Systemic hypothermia following spinal cord compression injury in the rat: axonal changes studied by β-APP, ubiquitin, and PGP 9.5 immunohistochemistry

Systemic hypothermia exerts neuroprotective effects in experimental ischemic CNS models caused by vascular occlusions. Recent experimental and clinical studies have also demonstrated beneficial effects of hypothermic treatment following brain trauma. The present study addresses the question as to whether systemic hypothermia has similar protective qualities following severe spinal cord compression trauma using beta-APP-, ubiquitin-, and PGP-9.5-immunohistochemistry combined with the ABC complex method as markers to identify axonal changes. Fifteen rats were randomized into three equally large groups and sustained to either thoracic laminectomy or to severe spinal cord compression trauma of the Th 8 - 9 segments. The non-trauma group contained laminectomized animals submitted to a hypothermic procedure in which the core temperature was reduced from 38 to 30 degrees C. The two trauma groups were either submitted to the same hypothermic procedure or kept normothermic during the corresponding time. All animals were sacrificed 24 h following the surgical procedure. In the hypothermic non-trauma group no axonal changes were seen. The number of abnormal axons, as indicated by accumulation of immunoreactive material in enlarged axons, was lower in the peri-injury zones of the hypothermic trauma group than in the normothermic trauma group. This difference was most obvious in the cranial peri-injury zones. No differences were seen between the groups in the trauma zones. This study demonstrates reduced axonal swelling in the peri-injury zones of spinal cord injured rats treated with systemic hypothermia. These changes could either indicate neuroprotective effects of the hypothermic treatment, or be results of reduced axonal transport or protein synthesis. To evaluate the clinical importance of our findings, further studies including reliable outcome measures of the animals must be performed.

Accumulation of SNAP-25 immunoreactive material in axons of Alzheimer's disease.

Neurofibrillary tangles and neuropil threads, both made of hyperphosphorylated tau proteins, point to an alteration of microtubules in Alzheimer's disease. The aim of this study was to test the consequences of these lesions on axoplasmic flow, which is dependent on intact microtubule assembly. We assessed the transport of synaptic proteins from the neuronal cell body to axonal terminals, using SNAP-25 (synaptosomal-associated protein of 25 kD) immunohistochemistry as a marker of impaired axonal transport. A sample from the supra-marginalis gyrus was obtained from 29 individuals over 75 years of age whose cognitive function had been prospectively assessed. Accumulation of immunoreactive material in swollen axons was observed in the white matter of severely demented individuals, and their number was correlated with the density of neurofibrillary tangles (r = 0.53, p = 0.005) and of focal Abeta deposits (r = 0.61, p = 0.001). This supports the hypothesis of a dysfunction of the cytoskeleton in Alzheimer's disease. An unexpected finding was the lack of correlation between SNAP-25 immunohistochemistry in the grey matter and the intellectual status or the density of neurofibrillary tangles, focal Abeta deposits and neuronal profiles. These results which question the role of synaptic markers as correlates of dementia, should be extended to other brain areas.

Characterization of a Novel 2′-5′ Oligoadenylate in Stimulated Lymphocytes

Abstract We have analysed Con A-stimulated mouse lymphocytes for the presence of 2′-5′-linked oligoadenylates using a radioimmunoassay based on a monoclonal antibody raised against adenylyl (2′-5′) adenosine (A2′pA). Time-course and Con A dose dependence were performed. We found that Con A induced, in a dose-dependant manner, the accumulation of immunoreactive material, together with the incorporation of 3H-thymidine in DNA. We showed that the immunoreactive material was constituted for the essential, by a novel 2′-5′ oligoadenylate. It was isolated and characterized as adenylyl 2′-5′adenylic acid (2′ and 3′P) according to the combination of criteria such as immunoreactivity, enzyme susceptibility, chromatographic behaviour and comparison with A2′pA3′(32P)p, A2′pA3′p acid A2′pA2′p that we have chemically synthetized. This is the first example of significant variations of a 2′-5′ oligoadenylate level in circumstances other than the antiviral mechanism of interferon.

Posttranslational processing of concanavalin A precursors in jackbean cotyledons.

Metabolic labeling of immature jackbean cotyledons with 14C-amino acids was used to determine the processing steps involved in the assembly of concanavalin A. Pulse-chase experiments and analyses of immunoprecipitated lectin forms indicated a complex series of events involving seven distinct species. The structural relatedness of all of the intermediate species was confirmed by two-dimensional mapping of 125I-tryptic peptides. An initial glycosylated precursor was deglycosylated and cleaved into smaller polypeptides, which subsequently reannealed over a period of 10-27 h. NH2-terminal sequencing of the abundant precursors confirmed that the intact subunit of concanavalin A was formed by the reannealing of two fragments, since the alignment of residues 1-118 and 119-237 was reversed in the final form of the lectin identified in the chase and the precursor first labeled. When the tissue was pulse-chased in the presence of monensin, processing of the glycosylated precursor was inhibited. The weak bases NH4Cl and chloroquine were without effect. Immunocytochemical studies showed that monensin treatment caused the accumulation of immunoreactive material at the cell surface and indicated that the ionophore had induced the secretion of a component normally destined for deposition within the protein bodies. Consideration of the tertiary structure of the glycosylated precursor and mature lectin showed that the entire series of processing events could occur without significant refolding of the initial translational product. Proteolytic events included removal of a peptide from the surface of the precursor molecule that connected the NH2- and COOH-termini of the mature protein. This processing activated the carbohydrate-binding activity of the lectin. The chase data suggest the occurrence of a simultaneous cleavage and formation of a peptide bond, raising the possibility that annealment of the fragments to give rise to the mature subunit involves a transpeptidation event rather than cleavage and subsequent religation.

Clinical applications of the radioimmunoassay of secretory tuberculoprotein.

A radioimmunoassay that measures a specific secretory tuberculoprotein was used to detect Mycobacterium tuberculosis in 9 of 30 liquid cultures of sputum. The accumulation of immunoreactive material in liquid cultures containing isoniazid was shown to reflect in vitro susceptibility of mycobacteria to the antibiotic effects of the drug.

Transport of cholecystokinin-octapeptide-like immunoreactivity toward the gut in afferent vagal fibres in cat and dog.

1. The distributions of gastrin- and cholecystokinin-like immunoreactivities in the dog and cat vagus nerves have been studied after nerve section and ligation. 2. In dogs, there was an increase in cholecystokinin-octapeptide-like immunoreactive material on the cranial side of ligatures on the thoracic or cervical vagi. When pairs of ligatures were tied on the cervical vagi there was accumulation proximal, and a slight decrease distal to, the upper ligature. There was also a modest increase distal to the lower ligature. 3. In cats, section of the vagus above the nodose ganglion, and hence degeneration of the efferent fibres, did not prevent increases in cholecystokinin-octapeptide-like immunoreactivity on the cranial side of ligatures which were later tied below the ganglion. Removal of the superior cervical ganglion had no effect on the accumulation of immunoreactive material above the ligatures. Section of the vagus below the nodose ganglion, and hence degeneration of both afferent and efferent fibres, abolished the accumulation on the cranial side of ligatures which were later tied below the section. Cholecystokinin-octapeptide-like material is therefore localized to afferent fibres with cell bodies in the nodose ganglion. 4. Immunoreactive forms were characterized by gel filtration and ion exchange chromatography, and the use of region-specific antisera. In all cats, and all but one dog, a molecule with the properties of sulphated cholecystokinin octapeptide was found to predominate. In some cats (30%) and dogs (26%) a molecule with the properties of heptadecapeptide gastrin (G17) was identified; concentrations of G17 were generally low compared with cholecystokinin octapeptide. In three dogs (20%) there was an accumulation of heptadecapeptide gastrin above the ligatures. 5. Axonal transport of cholecystokinin octapeptide in the vagus is consistent with a neuro-regulatory role for this peptide. However, the functional significance of its localization in afferent fibres, and transport towards the periphery, remains to be determined.

Neuronal systems immunologically related to the somatostatin system in the garden dormouse.

Cells and fibers containing somatostain (SRIF) or SRIF-like peptides were detected immunocytochemically in the brain of the garden dormouse (Eliomys quercinus L.). The periventricular preoptic nucleus and the paraventricular nucleus encompass a component of the SRIF-immunoreactive hypothalamo-infundibular and hypothalamo-neurohypophyseal systems. The suprachiastmatic, ventromedial and arcuate nuclei contain a number of SRIF-positive cells and receive a rich SRIF innervation. The extrahypothalamic systems containing SRIF can be subdivided into the following groups: (1) Afferents, the cellular origin of which is not always clearly evident, i.e., fibers of the septum, the lateral preoptic area, the thalamus, the superior olivary nucleus, the mesencephalic gray matter, and the subfornical organ; (2) dispersed cells with short projections (neocortex, caudate nucleus, putamen); (3) scattered cells with short projections (nucleus accumbens) or innervating remote territories (nucleaus interpeduncularis); (4) vascular organ of the lamina terminalis, a neurohemal area comparable to the median eminence. These observations lead to a theory of a functional bipotentiality of the somatostatin molecule. Immunocytological results depend on the antisera employed, the type of fixation and the experimental conditions. Adrenalectomy is followed by an accumulation of immunoreactive material in all SRIF-containing systems of the brain. These results clearly indicate the SRIF participates in the function of the CRF-ACTH-adrenal axis. The endocrine disturbance induced by adrenalectomy appears to modify the activity of both the neurohormonal and neuromodulator components of the SRIF system.

Substance P-, VIP-, and enkephalin-like immunoreactivity in the human vagus nerve

The human vagus nerve has been investigated for the presence of substance P (SP), vasoactive intestinal polypeptide (VIP), and enkephalin (ENK) using immunohistochemistry. After 0.5–4 hr of nerve ligation during surgical operations two right thoracic main truncs, two anterior subdiaphragmal trunks, and four anterior nerves of Latarjet were found to contain accumulation of immunoreactive material in nerve fibers above the ligation. Very high numbers of SP-, medium numbers of ENK-, and low numbers of VIP-immunoreactive fibers were seen. The relative proportions were similar at all levels studied. These data thus indicate the presence and axonal transport of SP-, ENK-, and VIP-like peptides in the human vagus nerve. Our observations in humans correlate well with results obtained from other species. Thus gastrointestinal vagal sensory mechanisms may be mediated by SP (and possibly VIP) and some motor mechanisms by ENK.
