon 3 presented with normal sperm counts and motility, whereas a patient with a loss-of-function mutation in exon 7 had low sperm count, motility, and morphology. CaSR is important for the sensing of Ca2+, Mg2+, and HCO3- in spermatozoa, and loss-of-function may impair male sperm function.

N-linked mannose glycoconjugates on shrimp thrombospondin, pmTSP-II, and their involvement in the sperm acrosome reaction.

Glycoconjugates in egg extracellular matrices are known to serve several functions in reproductive processes. Here, the presence of N-linked mannose (Man) glycoconjugates on shrimp thrombospondin ( pmTSP-II) and their physiological functions were investigated in the black tiger shrimp Penaeus monodon. A molecular analysis of pmTSP-II demonstrated anchorage sites for N-linked glycans in both the chitin-binding and TSP3 domains. The presence of Man residues was verified by concanavalin A lectin histochemistry on the purified fraction of pmTSP-II (250 kDa with protease inhibitor).The function of the Man glycoconjugates was evident by the Con A interference with the pmTSP-II-induced acrosome reaction (AR) as well as by the ability to recover the induction of the AR by the inclusion of Mans in the treatment mixture. In addition, the recombinant proteins of the three signature pmTSP-II domains expressed in E. coli (lacking glycosylation) and mannosidase-treated pmTSP-II showed a minimal ability to initiate the AR response. Together, these results provide evidence of the pivotal role that Man-linked pmTSP-II plays in modulating the shrimp sperm AR, a novel role for a TSP family protein in shrimp reproductive biology.

Seasonal variations in sperm acrosome reaction, osmotic tolerance and serum testosterone concentrations in rams

The study was conducted to evaluate the seasonal dynamics in the sperm acrosome reaction (AR), osmotic tolerance and serum testosterone concentration in rams. Blood and semen samples were collected from six mature rams during the winter, spring, summer and autumn seasons. The AR in sperm samples was induced by using lysophosphatidylcholine (LPC) and calcium ionophore (CI). The AR was monitored at various incubation times (15, 30, 60, 120 and 180 min) post-induction. To determine the osmotic tolerance through hypo-osmotic swelling test (HOST) the sperm were exposed to different osmotic conditions (50, 100, 290, 500 and 1000 mOsm/L). The serum testosterone concentration was assessed using a solid phase enzyme-linked immunosorbent assay (ELISA). The proportion of acrosome reacted sperm was greater (P < 0.05) during the winter season in the LPC and control groups, whereas, it was greater (P < 0.05) in two different seasons (winter and summer) at all incubation time points (15–180 min) in the CI group. The live HOST +ve, total HOST +ve and total live sperm were least (P < 0.05) during the winter season. The greatest membrane integrity (P < 0.05) was in the autumn season. The peak (P < 0.05) serum testosterone concentration was during the summer season. In conclusion, the osmotic tolerance and sensitivity of ram sperm to undergo the AR in response to LPC and CI vary with season. The increased testosterone concentration in the summer season might be the reason for a greater membrane integrity during the subsequent autumn season in Kivircik ram sperm.

Modification of membrane cholesterol and desmosterol in chicken spermatozoa improves post-thaw survival and prevents impairment of sperm function after cryopreservation.

During cryopreservation, spermatozoa are subjected to cryodamage that leads to a decline in fertilisation ability. Due to the complex nature of this process, the initial trigger for cryodamage remains unknown. Recently, we demonstrated that cryopreservation induces early apoptotic changes characterised by phosphatidylserine (PS) translocation via sterol loss from the plasma membrane of chicken spermatozoa. This led us to hypothesise that sterol incorporation into membranes minimises cryodamage, thereby improving the quality of cryopreserved chicken spermatozoa. In the present study, treating spermatozoa with 1.5mgmL-1 cholesterol- and 3mgmL-1 desmosterol-loaded cyclodextrin (CLC and DLC respectively) increased post-thaw survival and motility. These effects appeared to be highly dependent the amount of sterol loaded into the spermatozoa. Localisation experiments confirmed the incorporation of exogenous cholesterol into the sperm head region. Detection of PS translocation showed that elevation of these sterols inhibited early apoptotic changes, thereby enhancing post-thaw survival. Furthermore, CLC and DLC treatment suppressed spontaneous acrosome reaction after cryopreservation, preserving the ability of spermatozoa to undergo acrosome reactions in response to physiological stimulation. These results demonstrate that loading sterols into chicken spermatozoa before cryopreservation enhances their quality by inhibiting early apoptotic changes and spontaneous acrosome reactions. The present study provides new mechanistic insight into cryodamage in chicken spermatozoa.

Human sperm Toll-like receptor 4 (TLR4) mediates acrosome reaction, oxidative stress markers, and sperm parameters in response to bacterial lipopolysaccharide in infertile men.

To study the role of Toll-like receptor 4 (TLR4) in human spermatozoa and to assess sperm parameters, oxidative stress markers, and acrosome reaction in response to the stimulation of TLR4 by its ligand, the lipopolysaccharide (LPS), as a major endotoxin of Gram-negative bacteria. Our study was carried out in 73 sperm samples from patients undergoing semen analysis for couple infertility investigations. The studied patients were divided into three groups: normozoospermic fertile patients (n=13), patients with abnormal and leukospermic semen (n=13), and patients with abnormal and non-leukospermic semen (n=47). TLR4 expression in human spermatozoa was initially analyzed by western blot. Sperm samples were incubated in the presence of LPS (200ng/ml) for 18h. Then, sperm motility and vitality were evaluated by microscopic observation and oxidative stress markers as malondialdehyde (MDA) and carbonyl groups (CG) were spectrophotometrically assessed in neat and selected sperm. A triple-stain technique was also performed to evaluate acrosome reaction in 15 sperm samples from infertile patients. TLR4 expression was confirmed in human spermatozoa with a molecular weight of 69kDa. In the normozoospermic group, no significant differences in sperm parameters and oxidative stress markers were shown after incubation with LPS in neat and selected sperms. Regarding samples from the non-leukospermic group, LPS reduced spermatozoa motility and vitality rates in selected sperm (P=0.003; P=0.004, respectively). A significant increase of MDA and CG levels was also detected (P=0.01; P=0.02, respectively). However, only the MDA levels were significantly increased (P=0.01) in neat LPS-stimulated sperm. The same results were shown within the leukospermic group. The comparison between the two groups, leukospermic and non-leukospermic, in selected sperms showed a more important LPS effect in the leukospermic group significantly on motility and MDA rates (P=0.006; P=0.009, respectively). Furthermore, a significant decrease in reacted spermatozoa rate was detected in response to LPS in selected sperm samples from infertile men (P=0.03). These findings indicate that human spermatozoa express TLR4 and respond to LPS stimulationwith alterations in viability, motility, and theacrosome reaction implicating reactive oxygen species (ROS) production in sperm samples from infertile patients.

Effects of Panax ginseng, zearalenol, and estradiol on sperm function

BackgroundEstrogen signaling pathways are modulated by exogenous factors. Panax ginseng exerts multiple activities in biological systems and is classified as an adaptogen. Zearalenol is a potent mycoestrogen that may be present in herbs and crops arising from contamination or endophytic association. The goal of this study was to investigate the impact of P. ginseng, zearalenol and estradiol in tests on spermatozoal function. MethodsThe affinity of these compounds for estrogen receptor (ER)—alpha and beta (ERα and ERβ)—was assessed in receptor binding assays. Functional tests on boar spermatozoa motility, movement and kinematic parameters were conducted using a computer-assisted sperm analyzer. Tests for capacitation, acrosome reaction (AR), and chromatin decondensation in spermatozoa were performed using microscopic analysis. ResultsZearalenol—but not estradiol (E2)- or ginseng-treated spermatozoa—decreased the percentage of overall, progressive, and rapid motile cells. Zearalenol also decreased spontaneous AR and increased chromatin decondensation. Ginseng decreased chromatin decondensation in response to calcium ionophore and decreased AR in response to progesterone (P4) and ionophore. ConclusionZearalenol has adverse effects on sperm motility and function by targeting multiple signaling cascades, including P4, E2, and calcium pathways. Ginseng protects against chromatin damage and thus may be beneficial to reproductive fitness.

Enhancement of trypsin-like enzymes by A23187 ionophore is crucial for sperm penetration through the egg vestment of the giant freshwater prawn.

We report the presence of trypsin-like enzymes preferring Boc-QAR-MCA substrate in sperm collected from different portions of male reproductive tracts of the giant freshwater prawn, Macrobrachium rosenbergii and compare enzyme activities before and after an A23187 calcium ionophore treatment. Fluorogenic enzyme assays revealed that testicular sperm lysates showed high trypsin-like enzyme activity but the activity was relatively low in vas deferens sperm lysates as well as in the live sperm. Upon sperm treatment with A23187, trypsin-like activity was greatly enhanced in distal vas deferens sperm. Substrate- and inhibitor-based localization studies indicated that the sperm trypsin-like enzymes were not of a soluble type but were rather of a membrane-borne type, localized at the anterior spike and upper part of the main body. Notable structural changes were also evident in A23187-induced sperm including extensive ruffling of the sperm membrane structure at the base of the main body thereby supporting the acrosome reaction response in this species. We further proved by substrate inhibition assays that the enhanced trypsin-like enzyme activity participates in sperm penetration through the vitelline envelope, a novel sperm-egg penetration mechanism that is unique in this species.

Redistribution of the intra-acrosomal EGFP before acrosomal exocytosis in mouse spermatozoa.

Mammalian spermatozoa must undergo complex physiological and morphological alterations within the female reproductive tract before they become fertilization competent. Two important alterations are capacitation and the acrosome reaction (AR), by which spermatozoa become capable of penetrating the zona pellucida (ZP) of the oocyte. Although various biochemical stimulants have been reported to induce the AR, the true physiological inducer in vivo remains to be identified. Previously, it has been reported that most fertilizing spermatozoa undergo the AR before contacting the ZP and that only a small fraction of in vitro-capacitated spermatozoa can penetrate the ZP. Therefore, it is important to identify which capacitating spermatozoa undergo the AR in response to potential AR inducers such as progesterone. Here we show that spermatozoa undergo a dynamic rearrangement of the acrosome during in vitro capacitation. This involves the rapid movement of an artificially introduced soluble component of the acrosome, enhanced green fluorescent protein (EGFP), from the acrosomal cap region to the equatorial segment (EQ) of the sperm head. Spermatozoa exhibiting the EQ pattern were more sensitive to progesterone than were those without it. We suggest that spermatozoa that are ready to undergo acrosomal exocytosis can be detected by real-time EGFP imaging. This offers a promising new method for identifying where spermatozoa undergo the AR in the female reproductive tract in vivo.

Metabolic syndrome-associated sperm alterations in an experimental rabbit model: Relation with metabolic profile, testis and epididymis gene expression and effect of tamoxifen treatment

The influence of metabolic syndrome (MetS) on sperm quality and function is debated. Using a well-established high fat diet (HFD) rabbit model resembling human MetS, including development of hypogonadism, we demonstrate that HFD decreased sperm motility, morphology and acrosome reaction in response to progesterone and increased sperm cholesterol content. All the above parameters were associated with most MetS features, its severity and plasma testosterone (T) at univariate analysis. After T adjustment, sperm morphology and motility retained a significant association, respectively, with mean arterial pressure and circulating cholesterol levels. MetS modified the expression of inflammatory and tissue remodelling genes in the testis and of aquaporins in the epididymis. In a multivariate analysis, sperm morphology resulted associated with testis expression of fibronectin and collagen type 1 genes, whereas motility with epididymis aquaporin 1 gene. Administration of tamoxifen, used in the treatment of idiopathic male infertility, to HFD rabbits partially restored motility, but further decreased morphology and increased spontaneous acrosome reaction, without restoring responsiveness to progesterone. Overall our results indicate that development of MetS produces detrimental effects on sperm quality and functionality by inducing metabolic disorders leading to alterations in testis and epididymis functions and evidence a role of hypertension as a new determinant of abnormal sperm morphology, in line with a previous human study from our group.

Progesterone significantly enhances the mobility of boar spermatozoa

Progesterone released from the cumulus cells of the oocyte causes a number of physiological responses in human sperm cells including hyperactivation, acrosome reaction and chemotaxis. We employed a validated sperm mobility assay, which involves measuring the ability of sperm to penetrate an inert cell separation solution over time, to assess the ability of progesterone to enhance the mobility of boar spermatozoa. Cells maximally penetrate the solution over 50 minutes. 100nM progesterone significantly (P = 0.01) increased the mobility of non-capacitated sperm cells causing a doubling in the rate at which the cells penetrated through the cell separation solution (control half maximal penetration rate (Km) = 18.0±2.2; +100nM progesterone Km = 8.8±0.8min). Similarly, capacitated cells penetrated at a rate (Km = 19.2±3.0 min) not significantly different from non-capacitated cells and 100nM progesterone also significantly increased the rate of penetration of capacitated cells (Km = 9.5±1.0 min, P<0.05). The T-type voltage gated calcium channel blocker mibefradil (30mM) significantly inhibited both the control and progesterone enhanced mobility in non-capacitated and capacitated sperm. Only capacitated cells showed a significant increase in the acrosome reaction in response to 100nM progesterone (control non-reacted = 75±4%, +100nM progesterone non- reacted = 47±10%). Western blot analysis confirmed that there was an increase in the total protein tyrosine phosphorylation levels in capacitated cells. In conclusion, we have demonstrated that 100nM progesterone accelerates the mobility of boar sperm cells through an inert cell separation solution in an extracellular calcium dependent manner.

Characterization of the thrombospondin (TSP)‐II gene in Penaeus monodon and a novel role of TSP‐like proteins in an induction of shrimp sperm acrosome reaction

We have recently shown that water-soluble materials from the egg extracellular cortical rods (wsCRs) exert the ability to induce the sperm acrosome reaction in Penaeus monodon. In this study, we further demonstrated that the thrombospondin protein family (TSP) existed in wsCRs, and that their mRNA transcripts were detected in developing oocytes as early as stage I. Full sequence analysis revealed that our pmTSP sequence was considerably different from the recently reported pmTSP in the 5' nonconserved region and in many TSP signature domains, hence, the name pmTSP-II was given to our variant. The transcripts of pmTSP-II were detected only in early developing oocytes (stage-I and -II) while TSP-like proteins were detected in all developing oocytes, particularly at the outer rim of cortical rods situated in the extracellular crypts of the mature, stage-IV oocytes. In addition, wsCRs contained anti-TSP-reactive proteins, suggesting that TSP-like proteins are dissolved in and are part of the egg water during spawning. The functional importance of TSP-like proteins was evident by the interference of a wsCR-induced acrosome reaction response with anti-TSP in a concentration-dependent manner. In summary, we found that pmTSP-II transcripts were present in the developing oocytes and pmTSP-II protein accumulated in cortical rods, which are partly secreted and thus solubilized to produce dissolved TSP-like proteins that participate in induction of the sperm acrosome reaction-a novel reproductive role for TSP protein family.

Cholesterol-loaded cyclodextrin inhibits premature acrosomal reactions in liquid-stored rabbit spermatozoa

The effect of pretreatment of rabbit sperm cells with different concentrations of cholesterol-loaded cyclodextrin (CLC) on the occurrence of premature acrosome reactions during 72 h of liquid storage was investigated in three successive experiments. The aim of the first experiment was to establish a liquid storage model to facilitate premature acrosome reactions in rabbit sperm cells and, therefore, examined the relative effects of different dilution rates (1:5, 1:25 or 1:50) and storage temperatures (4 °C or 35 °C) on the occurrence of premature acrosome reactions. Increasing both dilution rate (from 1:5 to 1:25; P < 0.05) and storage temperature (from 4 °C to 35 °C; P < 0.0001) significantly enhanced the percentage of sperm cells that underwent premature acrosome reactions during storage. Therefore, a constant dilution rate of 1:25 and storage temperature of 35 °C was employed for the rest of the study. The second experiment examined the effect of different CLC concentrations (0, 0.5, 1.0 and 3.0 mg per 120 × 10 6 spermatozoa) on the occurrence of premature acrosome reactions in sperm cells. CLC supplementation of the extender inhibited ( P < 0.001) premature acrosome reactions in sperm cells in a dose-dependent manner during 72 h of storage. In the third experiment, the ability of CLC-pretreated sperm cells to undergo acrosome reactions induced by lysophosphatidylcholine (LPC) was evaluated following 72 h of storage. A considerable proportion of sperm cells pretreated with CLC (between 68.7 and 91.8%) underwent the acrosome reaction in response to LPC following 72 h of liquid storage. However, the ability of the sperm cells to undergo the acrosome reaction varied with regards to the dose levels of CLC pretreatment ( P < 0.001). In conclusion, CLC supplementation prevents premature acrosome reactions in liquid-stored rabbit spermatozoa.

In vitro induction of the acrosome reaction in bull sperm and the relationship to field fertility using low-dose inseminations

The acrosome reaction (AR) is a prerequisite for normal sperm fertilizing capability and can be studied in vitro after induction by various agents. The efficacy of a sperm population to undergo the AR in vivo is expected to influence male fertilizing potential. During the past two decades, a number of attempts have been made to relate the in vitro–induced AR to field fertility in several species. However, to our knowledge, no studies have combined in vitro induction of the AR with the simultaneous detection of sperm viability and acrosomal status using a high-precision flow cytometric technique. Furthermore, large-scale fertility trials using low-dose inseminations are pending. In the current study, the relationship between field fertility and the in vitro–induced AR was investigated using three ejaculates from each of 195 bulls, 156 Holstein and 39 Jersey bulls (Bos taurus), participating in a progeny test program including low-dose inseminations. A range of insemination doses, varying from 2.0×106 to 15×106 sperm/dose, was obtained by a controlled dilution process applied to each ejaculate. Different insemination doses were distributed at random among 75,610 experimental first inseminations in 4721 herds and 208 artificial insemination (AI) technicians. Simultaneous detection of sperm viability and acrosomal status was achieved using a triple color flow cytometric technique. Sperm samples from the bulls displayed a wide range of ability to acrosome react in response to calcium ionophore A23187. Both reproducibility of the AR response after induction and relationship between ability to acrosome react and field fertility was highly dependent on the definition of AR inducibility. Six basic and six combined AR indices were assessed. The AR index expressing the fraction of acrosome reacted sperm in the live sperm population after induction by ionophore had the highest repeatability, best described the biological variation in the studied population, and yielded the best significant predictive values on field fertility among the 12 indices considered. Moreover, the ability of sperm to acrosome react appeared to be a noncompensable trait that affects fertility regardless of the number of sperm per insemination dose. The current results therefore indicate that this sperm parameter is important in the field and also may play a role in the IVF laboratory.

Zona pellucida mediated acrosome reaction and sperm morphology.

Sperm samples from 29 men randomly selected from the andrology laboratory, were used to evaluate acrosome reaction response to solubilized human zona pellucida. Capacitated sperm samples were exposed to a solution containing 2 zona pellucidae (ZP) per microl for 60 min, after which acrosomal status were recorded using a PSA-FITC technique. Controls included samples supplied by fertile sperm donors. After completion of acrosome reaction studies, patient samples were divided according to the percentage of morphologically normal spermatozoa. Three basic groups were identified, namely, fertile donors, teratozoospermic (normal sperm morphology 5-14%; n = 25) and severely teratozoospermic (normal sperm morphology < 4%; n = 4) groups. The mean percent normal sperm were 15.8 +/- 0.9, 10.4 +/- 0.7 and 2.7 +/- 0.7, respectively, for normozoospermic donors, teratozoospermic and severely teratozoospermic men. The mean percentage (+/-SE) ZP mediated acrosome reacted sperm among teratozoospermic and severely teratozoospermic cases was 25.8% +/- 0.9 and 19.0% +/- 0.9 (P = 0.001), compared to 36.8% +/- 0.9 for the donor controls. Results were analysed and expressed as correlations between sperm morphology and acrosomal response to human solubilized zona pellucida, spontaneous and calcium ionophore induced acrosome reaction. Predictive values for acrosome responsiveness were depicted with ROC curve analyses. Sperm morphology evaluated by strict criteria correlated positively and highly significantly with the responsiveness of the acrosome reaction (r = 0.91, P = 0.0001). At a morphology cut-off value of 4%, the ROC curve analysis showed sperm morphology to be highly predictive of zona pellucida induced acrosome responsiveness with a sensitivity of 100% and negative predictive value of 100%. Spontaneous and calcium ionophore induced acrosome reactions revealed no correlation with sperm morphology. It was concluded that (i) morphological features of human spermatozoa are indicative of specific functional characteristics; (ii) zona pellucida induction of the acrosome reaction is superior, as a predictor of sperm morphology, compared to calcium ionophore induced and spontaneous acrosome reactions.

Effects of pentoxifylline treatment before freezing on motility, viability and acrosome status of poor quality human spermatozoa cryopreserved by the liquid nitrogen vapor method

The objective of the present study was to investigate the effects of the direct addition of pentoxifylline (PF) to the ejaculates of men with poor sperm quality before freezing on post-thaw sperm motility, viability, acrosome integrity, and agonist-induced acrosome reaction. Semen specimens from 16 infertile men with impaired sperm count and motility (oligoasthenozoospermia) were divided into two equal aliquots: one received no treatment (control) while the other was incubated with 5 mM PF (treated). Both aliquots were cryopreserved by the liquid nitrogen vapor method. Motility was assessed according to WHO criteria. Acrosome integrity and spontaneous and calcium ionophore-induced acrosome reactions were assessed with fluorescein isothiocyanate-conjugated peanut agglutinin combined with a supra-vital dye (Hoechst-33258). Cryopreservation impaired sperm motility (percentage reduction: 87.4 (interquartile range, IQ: 70.3-92.9) vs 89.1 (IQ: 72.7-96.0%)), viability (25.9 (IQ: 22.2-29.7) vs 25.6 (IQ: 19.7-40.3%)) and acrosome integrity (18.9 (IQ: 5.4-38.9) vs 26.8 (IQ: 0.0-45.2%)) to the same extent in both treated and control aliquots. However, PF treatment before freezing improved the acrosome reaction to ionophore challenge test scores in cryopreserved spermatozoa (9.7 (IQ: 6.6-19.7) vs 4.8 (IQ: 0.5-6.8%); P = 0.002). These data show that pre-freeze treatment of poor quality human sperm with pentoxifylline did not improve post-thaw motility or viability nor did it prevent acrosomal loss during the freeze-thaw process. However, PF, as used, improved the ability of thawed spermatozoa to undergo the acrosome reaction in response to calcium ionophore. The present data indicate that treatment of poor quality human sperm with PF may enhance post-thaw sperm fertilizing ability.

Biochemical characterization and physiological role of cortical rods in black tiger shrimp, Penaeus monodon

Cortical rods (CRs), precursors of egg jelly investment in many penaeoid shrimp, are composed of different proportions of proteins and carbohydrates, the physiological role of which still requires extensive investigation. In this study, we demonstrated the biochemical properties of the CRs and their role in the induction of the acrosome reaction (AR). Profiles of the isolated CRs revealed a number of major protein bands ranging from 35 to 230 kDa. These CR proteins were extensively glycosylated and sulfated. Lectin-based carbohydrate analysis further revealed the highest reactivity of concanavalin A (Con A) among other lectins used. In addition, the selective interference of Con A binding with mannose but not glucose indicated that CR glycoproteins were of high-mannose type. Using immunoblotting with anti-CR antibody, we further demonstrated that part of egg water (EW, a natural AR inducer) was derived from miscible components of the CRs. Physiological tests of water-soluble CR (wsCR) revealed its high AR inducing competency comparable to that of EW, which was far superior to that of acid-urea treated CR (auCR). Furthermore, the wsCR-induced AR was selectively inhibited by Con A, suggesting the significance of the exposing mannose residues in regulating P. monodon sperm AR response.
