Production of phenolic compounds, flavonoids, and lupeol by Lycium schweinfurthii suspension cultures in shake flasks and a rocking-motion bioreactor

AbstractLycium schweinfurthii, a wild shrub of the Solanaceae family, has received increasing attention in the last decade for its therapeutic potential in traditional medicine due to its diverse array of secondary metabolites, including phenolic substances and terpenoids. The aim of this study was to investigate the accumulation of phenolics, flavonoids, and the terpenoid lupeol in L. schweinfurthii cell suspension shake flask cultures and a single-use 2-dimensional rocking motion bioreactor. Three different media formulations were compared for in vitro cell cultures. Various parameters, such as biomass accumulation, settled cell volume, cell viability (assessed via a 2,3,5-triphenyl tetrazolium chloride assay), and sucrose consumption were determined as indicators of cell activity and growth. Total phenolic and flavonoid contents were estimated spectrophotometrically, lupeol was quantified via High-Performance Thin Layer Chromatography (HPTLC). Although a higher fresh biomass concentration of 464 g L− 1 was obtained in MS medium supplemented with a combination of each, 1 mg L− 1 of 2,4-Dichlorophenoxyacetic acid (2,4-D) and 1-Naphthaleneacetic acid (NAA), the rocking-motion bioreactor cultivation was performed with 2 mg L− 1 NAA due to its superior reproducibility in viability, productivity, and content of bioactive compounds (e.g., phenolics, flavonoids, lupeol). A final fresh biomass concentration of 185 g L− 1 was achieved in a 16 L cultivation scale with a notable increase in the concentration of phenolics (1.4-fold) and flavonoids (1.7-fold). Most importantly, the concentration of lupeol, a pentacyclic triterpenoid known for its anti-inflammatory, antibacterial, and anti-atherogenic properties, exhibited a remarkable 5.5-fold increase in the bioreactor cultivation (585 µg g− 1) compared to shake flask cultivations (106 µg g− 1). The current study demonstrated the profound impact of media composition and non-limited fed-batch conditions in a rocking-motion bioreactor on the accumulation of bioactive compounds. The findings are also relevant to other plant cell cultures.

Carvacrol maintains antioxidant capacity in goji fruit by increasing the content of bioactive compounds

Fresh goji berries are prone to decay and spoilage, resulting in economic losses. We investigated the effects of carvacrol (CVR) on the natural disease incidence and quality of goji berries by fumigating with varying CVR concentrations (0.06, 0.12, and 0.24 μL/mL). The results showed that CVR treatment inhibited the natural decay of goji berries by enhancing their quality, color, and total soluble solids/titratable acidity ratio. Moreover, 0.12 μL/mL CVR was determined to be the optimal concentration, as it maintained the sensory quality (except for aroma) and inhibited the disease incidence and preliminary disease expansion of goji fruit inoculated with Alternaria alternata. The treated berries exhibited elevated total phenolic content, total flavonoid content, ascorbic acid, glutathione, melatonin, total chlorophyll, and carotenoid levels. Higher hydrophilic and lipophilic total antioxidant activities and free radical scavenging capacities were also observed in the treated berries. Targeted metabolomics showed that treatment with 0.12 μL/mL CVR increased the levels of four phenolic acids (caffeic acid, chlorogenic acid, ferulic acid, and protocatechuic acid) and two flavonoids (naringenin and quercetin) in goji berries, thereby activating the gene expression of the phenylpropanoid metabolic pathway. In summary, CVR treatment delayed fruit senescence and the decline in fruit quality, promoted the accumulation of bioactive compounds, and improved antioxidant capacity and disease resistance by activating the phenylpropanoid metabolic pathway.

Metabolic and cannflavin profiles of germinated hemp seed (Cannabis sativa L.)

This study aimed to identify the optimal germination period for maximizing cannflavin A (CFL-A) and cannflavin B (CFL-B) production in hemp seeds. Utilizing ultra-high performance liquid chromatograph with orbitrap mass spectrometry (uHPLCOrbitrap-MS/MS), the research measured metabolomic activities, complemented by spectrophotometric analysis of antioxidant activity. The findings indicated that CFL-A and CFL-B concentrations progressively increased, reaching a peak at 72 h. Notably, the polar compounds saw the emergence of 4-hydroxybenzaldehyde, a derivative of enzymatic sugar-to-aldehyde conversion. In contrast, the non-polar fraction exhibited a decrease in cannabidiolic acid levels. The study also noted the potential formation of alpha-linolenic acid (omega-3 fatty acid) through hydrolysis, which serves as an energy source for germination. Metabolomic profiles were categorized by Agglomerative Hierarchical Clustering (AHC) and Principal Component Analysis (PCA), revealing significant changes from Day 0 to Day 1, followed by relative stability until Day 3. A substantial reduction in DPPH IC50 values was observed from 93.00 mg/kg (Day 0) to 43.18 mg/kg (Day 3), indicated stronger radical scavenging capacity. The highest FRAP and TPC value was recorded on Day 3, reflecting the accumulation of bioactive compounds, resulting in improved antioxidant activity.

Genomic and metabolomic insights into the selection and differentiation of bioactive compounds in citrus

Bioactive compounds are playing an increasingly prominent role in breeding functional and nutritive fruit crops such as citrus. However, the genomic and metabolic basis for the selection and differentiation underlying bioactive compounds variations in citrus remain poorly understood. Here, we constructed a species-level variation atlas of genomes and metabolomes using 299 citrus accessions. A total of 19,829 significant SNPs were targeted to 653 annotated metabolites, among which multiple significant signals were identified for secondary metabolites, especially flavonoids. Significantly differential accumulation of bioactive compounds in phenylpropane pathway, mainly flavonoids and coumarins, were unveiled across ancestral citrus species during differentiation, which is likely associated with the divergent haplotype distribution and/or expression profiles of relevant genes, including p-coumaroyl coenzyme A 2'-hydroxylases, flavone synthases, cytochrome P450 enzymes, prenyltransferases and UDP-glycosyltransferases. Moreover, we elucidated the citrus varieties with excellent antioxidant and anticancer capacities, clarifying the robust associations between distinct bioactivities and specific metabolites. Thus, these findings provide citrus breeding options for enrichment of beneficial flavonoids and avoidance of the potential risk of coumarins. This study will illuminate the application of genomic and metabolic engineering strategies in developing modern healthy citrus cultivars.

Constitutive and inducible tomato defenses contribute to Bacillus thuringiensis lethality against Spodoptera exigua

In nature, insect herbivory exerts continuous selective pressure on plants that, in turn, have developed a wide array of constitutive and inducible defenses to fight against attackers. Since plant defenses may affect higher trophic levels, including entomopathogens, further research is required to understand how plant compounds influence insect-pathogens interactions and their implications for integrated pest management programs. Here, we evaluated the impact of tomato defenses on the lethality produced by the bacterial entomopathogen Bacillus thuringiensis (Bt) against second instar larvae of Spodoptera exigua. We first examined the effect of constitutive defenses from fifteen wild tomato species and forty cultivated varieties (Solanum lycopersicum) on S. exigua larval growth and susceptibility to Bt. The results showed larvae fed on wild tomato species had a reduced larval growth compared to larvae fed on cultivated varieties, whereas susceptibility to Bt was similar between both groups. We then selected six cultivated varieties, including those with high and low impacts on larval growth and Bt-induced mortality, to further explore the effect of inducible defenses. Elicitation of defenses by methyl jasmonate (MeJA) reduced larval growth and increased basal mortality. Additionally, when larvae were infected with Bt, MeJA treatment further increased their susceptibility to the entomopathogen. Metabolomic analysis confirmed a reprogramming of tomato leaf metabolism following MeJA elicitation, with an induced accumulation of bioactive compounds, such as saponins or flavonoids, known for their anti-herbivory properties in most tomato varieties. Overall, these data reveal that both constitutive and inducible tomato defenses not only protect the plant by directly affecting the insect pest but also enhance the efficacy of bacterial entomopathogens

Effect of ultrasound pretreatment combined with different drying methods on the quality and bioactive compounds of broccoli stems

SummaryThe effects of ultrasound (US) pretreatment combined with drying process on storage quality maintenance and bioactive compound accumulation of broccoli stems were investigated. Results indicated that US pretreatment (λ = 8.5 mm) delayed the decline of L* value and ascorbic acid content but inhibited the increase of a* and b* values, browning index, and microbial counts of fresh‐cut broccoli stems. Furthermore, the US application enhanced total phenolics content by activating phenylalanine ammonia‐lyase, cinnamate‐4‐hydroxylase, and 4‐coumarate‐CoA ligase in phenylpropane metabolism. Meanwhile, it also promoted γ‐aminobutyric acid (GABA) accumulation by upregulating activities of diamine oxidase, polyamine oxidase, and aminoaldehyde dehydrogenase in GABA shunt and polyamine degradation pathway, respectively, which in turn, accelerated the biosynthesis of GABA from polyamines and glutamate. In addition, compared with other three drying methods, the combined vacuum freeze drying (VFD) could further retain the sensory attributes and bioactive compounds to a maximum extent in dehydrated broccoli stems. Hence, current findings illustrated that US pretreatment combined with VFD process is beneficial for commercial quality maintenance and nutritional value enhancement, thus providing new perspectives on the comprehensive utilisation of broccoli stems.

Spatial Metabolomic Profiling of Pinelliae Rhizoma from Different Leaf Types Using Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry Imaging.

Pinelliae Rhizoma (PR), a highly esteemed traditional Chinese medicinal herb, is widely applied in clinical settings due to its diverse pharmacological effects, including antitussive, expectorant, antiemetic, sedative-hypnotic, and antitumor activities. Pinellia ternata exhibits morphological variation in its leaves, with types resembling peach, bamboo, and willow leaves. However, the chemical composition differences among the corresponding rhizomes of these leaf phenotypes remain unelucidated. This pioneering research employed Matrix-Assisted Laser Desorption/Ionization Mass Spectrometry Imaging (MALDI-MSI) to conduct the in situ identification and spatial profiling of 35 PR metabolites in PR, comprising 12 alkaloids, 4 organic acids, 12 amino acids, 5 flavonoids, 1 sterol, and 1 anthraquinone. Our findings revealed distinct spatial distribution patterns of secondary metabolites within the rhizome tissues of varying leaf types. Orthogonal Partial Least Squares Discriminant Analysis (OPLS-DA) effectively differentiated between rhizomes associated with different leaf morphologies. Furthermore, this study identified five potential differential biomarkers-methylophiopogonanone B, inosine, cytidine, adenine, and leucine/isoleucine-that elucidate the biochemical distinctions among leaf types. The precise tissue-specific localization of these secondary metabolites offers compelling insights into the specialized accumulation of bioactive compounds in medicinal plants, thereby enhancing our comprehension of PR's therapeutic potential.

Oxidative stress and culture atmosphere effects on bioactive compounds and laccase activity in the white rot fungus Phlebia radiata on birch wood substrate

Wood-decaying white rot fungi live in changing environmental conditions and may switch from aerobic to fermentative metabolism under oxygen depletion. Decomposition of wood and lignocellulose by fungi is dependent on enzymatic and oxidative biochemistry including generation of reactive oxygen species. In this study, we subjected semi-solid wood-substrate cultures of the white rot fungus Phlebia radiata to oxidative stress by addition of hydrogen peroxide under aerobic and anaerobic cultivation conditions. Wood decomposition and fungal metabolism were followed by analysis of extracellular organic compounds, mycelial growth, and laccase activity. Under both atmospheric conditions, accumulation of bioactive aromatic compounds from birch wood occurred into the culture supernatants after hydrogen peroxide treatment. The supernatants inhibited both fungal growth and laccase activity. However, the fungus recovered from the oxidative stress quickly in a few days, especially when cultivated under regular aerobic conditions. With repeated hydrogen peroxide treatments, laccase suppressive-recovery effect was observed. Culture supernatants demonstrated antioxidant and antimicrobial effects, in concert with emergence of chlorinated birch-derived organic compounds. Bioactivities in the cultures disappeared in the same pace as the chlorinated compounds were transformed and de-chlorinated by the fungus. Our results indicate tolerance of white rot fungi against excessive oxidative stress and wood-derived, growth-inhibiting and harmful agents.

Determination of Biologically Active Compounds and Antioxidant Capacity In Vitro in Fruit of Small Cranberries (Vaccinium oxycoccos L.) Growing in Natural Habitats in Lithuania.

The composition of flavonols, proanthocyanidins, anthocyanins, triterpene compounds, and chlorogenic acid in small cranberry fruit samples collected in natural habitats in Lithuania and variation in the antioxidant capacity of cranberry fruit extracts was determined. This study showed that in the flavonol group, hyperoside and myricetin-3-O-galactoside predominated in cranberry fruit samples; in the anthocyanin group, the predominant compounds were cyanidin-3-O-galactoside, cyanidin-3-O-arabinoside, peonidin-3-O-galactoside, and peonidin-3-O-arabinoside, and in the group of triterpene compounds, ursolic acid was predominant. The highest total amounts of flavonols and anthocyanins were found in the samples collected in Čepkeliai State Strict Nature Reserve (2079.44 ± 102.99 μg/g and 6993.79 ± 350.22 μg/g, respectively). Cluster analysis of the chemical composition of small cranberry fruit samples revealed trends in the accumulation of bioactive compounds in cranberry fruit. Cranberry fruit samples collected in central Lithuania had higher levels of triterpene compounds. Statistical correlation analysis showed the strongest correlation between the quantitative composition of cyanidin-3-O-arabinoside and peonidin-3-O-arabinoside and the reducing capacity of the ethanolic extracts of the cranberry fruit samples assessed in vitro by the FRAP assay (r = 0.882, p < 0.01 and r = 0.805, p < 0.01, respectively). Summarizing the results, the geographical factor affects the variation of the quantitative composition of biologically active compounds in cranberry fruit samples.

Argovit™ silver nanoparticles transform agro-waste into phenolic biofactories: Postharvest stress for high-value compound production in prickly pear peels

The growing demand for food has resulted in an increased generation of plant waste residues. These waste materials contain various bioactive compounds, but their concentrations are typically low, rendering their extraction economically unviable. In this study, we assessed the impact of abiotic stress induced by silver nanoparticles (AgNPs) on the compounds derived from red and green prickly pear peels. Six different AgNP lots were tested at varying concentrations. The total phenol content was consistently higher than that of the control group across all lots. Notably, the AgNP systems exhibited selectivity in inducing the formation of specific compounds, such as caffeic (10.36%), vanillic (6.71%), p-coumaric (6.63%), malic (4.86%) acids, and catechin (46.15%). Moreover, the betanin content in red prickly pear increased by up to 2.4 times compared to the control group. However, the data analysis indicated a tendency towards increased variability in phenolic acids and flavonoids. This suggests that the AgNP systems might be activating these metabolic pathways. These findings are highly relevant as they demonstrate the potential of AgNP systems to enhance the accumulation of bioactive compounds in agro-industrial waste materials, which can then be utilized in the development of functional ingredients.

Ultraviolet-to-Blue Light Conversion Film Affects Both Leaf Photosynthetic Traits and Fruit Bioactive Compound Accumulation in Fragaria × ananassa

The influence of light downconversion films (red, pink and blue films) on leaf physiological features and fruit bioactive compound accumulation was studied in strawberry plants. Red, pink and blue films were able to convert light less utilised by plants into more efficient light wavebands with various possibilities depending on the film characteristics (blue film shifts UV into blue radiation; red film shifts green into red radiation, pink film shifts UV and green into blue and red radiation but to a lesser extent than red film). Indeed, by enhancing the quality of light available for photosynthesis, the utilization of these films holds the potential to improve agricultural productivity and sustainability. All of these light downconversion films resulted in higher plant fresh weight compared to a control colourless (Cnt) film, with plants grown under blue film (UV-to-blue light conversion) showing the most impressive results in terms of total leaf area (+25%), leaf thickness (+10%) and leaf mass per area (+15%). Simultaneously, during the flowering stage, plants under blue film had a higher net photosynthetic rate due to the increase in photosynthetically efficient wavelengths. Moreover, blue film resulted in the highest total phenolic (+40% and +28% than red and pink films, respectively) and flavonoid content (+54%, +84%, +70% than Cnt, red and pink films, respectively) in fruit, with specific effects on targeted phenols, i.e., quercetin, ellagic acid and its glycoside, ellagitannins, and procyanidins. In conclusion, the use of the UV-to-blue conversion light film tested herein represents an innovative solution to increase strawberry yield and promote fruit nutraceutical features, playing a pivotal role in ensuring food quality and security and sustainable agricultural practices.

Unveiling the potential of native arbuscular mycorrhizal fungi for growth promotion and phytochemical enrichment in Valeriana jatamansi Jones.

Medicinal plants are rich sources of pharmaceutically important compounds and have been utilized for the treatment of various diseases since ancient times. Valeriana jatamansi Jones, also known as Indian valerian, holds a special place among temperate Himalayan medicinal plants and is renowned for its therapeutic properties in addressing a variety of ailments. The therapeutic potential of V. jatamansi is attributed to the presence of valuable compounds such as valepotriates, sesquiterpenoids, valeriananoids, jatamanins, lignans, cryptomeridiol, maaliol, xanthorrhizzol, and patchouli alcohol found in its rhizome and roots. This study employed various treatments, including the cultivation of V. jatamansi with the inoculation of Funneliformis mosseae, F. constrictus, and a consortium of arbuscular mycorrhizal fungi (AMF), to investigate their influence on biomass production, chlorophyll content, and the accumulation of bioactive compounds in V. jatamansi. The results revealed significant improvement in these parameters in the inoculated plants. The parameters of plants inoculated with F. mosseae were the highest, followed by those of plants inoculated with F. constrictus and a mixture of AMFs. This study not only underscores the potential of native AMF for promoting the growth of V. jatamansi but also elucidates their role in influencing the synthesis of bioactive compounds. The cultivation of V. jatamansi with native AMF has emerged as a sustainable and eco-friendly approach, providing the dual benefit of enhancing both the medicinal and economic value of this valuable plant. This research contributes valuable insights into the practical application of mycorrhizal associations for the cultivation of medicinal plants, bridging the realms of agriculture and pharmaceuticals.

Accumulation of 5-methyltetrahydrofolate and other bioactive compounds, in the course of fermentation of green tea (Camellia sinensis) kombucha

Kombucha is a potential probiotic tea-based drink with increasing worldwide consumption. Studies on this probiotic beverage are growing rapidly, especially about micronutrients and microbial population. As such, the present study performed the molecular identification of the microorganism and evaluated 5-methyltetrahydrofolate content by HPLC-DAD, phenolic compounds, flavonoids, carotenoids, antioxidant activity by spectrophotometric methods, and physicochemical composition of green tea kombucha on fermentation days 1, 3, 7, 14, and 21. DNA sequencing identified the Microbacterium genus as predominant. However, was unable to safely determine the species level because of the rRNA 16S gene sequence similarity between four species M. ureisolvens, M. yannicii, M. chocolatum e M. atlanticum. The concentration of 5-methyltetrahydrofolate found on the third day was 39.12 ± 1.32 μg/mL (liquid) and 45.78 ± 8.42 μg/mL (polymeric biofilm); On the twenty-first day it was 50.87 ± 3.56 μg/mL (liquid) and 54.88 ± 3.89 μg/mL (polymeric biofilm). Total phenolic compounds increased with fermentation; however, flavonoids and carotenoids were degraded by the process. The information on 5-methyltetrahydrofolate is unprecedented and highly relevant for food guidelines, since related deficiencies can lead to fetal malformation in the first three months of pregnancy. Lastly, the best fermentation time to obtain 5-methyltetrahydrofolate and others bioactive compounds is between days 7–14. Further analyses are also encouraged to understand the bioavailability of the vitamin.

Response to Various Water Regimes of the Physiological Aspects, Nutritional Water Productivity, and Phytochemical Composition of Bush Tea (Athrixia phylicoides DC.) Grown under a Protected Environment

The influence of water regimes on plants is crucial for integrating bush tea (Athrixia phylicoides DC.) into strategies in Sub-Saharan Africa to tackle food and nutritional insecurity by considering physiological aspects, nutritional yield, nutritional water productivity, and metabolite composition. The objective of the study was to determine the physiological aspects, including leaf gas exchange and chlorophyll fluorescence, nutritional yield, nutritional water productivity, and metabolite composition of bush tea under varying water regimes. The tunnel experiment was laid out in a randomized complete block design (RCBD) with treatments consisting of three water regimes: 100% of crop water requirement (ETa), 30% of ETa, and a control (no irrigation), all replicated three times. The morphological aspects were recorded on a weekly basis. However, yield, nutrient content, nutritional water productivity (NWP), and phytochemical composition were determined at harvest. The phytochemical analysis by liquid chromatography mass spectrometry (LC-MS), coupled with visualization of the detected chemical spaces through molecular networking, indicated Athrixia phylicoides DC. to be rich in various bioactive compound derivatives, including methyl chlorogenate, flavonoids, tartaric acid, caffeoylquinic acid, and glutinane. The results showed that 30% ETa enhanced plant growth, nutrient content, and nutritional water productivity compared to other water treatments. Nevertheless, 100% ETa yielded more (95.62 kg ha−1) than 30% ETa (60.61 kg ha−1) and control (12.12 kg ha−1). The accumulation of chlorogenic acids was higher under 30% ETa compared to 100% ETa and control. Therefore, this study is the first to determine the accumulation of various bioactive compounds in bush tea leaf extracts under varying water regimes. This confirms that in areas with low water availability, bush tea is well adapted for production without limiting nutrients.

Metabolome and transcriptome reveal high abundance of bioactive substances in albino jujube fruit as potential function food

Little is known about the fruit quality of albino mutants, due to generally exhibited lethal phenotypes caused by restrained photosynthesis and energy metabolism. In this study, a non-lethal bud mutant of jujube that could bear fruit with higher nutrient content after leaf albinism was described. Compared to normal mature fruit, the albino fruit contained lower soluble sugar and titratable acid, as well as higher ascorbic acid, phenols and flavonoids contents. Through metabolome and transcriptome analyses, 109, 117, and 126 differentially accumulated metabolites (DAMs) and 1169, 797, and 3059 differentially expressed genes (DEGs) were identified in the young, white-ripened, and mature stages of albino and normal fruits, respectively. KEGG annotation indicated that DEGs and DAMs were mainly enriched in phenylpropanoid and flavonoid biosynthesis, glutathione metabolism, and α-linolenic acid metabolism. Moreover, ADH, GST17, and POD16 were identified as hub genes that play major roles in high abundance of chalcone, (−)-epigallocatechin, GSH, and total ascorbate. The alteration of bioactive substance content in albino fruits was an adaptive molecular mechanism that responded to adverse mutations, and reflected a higher antioxidant capacity. These findings provide new insights into bioactive compounds accumulation in albino mutants, and provide experimental evidence for albino fruit as a potential nutraceutical or functional food.

Effect of Preharvest Treatment Using Jasmonic Acid and Methyl Jasmonate on the Physicochemical Properties and Antioxidant Activities of Red-fleshed Dragon Fruit (Hylocereus polyrhizus L.)

This study investigated the effect of exogenous plant growth regulators (PGR), namely jasmonic acid (JA) and methyl jasmonate (MeJA), on the physicochemical properties of flesh and peels of red-fleshed dragon fruit (Hylocereus polyrhizus). The fruit was sprayed with 100 and 1,000 ppm of JA and MeJA at 15 and 22 days of anthesis and harvested after 35 days. Then, the flesh and peels were analyzed for total soluble solids (TSS), total betacyanins, betanin, total phenolics (TP), total flavonoids (TF), and color characteristics. The fruit peels contained significantly higher (p&lt;0.05) TP and antioxidant activities compared to flesh. No significant difference was detected between the variables in the peels, except for significantly higher (p&lt;0.05) of total betacyanins (~295.6 and ~299.9 mg/100 g) and TP (~614.1 and 566.1 mg GAE/100 g) were recorded in control and 100 ppm MeJA, respectively. In the flesh, 1,000 ppm MeJA-treated fruit possessed the highest total betacyanins (~139.2 mg/100 g), betanin (~356.0 mg/g), TP (~244.9 mg GAE/100 g), TF (~329.0 mg CE/100 g), Trolox equivalent antioxidant capacity (TEAC) (63.2 µmol TE/g) and reducing power (~21.5 µmol TE/g). Overall, 1,000 ppm MeJA was more effective in enhancing the accumulation of bioactive compounds and antioxidant activities in the flesh of red-fleshed dragon fruit compared to other PGR treatments.

Arbuscular mycorrhizal fungi promote growth and enhance the accumulation of bioactive compounds in Tomato (Solanum lycopersicum L.).

Arbuscular mycorrhizal fungi (AMF) have been known to enhance plant growth and nutrient uptake. In this study, we investigated the effects of Funneliformis mosseae, Rhizophagus intraradices, and their co-inoculation on the growth and biochemical composition of tomato (Solanum lycopersicum L.) plants. The findings demonstrated that the inoculation of AMF significantly enhanced shoot and root length, shoot and root dry weight, number of fruits per plant, as well as concentrations of anthocyanin, phenolic compounds, and antioxidants in tomato plants. Both individual and co-inoculation of AMF also significantly increased nitrogen, phosphorus, and potassium concentrations in tomato plants. Our findings suggest that AMF can be used as a potential biofertilizer to enhance the growth and biochemical composition of tomato plants.

Salty treatment increased bioactive compounds accumulation during agarwood development in Aquilaria sinensis trees

To compare the bioactive compounds in agarwood induced by different methods in Aquilaria sinensis(Lour.) Gilg trees, a two dimensional thin layer chromatograph(2D-TLC) combined with effect directive analysis(EDA) was developed. Three antioxidants were found by 2D-TLC-DPPH and further identified as 2-(2-phenylethyl) chromones(PECs) with LC-MS/MS. The 3 antioxidants decreased along agarwood formation and their compositions in drilling induced agarwood differed with those in microbe culture induced agarwood. Further study showed NaCl treatment promoted antioxidants accumulation in agarwood induced by drilling or hot drilling. Hot drilling combined with salty stimulation was most efficient in some chemicals accumulation, which were identified as PECs with antioxidant, tyrosinase or β-glucosidase inhibiting activities by 2D-TLC-EDA-LC-MS/MS. This study provided a 2D-TLC-EDA-LC-MS/MS method for bioactive compounds screen and qualification of agarwood. Based on this method, non-conventional methods were found to accelerate the accumulation of some bioactive PECs in A. sinensis trees.

Effect of UV-A Irradiation on Bioactive Compounds Accumulation and Hypoglycemia-Related Enzymes Activities of Broccoli and Radish Sprouts

In the present study, different intensities of UV-A were applied to compare their effects on growth, bioactive compounds and hypoglycemia-related enzyme activities in broccoli and radish sprouts. The growth of sprouts was decreased after UV-A irradiation. A total of 12 W of UV-A irradiation resulted in the highest content of anthocyanin, chlorophyll, polyphenol and ascorbic acid in broccoli and radish sprouts. The highest soluble sugar content was recorded in sprouts under 8 W of UV-A irradiation, while no significant difference was obtained in soluble protein content among different UV-A intensities. Furthermore, 12 W of UV-A irradiation induced the highest glucosinolate accumulation, especially glucoraphanin and glucoraphenin in broccoli and radish sprouts, respectively; thus, it enhanced sulforaphane and sulforaphene formation. The α-amylase, α-glucosidase and pancrelipase inhibitory rates of two kinds of sprouts were enhanced significantly after UV-A irradiation, indicating UV-A-irradiation-treated broccoli and radish sprouts have new prospects as hypoglycemic functional foods.
