The redox status of salinity-stressed Chenopodium quinoa under salicylic acid and sodium nitroprusside treatments.

Spreading the cultivation of crops with high nutritional values such as quinoa demands a wide area of research to overcome the adverse effects of environmental stress. This study aimed at investigating the role of salicylic acid (SA) and sodium nitroprusside (SNP) as a nitric oxide donor, priming at improving the antioxidant defense systems in boosting salinity tolerance in Chenopodium quinoa. These two treatments, SA (0.1 mM) and SNP (0.2 mM), individually or in combination, significantly improved the function of both enzymatic and non-enzymatic antioxidants. SA and SNP priming significantly reduced superoxide dismutase activity, which was accompanied by a significant decrease in hydrogen peroxide accumulation under salinity stress (100 mM NaCl). The SA and SNP treatment increased the activity of enzymatic antioxidants (e.g., catalase, ascorbate peroxidase, peroxidase, and glutathione reductase) and the accumulation of non-enzymatic antioxidants (e.g. ascorbate-glutathione pools, α-tocopherol, phenols, flavonoids, anthocyanins, and carotenoids) to suppress the oxidative stress induced by salinity stress. Under SA and SNP treatment, the upregulation of antioxidant mechanisms induced a significant increase in chlorophyll florescence, chlorophylls, carotenoids, and proteins, as well as a significant reduction in the malondialdehyde content in salinity-stressed plants. In addition, the foliar application of SA or/and SNP led to a significant increase in the accumulation of osmoprotectant molecules of sugars and proline to overcome osmotic stress induced by salinity stress. In conclusion, SA and SNP priming can effectively combat salinity stress through improving the redox status of plants.

Molybdenum-induced endogenous nitric oxide (NO) signaling coordinately enhances resilience through chlorophyll metabolism, osmolyte accumulation and antioxidant system in arsenate stressed-wheat (Triticum aestivum L.) seedlings

There is little information available to decipher the interaction between molybdenum (Mo) and nitric oxide (NO) in mitigating arsenic (AsV) stress in plants. The present work highlights the associative role of exogenous Mo and endogenous NO signaling in regulating AsV tolerance in wheat seedlings. Application of Mo (1 μM) on 25-day-old wheat seedlings grown in the presence (5 μM) or absence of AsV stress caused improvement of photosynthetic pigment metabolism, reduction of electrolytic leakage and reactive oxygen species (ROS), and higher accumulation of osmolytes (proline and total soluble sugars). The molybdenum treatment upregulated antioxidative enzymes, such as superoxide dismutase, ascorbate peroxidase and glutathione reductase. In addition, the accumulation of nonenzymatic antioxidants (ascorbate and glutathione) was correlated with an increase in ascorbate peroxidase and glutathione reductase activity. The application of cPTIO (endogenous NO scavenger; 100 μM) reversed the Mo-mediated effects, thus indicating that endogenous NO may accompany Mo-induced mitigation of AsV stress. Mo treatment stimulated the accumulation of endogenous NO in the presence of AsV stress. Thus, it is evident that Mo and NO-mediated AsV stress tolerance in wheat seedlings are primarily operative through chlorophyll restoration, osmolytes accumulation, reduced electrolytic leakage, and ROS homeostasis.

Overexpression of AtDREB1 and BcZAT12 genes confers drought tolerance by reducing oxidative stress in double transgenic tomato (Solanum lycopersicum L.).

Double transgenic tomato developed by AtDREB1A and BcZAT12 genes pyramiding showed significant drought tolerance by reducing oxidative stress with enhanced yield. Although a large number of efforts have been made by different researchers to develop abiotic stress tolerance tomato for improving yield using single gene, however, no reports are available which targets AtDREB1 and BcZAT12 genes together. Hence, in the present study, double transgenic plants were developed using AtDREB1 and BcZAT12 genes to improve yield potential with better drought tolerance. Double transgenic (DZ1-DZ5) tomato lines showed enhanced drought tolerance than their counterpart non-transgenic and single transgenic plants at 0, 07, 14, and 21days of water deficit, respectively. Double transgenic plants showed increased activity of antioxidant enzymes, like catalase (CAT), superoxide dismutase (SOD), glutathione reductase (GR), ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR), monodehydroascorbate reductase (MDHAR) and guaiacol peroxidase (POD), and accumulation of non-enzymatic antioxidants like ascorbic acid, glutathione as compared to non-transgenic and single transgenic. Additionally, the transcript analysis of antioxidant enzymes revealed the increased level of gene expression in double transgenic tomato lines. Developed double-transgenic tomato plants co-over-expressing both genes exhibited more enzymatic and non-enzymatic anti-oxidative activities as compared to the non-transgenic and single transgenic control, respectively. This is the preliminary report in tomato, which forms the basis for a multigene transgenic approach to cope with drought stress.

Effects of tropospheric ozone on groundnut (Arachis hypogea L.) cultivars: Role of plant age and antioxidative potential

Ozone (O3) is a secondary atmospheric pollutant present in the tropospheric environment. Ozone enters in internal tissue of leaf through stomata and produces reactive oxygen species (ROS) which damages cellular membrane, alter gene expression and metabolic activities and ultimately caused death of the plant tissues. Certain defensive mechanism occurs in the internal part of leaves to eliminate generated ROS. However, there is significant change in the levels and kind of defence traits as plants develop from the seedling to juvenile to mature and senescent stages. A field study was conducted for two consecutive seasons with five cultivars of groundnut (Arachis hypogea L cv. TG-37A, TPG-41, TAG-24, GG-20, and Dh-86) to assess the intra-specific variation in defensive response against ozone at various plant developmental stage. Results of the study showed that ozone significantly affected biomass and primary metabolites, generate ROS and increases membrane permeability in all the groundnut cultivars. In response to ROS, groundnut cultivars produced non-enzymatic and enzymatic antioxidants. Study found that the severity of the effect varied among the cultivars and by developmental stages. Cultivar Dh-86 showed higher antioxidative activity and least biomass sensitivity to ozone. Although accumulation of non-enzymatic antioxidants was present to provided defence against ozone, activity of enzymatic antioxidants varied at different growth stages and corresponds to the biomass sensitivity. Study concluded that the ozone phytotoxicity is dependent on the antioxidative potential of the cultivars and the growth stage of the plant. Further, the activity of APX played a measure role for age dependent plant sensitivity in groundnut.

Low UVA intensity during cultivation improves the lettuce shelf-life, an effect that is not sustained at higher intensity

• The cultivation UVA level affects lettuce quality during shelf-life. • Low UVA intensity postpones the decrease of the primary energy reserves. • Low UVA intensity delays the decline of chlorophyll content, and decreases PPO activity. • Low UVA intensity reduces the ROS levels, owing to enhanced antioxidant activity. • The high UVA intensity mostly behaves comparably to the control. Sunlight includes UVA (320–400 nm). Since it is irrelevant for driving photosynthesis, less recognition is given to its usefulness in indoor cultivation. We examined how the cultivation under different UVA levels (0, 10 and 40 μmol m −2 s −1 ) affects the lettuce postharvest quality by evaluating the temporal dynamics of several underlying aspects, including weight loss, chlorophyll fluorescence, protein content, primary energy reserves (starch, sugars), pigments (chlorophyll, carotenoids), the activity of enzymes related to enzymatic browning [phenylalanine ammonia lyase, polyphenol oxidase (PPO)], the activity of antioxidant machinery enzymes (superoxide dismutase, catalase), the accumulation of non-enzymatic antioxidants (polyphenols, ascorbic acid) and the accumulation of reactive oxygen species (ROS; H 2 O 2 , O 2 − ). Evaluations were conducted (5 d intervals) on dark-stored excised leaves for 15 d (16 ℃, 70 % relative air humidity). Control samples gradually underwent a decrease in both primary energy reserves and protein content. They also suffered from color degradation owing to both a decrease in chlorophyll content and an enhanced PPO activity. Increased electrolyte leakage was also observed as a result of enhanced ROS levels. The low UVA level clearly improved all quality parameters, whereas the application of high UVA intensity caused limited effects in comparison to the control. In the former case, ROS accumulation was counteracted by an enhanced stimulation of the non-enzymatic and enzymatic antioxidant machinery. Overall, the results show that the promotive effect of UVA during cultivation on lettuce nutritional quality and shelf-life is strongly intensity-dependent and is mediated by diverse processes.

Differential physiological and biochemical responses of Quercus infectoria and Q. libani to drought and charcoal disease.

The vast oak-dominated forests of the Zagros Mountains in southwestern Iran currently undergo large-scale dieback driven by a combination of drought and increasing incidence of charcoal disease caused by the fungal pathogens Biscogniauxia mediterranea and Obolarina persica. Here, we explore the interactive effects between drought and charcoal disease agents on the physiology and biochemistry of Quercus infectoria and Quercus libani seedlings. The combination of pathogen attack and water limitation hampered plant development, especially in Q. libani seedlings, negatively affecting growth, biomass production, photosynthetic efficiency, and leaf water potential. An increase in markers of oxidative damage together with the upregulation of the antioxidant defense revealed that drought stress and pathogen infection led to pro-oxidative conditions in both oak species, especially in Q. libani, where larger changes in malondialdehyde and hydrogen peroxide occurred. The upregulation of the antioxidant system was more prominent in Q. infectoria than in Q. libani, resulting in enhanced enzyme activity and accumulation of non-enzymatic antioxidants. Fungal infection stimulated the activity of chitinase, phenylalanine ammonia lyase and β-1,3-glucanase in Q. infectoria leaves and this response became more pronounced under water shortage. Our study highlights that drought stress greatly intensifies the effects of the charcoal disease. Moreover, our findings imply superior stress resistance of Q. infectoria conferred by a highly efficient antioxidant system, strong osmotic adjustment (through proline), and increases in resistance enzymes and secondary metabolites (phenols and flavonoids). Future investigations should focus on adult trees in their natural habitat including interactions with soil factors and other pathogens like nematodes, bacteria and other fungi. Because the present research was conducted on oak seedlings, the findings can be considered by forest nursery managers.

Investigating the effect of methyl jasmonate and melatonin on resistance of Malus crabapple 'Hong Jiu' to ozone stress.

Ozone (O3) is an adverse environmental factor posing damage to ornamental plants. Thus, it is important to seek an effective way of enhancing plant tolerance to O3-induced damage. Methyl jasmonate (MJ) and melatonin (MT) are plant growth regulators (PGRs) involved in plant abiotic stress responses. In this study, compared with the control group of plants without ozone, the influence of exogenous MJ (0, 10, 50, 100, and 150μM) and MT (0, 0.1, 0.5, 2.5, and 12.5μM) on the resistance of Malus crabapple 'Hong Jiu' was evaluated under O3 stress (100 ± 10nL/L for 3h). Our data revealed that levels of MDA were significantly enhanced following O3 treatment compared with plants without O3. O3 induced the activities of antioxidant enzymes and the accumulation of non-enzymatic antioxidants. While lower malondialdehyde (MDA) content, greater activities of antioxidant enzymes, and higher levels of soluble protein and non-enzymatic antioxidants were observed in PGRs-pretreated plants than in non-PGRs-pretreated plants under O3 stress. Based on the above results and air pollution tolerance index (APTI), an exogenous supply of MJ and MT to Malus crabapple 'Hong Jiu' seedlings was protective for O3-induced toxicity. The present study provides new insights into the mechanisms of MJ and MT amelioration of O3-induced oxidative stress damages in Malus crabapple 'Hong Jiu.'

Nitric oxide and abscisic acid protects against PEG-induced drought stress differentially in Brassica genotypes by combining the role of stress modulators, markers and antioxidants

The present study was designed to see the effect of exogenous nitric oxide (NO) and abscisic acid (ABA) and their interaction on physiological and biochemical activities in leaves and roots of two Indian mustard (Brassica juncea) cultivars [cv. Pusa Jagannath (PJN) and Varuna (VAR)] exposed to polyethylene glycol (PEG)-induced drought stress. Seven days old hydroponically grown seedlings were treated with PEG (10%), sodium nitroprusside, a NO donor [NO (100 μM)] and abscisic acid [ABA (10 μM)], using different combinations as: Control, ABA, NO, PEG, PEG + ABA, PEG + NO, and PEG + NO + ABA. Results revealed that in response to PEG-induced drought stress leaf relative water content, chlorophyll, carotenoid and protein content decreased with increased production of O2−●, MDA, H2O2, cysteine content and non-enzymatic antioxidants (including proline, flavonoid, phenolic, anthocyanin, and ascorbic acid), whereas, the enzymatic antioxidants (including SOD, CAT, APX, GR) showed the response range from no effect to increase or decrease in certain enzymes in both Brassica cultivars. The application of NO or/and ABA in PEG-stressed cultivars showed that both enzymatic and non-enzymatic antioxidants responded differently to attenuate oxidative stress in leaves and roots of both cultivars. Overall, PJN had the antioxidant protection mainly through the accumulation of non-enzymatic antioxidants, whereas VAR showed tolerance by the enhancement of both enzymatic and non-enzymatic antioxidant activities. Altogether, the study concluded that the independent NO and its interaction with ABA (PEG + NO and PEG + NO + ABA) were much effective than independent ABA (PEG + ABA) in lowering PEG-drought stress in Brassica cultivars.

Antioxidant potential of ragweeds: Ambrosia artemisiifolia, A. trifida and Iva xanthifolia

The purpose of this study was to analyze antioxidant systems among three invasive ragweed species, Ambrosia artemisiifolia L., A. trifida L. and Iva xanthifolia Nutt. Antioxidant capacity could be a possible marker of adaptation to variable environmen?tal conditions, since change in amount of antioxidants represents one of the first responses to various environmental stimuli. Among investigated ragweeds, I. xanthifolia leaves had more pronounced guaiacol peroxidase activity (87.5 and 62.5%) and reduced glutathione content (2.3 and 28.8%) than A. artemisiifolia and A. trifida, respectively. However, super?oxide dismutase activity was invariable in all investigated plants (234.1-247.5 U g-1 fresh weight). The highest content of total phenolics, tannins, flavonoids and proanthocyanidins were detected in A. trifida leaves (up to 3.7 - fold the amount of the others). According to antioxidant activity tests, investigated ragweed species could be presented in a scale: A. trifida > I. xanthifolia > A. artemisiifolia. Accumulation of non-enzymatic antioxidants and lower content of reduced glutathione point to different oxidative stress avoidance strategies of A. trifida when compared to A. artemisiifolia and I. xanthifolia within the same environ?mental conditions.

Morpho-physiological and biochemical responses of muskmelon genotypes to different degree of water deficit

Morpho-physiological and biochemical analyses were carried out in eight diverse indigenous muskmelon (Cucumis melo L.) genotypes exposed to different degrees of water deficit (WD). The ability of genotypes MM-7, and especially MM-6, to counteract better the negative effect of WD was associated with maintaining higher relative water content (RWC), photosynthetic rate, efficiency of PSII, and photosynthetic pigments compare to other genotypes. Furthermore, MM-6 showed a better ability to maintain cellular homeostasis than the others. It was indicated by a stimulated antioxidative defense system, i.e., higher activities of antioxidant enzymes, accumulation of nonenzymatic antioxidants together with lower concentration of reactive oxygen species and malondialdehyde. However, the genotypes MM-2 and MM-5 suffered greatly due to WD and showed reduced RWC, photosynthetic rates, pigment content, and exhibited higher oxidative stress observed as lower antioxidant enzyme activities.

Natural silicates mixed with organic fertilizers enhance corn adaptation to salt stress and improve physical characteristics of sandy soil

ABSTRACTSoil salinity is one of the major environmental constraints to crop productivity worldwide. Therefore, the development of cost-effective and environment-friendly techniques allowing increased crop productivity and soil fertility under saline conditions is rather urgent today. The objective of this investigation was to study the effects of mixtures containing natural silicates (analcite, bergmeal, and potassium silicate) and organic fertilizers (sapropel, peat) in corn (Zea mays L.). We specifically evaluated tolerance of corn to salinity stress and certain characteristics of saline soil (viz., redox potential, conductivity, and phytotoxicity) using a factorial pot experiment, modeling NaCl salinity levels of 0, 50, 100, 150, and 200 mM under greenhouse conditions. Growth, water balance, photosynthesis, catalase activity, and accumulation of nonenzymatic antioxidants (flavonoids and anthocyanins) were measured and evaluated. Salinity stress reduced shoot and root biomass by 8–49%, photosynthetic pigment content in leaves by 15–30%, deteriorated water balance, and activated nonspecific adaptive reactions (i.e., accumulation of enzymatic and nonenzymatic antioxidants) in the corn seedlings. All the tested silicon-containing mixtures stimulated corn seedling resistance to salt stress and reduced soil phytotoxicity. This was reflected in the stimulation of growth of the corn seedlings (accumulation of shoot biomass, and formation and growth of lateral roots). The content of photosynthetic pigments, flavonoids, anthocyanins, catalase activity increased 1.3–2 times compared with plants that received NaCl only. The difference between treatments and control was most pronounced at moderate levels of salinity (100–150 mM). The mixture containing silicon minerals and sapropel (9:1 proportion) showed the highest protective effect against salinity stress.

Arbuscular mycorrhizal symbiosis modulates antioxidant response in salt-stressed Trigonella foenum-graecum plants

An experiment was conducted to evaluate the influence of Glomus intraradices colonization on the activity of antioxidant enzymes [superoxide dismutase (SOD), catalase (CAT), peroxidase (PX), ascorbate peroxidase (APX), and glutathione reductase (GR)] and the accumulation of nonenzymatic antioxidants (ascorbic acid, α-tocopherol, glutathione, and carotenoids) in roots and leaves of fenugreek plants subjected to varying degrees of salinity (0, 50, 100, and 200mM NaCl) at two time intervals (1 and 14days after saline treatment, DAT). The antioxidative capacity was correlated with oxidative damage in the same tissue. Under salt stress, lipid peroxidation and H2O2 concentration increased with increasing severity and duration of salt stress (DoS). However, the extent of oxidative damage in mycorrhizal plants was less compared to nonmycorrhizal plants. The study reveals that mycorrhiza-mediated attenuation of oxidative stress in fenugreek plants is due to enhanced activity of antioxidant enzymes and higher concentrations of antioxidant molecules. However, the significant effect of G. intraradices colonization on individual antioxidant molecules and enzymes varied with plant tissue, salinity level, and DoS. The significant effect of G. intraradices colonization on antioxidative enzymes was more evident at 1DAT in both leaves and roots, while the concentrations of antioxidant molecules were significantly influenced at 14DAT. It is proposed that AM symbiosis can improve antioxidative defense systems of plants through higher SOD activity in M plants, facilitating rapid dismutation of O2 (-) to H2O2, and subsequent prevention of H2O2 build-up by higher activities of CAT, APX, and PX. The potential of G. intraradices to ameliorate oxidative stress generated in fenugreek plants by salinity was more evident at higher intensities of salt stress.
