Effect of PDGF-B Gene-Activated Acellular Matrix and Mesenchymal Stem Cell Transplantation on Full Thickness Skin Burn Wound in Rat Model.

Full thickness burn wounds are lack of angiogenesis, cell migration, epithelialisation and finally scar tissue formation. Tissue engineered composite graft can provide sustained release of growth factor and promote the wound healing by cell migration, early angiogenesis and proliferation of extracellular matrix and wound remodeling. The objective of this study was to evaluate the gene embedded (pDNA-platelet-derived growth factor, PDGF-B) porcine acellular urinary bladder matrix with transfected mesenchymal stem cells (rBMSC) on healing of full thickness burn wound in rat model. Full thickness burn wound of 2 × 2cm size was created in dorsum of rat model under general anesthesia. Burn wounds were treated with silver sulfadiazine; porcine acellular urinary bladder matrix (PAUBM); PAUBM transfected with pDNA-PDGF-B; PAUBM seeded with rBMSC; PAUBM seeded with rBMSC transfected with pDNA-PDGF-B in groups A, B, C, D and E respectively. The wound healing was assessed based on clinical, macroscopically, immunologically, histopathological and RT-qPCR parameters. Wound was significantly healed in group E and group D with early extracellular matrix deposition, enhanced granulation tissue formation and early angiogenesis compared to all other groups. The immunologic response against porcine acellular matrix showed that PDGF-B gene activated matrix along with stem cell group showed less antibody titer against acellular matrix than other groups in all intervals. PDGF gene activated matrix releasing the PDGF-B and promote the healing of full thickness burn wound with neovascularization and neo tissue formation. PDGF gene also enhances secretion of other growth factors results in PDGF mediated regenerative activities. This was confirmed in RT-qPCR at various time intervals. Gene activated matrix encoded for PDGF-B protein transfected stem cells have been clinically proven for early acceleration of angiogenesis and tissue regeneration in burn wounds in rat models. Regeneration of lingual musculature in rats using myoblasts over porcine bladder acellular matrix.

To use tissue engineering muscle repair (TEMR) for regenerating the lingual musculature of hemiglossectomized rats using neonatal myoblasts (NM) on porcine acellular urinary bladder matrix (AUBM). The study used 80 male rats. A volumetric muscle loss (VML) injury was created on the left side of the tongue. The rats were randomized into four groups: Group 1 (AUBM+myoblasts); Group 2 (AUBM); Group 3 (myoblasts); and Group 4 (control). NM were obtained from neonatal rats. The animals were weighed on day 0 and just before euthanasia. Five rats in each group were euthanized at days 2, 14, 28, and 42; the tongues were prepared for morphometric analysis, postoperative left hemitongue weight, and immunohistochemical analysis (desmin, CD-31, and anti-neurofilament). The weight gain from greatest to least was as follows: AUBM+myoblasts>myoblasts>AUBM>control. The tongue dorsum occupied by VML, and difference in mg between control side and intervened side from least to great was as follows: AUBM+myoblasts<myoblasts<AUBM<control. The order from highest to lowest antibody positivity was as follows: AUBM+myoblasts>myoblasts>AUBM>control. The use of porcine AUBM and NM for the regeneration of lingual musculature was found to be an effective TEMR treatment for repairing tongue VML injury.

Evaluation of two biological matrices for repairing of ventral hernia in bucks

The aim of this study was to estimate the changes in ventral hernia repairing in Iraqi bucks by using two biological matrixes derived from bovine (pericardium and urinary bladder matrix) through histopathological examination. All bucks right lower flank awas prepared surgically, sedation were done by using (2% Xylazine hydrochloride) at a dose of 0.2mg/kg intramuscular, and surgical site anesthetized through an inverted (L) shape local infiltration technique using lidocaine hydrochloride (2%) at a dose of 8mg/kg. Ventral abdominal hernias were induced in (24) bucks through elliptical resection of abdominal muscles to made hernia ring (6-8cm) with avoiding peritoneum perforation. After 30 days of operation bucks were divided into two equal groups. cellular Bovine pericardium group and cellular urinary bladder matrix group. In two groups hernias were treated with only replacement of acellular bovine pericardium and cellular urinary bladder matrix respectively which fixed with interrupted horizontal mattress 2cm far from hernia ring by Polypropylene (No.1) suture material. Histopathological biopsies were taken at 2nd, 8th and 16th week post treatment (4 bucks\ period). Both groups’ successes in reconstruction of large hernia in bucks through prevent recurrent or others post-operative complications. In addition the histopathological examination showed that the acellular urinary bladder matrix superior than acellular bovine pericardium matrix in enhance healing based on acellular urinary bladder matrix role in augment early initiation of inflammatory cells infiltration, fibroblast proliferation and marked collagen deposition, in addition to its early degradation, incorporation and remodeling.

Effects of topical applications of porcine acellular urinary bladder matrix and Centella asiatica extract on oral wound healing in a rat model.

To evaluate the effects of topical applications of porcine acellular urinary bladder matrix (AUBM) and Centella asiatica extract (CAE) on the healing of tongue wounds in a rat model. Wounds were made in the tongue using a punch tool in 64 male Sprague-Dawley rats, randomized into four groups (n = 16 per group): group 1 (control), group 2 (CAE), group 3 (AUBM mixed with orabase), and group 4 (orabase). No product was applied in group 1 and groups 2-4 received three daily topical applications. The animals were weighed on day 0 and at the time of euthanasia. Four rats in each group were euthanized at days 2, 7, 14, and 21 and the tongues were processed for: macroscopic morphometric analysis, myeloperoxidase (MPO) and malondialdehyde (MDA) levels, histological wound repair (degree of reepithelialization and inflammation), and CD31 positivity. The animals' weight gain, histological wound repair, and CD31 positivity from greatest to least were: AUBM > CAE > orabase > control. Percentage of tongue occupied by wound, MPO, and MPA levels from least to greatest were: AUBM < CAE < orabase < control, whereby the AUBM group showed significant differences (p ≤ 0.05) in comparison with the other groups on days 2, 7, 14, and 21 for percentage of tongue occupied by wound and MDA and on days 7, 14, and 21 for MPO. CAE is effective for oral tissue regeneration, while AUBM is an even more potent means of oral mucosa regeneration. AUBM may be beneficial to patients with oral wounds; this finding requires further clinical and laboratory investigation.

Co-delivery of micronized urinary bladder matrix damps regenerative capacity of minced muscle grafts in the treatment of volumetric muscle loss injuries.

Minced muscle grafts (MG) promote de novo muscle fiber regeneration and neuromuscular strength recovery in small and large animal models of volumetric muscle loss. The most noteworthy limitation of this approach is its reliance on a finite supply of donor tissue. To address this shortcoming, this study sought to evaluate micronized acellular urinary bladder matrix (UBM) as a scaffolding to promote in vivo expansion of this MG therapy in a rat model. Rats received volumetric muscle loss injuries to the tibialis anterior muscle of their left hind limb which were either left untreated or repaired with minced muscle graft at dosages of 50% and 100% of the defect mass, urinary bladder matrix in isolation, or a with an expansion product consisting of a combination of the two putative therapies in which the minced graft is delivered at a dosage of 50% of the defect mass. Rats survived to 2 and 8 weeks post injury before functional (in vivo neuromuscular strength), histological, morphological, and biochemical analyses were performed. Rats treated with the expansion product exhibited improved neuromuscular function relative to untreated VML after an 8 week time period following injury. This improvement in functional capacity, however, was accompanied with a concomitant reduction in graft mediated regeneration, as evidenced cell lineage tracing enable by a transgenic GFP expressing donor, and a mixed histological outcome indicating coincident fibrous matrix deposition with interspersed islands of nascent muscle fibers. Furthermore, quantitative immunofluorescence and transcriptional analysis following the 2 week time point suggests an exacerbated immune response to the UBM as a possible nidus for the observed suboptimal regenerative outcome. Moving forward, efforts related to the development of a MG expansion product should carefully consider the effects of the host immune response to candidate biomaterials in order to avoid undesirable dysregulation of pro-regenerative cross talk between the immune system and myogenic processes.

The Use of Acellular Urinary Bladder Matrix as Coverage for Fasciocutaneous Free Flap Donor Sites: An Alternative to Traditional Grafting Procedures

The purpose of this review is to 1) evaluate our early experience with urinary bladder matrix as a dressing for the management of complex wounds subsequent to fasciocutaneous or osteocutaneous vascularized tissue harvests, 2) assess coverage of exposed tendons and the duration of granulation and epithelial coverage, and 3) assess esthetic and functional outcomes compared with traditional means for the management of vascularized tissue harvest sites. This prospective study consisted of 8 consecutive patients in whom a substantial cutaneous component was harvested as part of a vascularized tissue reconstruction, which did not permit for primary closure (n=7) or had skin graft failure (exposed tendon) that required additional treatment (n=1). Functional and esthetic outcomes were comparable to those of traditional methods of donor site closure as determined by clinical evaluation and subjective assessment by the patient. In all patients, no additional grafting was required to achieve full coverage. Irregularities of the wound achieved a level plane with granulation tissue an average of 3 to 4weeks after surgery, followed by an additional 5 to 8weeks for epithelialization. Wound care was minimal and included twice-daily wet-to-dry dressings. It was noted that complete wound healing was delayed in the urinary bladder matrix group, which required more attention during wound care when compared with published data. Advantages included facilitation of a robust granulation layer that leveled wound irregularities and avoidance of an additional skin graft site. The use of urinary bladder matrix is a viable option for the rehabilitation of donor sites of vascularized tissue that include skin. Benefits include the avoidance of an additional skin graft donor site, facilitation of epithelialization over exposed tendons, leveling of the donor site texture, and an equivalent esthetic result compared with current practices for wound coverage. The success of this technique may be limited by the cost of materials and the protracted course for wound epithelialization.

Acellular Urinary Bladder Matrix in a Collagenase Model of Superficial Digital Flexor Tendonitis in Horses

The objective of the present study was to determine the efficacy of urinary bladder matrix (UBM) in collagenase-induced superficial digital flexor (SDF) tendonitis by using clinical, ultrasonographic, and histologic data. A total of eight healthy adult horses were used in this study. Bilateral forelimb SDF tendonitis was created in the horses by injecting collagenase. After 14 days, one randomly selected forelimb SDF tendon was blindly treated with UBM and the opposite tendon was treated with a control (saline). Clinical and ultrasonographic parameters including lameness, lesion size, ultrasonographic fiber pattern, and echogenicity were measured throughout the study. After 84 days, horses were euthanized and SDF tendon lesions from the two groups were compared statistically using an analysis of variance with significance set at P ≤ .05.Results showed that there were no significant differences between the treated and control tendons for any of the clinical, ultrasonographic, gross, or histologic variables. UBM does not appear to be an effective treatment for collagenase-induced SDF tendonitis. However, there may be differences in clinical tendonitis that might render the treatment more effective in the clinical setting.
