Translocation of platelets into Disse spaces and their entry into hepatocytes in response to lipopolysaccharides, interleukin-1 and tumour necrosis factor: the role of Kupffer cells

Background/Aims: Injection into mice of a small dose of either a lipopolysaccharide or interleukin-1 induces a slowly developing accumulation of 5-hydroxytryptamine, predominantly in the liver. We have established that this 5-hydroxytryptamine accumulation is the result of the translocation of platelets to hepatic sinusoidal spaces and, further, into Disse spaces, and that the platelets make direct contact with hepatocytes. In the present study, we report our recent findings on this phenomenon. Methods: Platelets contain a large amount of 5-hydroxytryptamine, but the 5-hydroxytryptamine content of the liver is normally very small. Therefore, the translocation of platelets to the liver was assessed by measuring 5-hydroxytryptamine as in previous studies, and it was also analysed by electron microscopy. Results: Anti-platelets agents, such as heparin and inhibitors of prostaglandin synthesis, were ineffective in preventing the lipopolysaaccharide-induced accumulation of 5-hydroxytryptamine in the liver. Of the various cytokines tested, only interleukin-1 and tumour necrosis factor induced such an accumulation of 5-hydroxytryptamine. Intravenous injection of liposomes encapsulating dichloromethylene bisphosphonate resulted in an almost complete depletion of macrophages from the liver. The lipopolysaccharide- and cytokine-induced hepatic accumulations of 5-hydroxytryptamine were abolished almost completely in such macrophage-depleted mice. Electron microscopy revealed no accumulation of platelets in the liver after injection of lipopolysaccharide into the macrophage-depleted mice. Surprisingly, in normal mice injected with lipopolysaccharide, several platelets were found inside some hepatocytes, even though there was no visible damage to these hepatocytes. In fact, there were many polysomes around the degranulated platelets within the hepatocytes, suggesting an enhanced protein synthesis. Conclusion: These results suggest that, in response to lipopolysaccharide, interleukin-1 or tumour necrosis factor, platelets translocate into the liver in a way that is different from aggregation, and that some, at least, enter hepatocytes. During these processes, hepatic macrophages play an essential role.

Accumulation of 5-Hydroxytryptamine by Aging Platelets: Studies in a Model of Suppressed Thrombopoiesis in Dogs

Thrombocytopenia was induced in healthy, male mongrel dogs by intramuscular injection of a single dose of estradiol valerate (1 mg/kg). A steady, almost linear decay of the blood platelet count starting about day 6 post-estradiol and attaining a mean value of 14 x 10(3) platelets/microliters one week later was observed. Thrombocytopenia is explained mainly by suppression of thrombocytopoiesis, as established by two independent ways: 1. Megakaryocytes in the bone marrow were markedly reduced. 2. Kinetic studies with 111In labeled autologous platelets revealed a nearly linear decay of the radioactivity and mean survival times within the expected range. The progressive reduction in the platelet count is associated with an increase in the mean age of the platelets still circulating. Following estradiol injection, platelet 5-hydroxytryptamine (5-HT) increased from a basal value of 130 +/- 30 ng/10(8) platelets (platelet count of 351 +/- 53 x 10(3) platelets/microliters) to 343 +/- 100 ng/10(8) platelets eleven days latter, when the platelet count dropped to 32 +/- 18 x 10(3) platelets/microliters. No significant changes in the number or affinity of the 5-HT uptake receptors could be demonstrated in platelets exposed in vitro and in vivo to estradiol. Our results indicate that aging platelets accumulate 5-HT, probably by a sustained exposure to the monoamine in plasma, confirming previous observations based on models in which thrombopenia was induced by immune and mechanical means.

The effect of lipopolysaccharide, interleukin-1 and tumour necrosis factor on the hepatic accumulation of 5-hydroxytryptamine and platelets in the mouse.

1. Injection of lipopolysaccharide (LPS; 0.5-500 microgram kg-1) into mice induced a dose-dependent, slowly developing increase in hepatic content of 5-hydroxytryptamine (5-HT). This sustained increase could not be attributed to an LPS-induced alteration of the pharmacokinetic handling of 5-HT by stimulation of its uptake or inhibition of its degradation. 2. Regional differences were apparent in the tissue content of histamine and 5-HT between mast cell-deficient (W/Wv) and normal (+/+) mice. LPS administration (0.5 mg kg-1) gave comparable increases in the hepatic level of 5-HT in mast cell-deficient and normal mice. 3. Reserpine pretreatment (1 mg kg-1) selectively reduced 5-HT levels in the blood, spleen, liver, brain and lung of normal mice. Prior treatment with this agent also abolished the LPS (0.5 mg kg-1)-induced hepatic accumulation of 5-HT. 4. Accumulation of 5-HT in the liver by LPS (0.1 mg kg-1) was temporally associated with both a fall in the levels of circulating platelets, and a reduction in the concentration of 5-HT in the blood. The LPS dose-dependent (0.5-500 micrograms kg-1) increase in hepatic 5-HT content was associated with a similar dose-dependent reduction in the circulating levels of 5-HT. 5. Interleukin-1, alpha and beta (10 micrograms kg-1) and tumour necrosis factor alpha (TNF alpha) (1 mg kg-1) significantly enhanced the accumulation of 5-HT within the liver. Administration of TNF alpha (10 micrograms kg-1) potentiated the increase in hepatic 5-HT content seen with IL-1 beta (10 micrograms kg-1). 6. Electron microscopy revealed numerous platelets in the sinusoidal and perisinusoidal Disse spaces within the liver, in animals pretreated with LPS (0.1 mg kg '). The platelets retained their intact structure and showed no evidence of degranulation. 7. These data suggest that the LPS and cytokine-induced mobilization of 5-HT in the liver is associated with the hepatic translocation of platelets. This migration appears to be independent of platelet aggregation.

Cerebrovascular nerves in old rats show reduced accumulation of 5-hydroxytryptamine and loss of nerve fibres

The effect of old age on the net accumulation of 5-hydroxytryptamine (serotonin) into cerebrovascular nerves of rats was studied using a new, densitometric application of image analysis of immunohistochemical staining. The results showed evidence of reduced accumulation in old animals in nerves supplying arteries of the circle of Willis. Measurements of the density of nerves stained with an antibody to the neuronal marker, PGP9.5, showed that loss of nerve fibres may occur in some cerebral vessels in old age.

Catabolism and disposition of exogenous 5-hydroxytryptamine in cockroach, Periplaneta americana, tissues

The concentrations of tryptophan, 5-hydroxytryptophan, 5-hydroxytryptamine, N-acetyl 5-hydroxytryptamine and N-acetyl dopamine were determined in the cerebral ganglia, haemolymph and Malpighian tubules of the cockroach Periplaneta americana, using high performance liquid chromatography with electrochemical detection. Injected 5-hydroxytryptamine was rapidly removed from the haemolymph with a concomitant elevation of circulating N-acetyl 5-hydroxytryptamine and little accumulation of 5-hydroxytryptamine in the cerebral ganglia. N-acetyl 5-hydroxytryptamine and N-acetyl dopamine were also rapidly removed from the haemolymph. Incubation of haemolymph from 5-hydroxytryptamine-injected insects and glucosidase or phosphatase, indicated that most of the injected 5-hydroxytryptamine had been converted to a sugar conjugate of N-acetyl 5-hydroxytryptamine. Whole haemolymph did not catabolize 5-hydroxytryptamine or N-acetyl 5-hydroxytryptamine whereas Malpighian tubules N-acetylated both 5-hydroxytryptamine and dopamine and metabolized N-acetyl 5-hydroxytryptamine. Injection of p-chlorophenylalanine (200 and 500 μg/g) had no effect on 5-hydroxytryptamine concentrations in the cockroach cerebral ganglia.

Accumulation of 5-hydroxytryptamine leads to dysfunction of adrenergic nerves in canine coronary artery following intimal damage in vivo.

Previous in vitro studies have demonstrated that coronary artery adrenergic nerves are a principal site of accumulation of 5-hydroxytryptamine released from aggregating platelets. The purpose of this study was to determine whether 5-hydroxytryptamine is accumulated by adrenergic nerves at sites of endothelial damage and platelet aggregation in vivo. Coronary artery 5-hydroxytryptamine content and response to in vitro adrenergic nerve stimulation were studied in dogs 24 hours following balloon catheter-induced intimal injury. 5-Hydroxytryptamine content was significantly increased in the catheter-damaged arteries, and there was a coincident decrease in the content of norepinephrine. The relaxation caused by acetylcholine was abolished in the catheter-injured arteries, indicating loss of this endothelial cell-mediated function. The normal beta-adrenergic relaxation caused by nerve stimulation was inhibited, and in some cases, contractions resulted; these effects were prevented by serotonergic receptor antagonists. The sensitivity to exogenously added norepinephrine was unchanged, indicating that the changes in the response to nerve stimulation were not due to an altered smooth muscle response to the native neurotransmitter. These observations indicate that following intimal damage, which produces platelet aggregation on the luminal surface of the blood vessel, 5-hydroxytryptamine can assume a transmitter role in coronary artery adrenergic nerves and thereby cause their dysfunction.

Inhibition of 5-hydroxytryptamine accumulation and deamination by substituted phenylalkylamines in hypothalamic synaptosomes from normal and reserpine-pretreated rats

1. In the present study the abilities of different compounds to inhibit MAO inside and outside the serotonergic neurons, to inhibit the accumulation of 5-HT and to release 5-HT were separated by using different in vitro techniques. With these methods a number of substituted phenylalkylamines, which are reversible inhibitors of monoamine oxidase (MAO) type A, were characterized. 2. The compounds were examined regarding their ability to inhibit the accumulation of 5-HT and to inhibit MAO in the same synaptosomal preparation of hypothalamus from normal and reserpine-pretreated rats. The difference in the uptake of 14C-5-HT (0.1 mumol/l) in the absence and presence of citalopram (0.25 mumol/l) was taken as a measure of the accumulation into the serotonergic synaptosomes. The deamination of 14C-5-HT (0.1 mumol/l) in the presence of citalopram (0.25 mumol/l) was considered as that brought about outside the serotonergic synaptosomes, whereas the difference between the deamination in the absence and presence of citalopram was taken as the MAO activity inside the serotonergic synaptosomes. 3. Most of the phenylalkylamines were slightly more potent as MAO inhibitors outside serotonergic synaptosomes than as inhibitors of 5-HT accumulation in normal rats. The most potent MAO inhibitors, both in absolute terms and in comparison with uptake inhibitory potency, were the 2,6-dichloro-(FLA 365) and the phenylpropylene-(FLA 417) derivatives. 4. A difference in potency on the accumulation in synaptosomes from normal and reserpine-pretreated rats was found for many of the phenylalkylamines with the exception of FLA 365, FLA 417 and the 2,5-dimethyl derivative RAN 113.(ABSTRACT TRUNCATED AT 250 WORDS)

Putrescine and 5-hydroxytryptamine accumulation in rat lung slices: Cellular localization and responses to cell-specific lung injury

The cellular localization of putrescine (1,4-diaminobutane) and 5-hydroxytryptamine (5HT) following the accumulation of tritium-labeled putrescine (2.5 μ m) or 5HT (0.5 μ m) into rat lung slices was determined by autoradiography at the light microscope level. Putrescine labeling was found to occur in type II alveolar epithelial cells and in bronchiolar nonciliated (Clara) cells, and possibly also in type I alveolar epithelial cells. The pattern of 5HT labeling was clearly different from that with putrescine, since the parenchyma was diffusely labeled with no preferential location in type II cells, but with strong labeling of the endothelium of large vessels and also the pleural mesothelium. The apparent kinetic parameters for the tissue uptake of [ 3H]putrescine (2.5 to 80 μ m) and [ 14C]5HT (0.5 to 16 μ m; both being simultaneously present in a 5 to 1 molar ratio) were studied in lung slices from normal rats and rats pretreated with O,S,S-trimethyl phosphorodithioate (OSSMe, 11 to 95 mg/kg, po), with paraquat (20 mg/kg, ip), or with α-naphthylthiourea (ANTU, 5 or 10 mg/kg, ip). OSSMe and paraquat were used as models for pulmonary epithelium-damaging agents, and ANTU was taken as a model for a pulmonary endothelium-damaging agent. The V max for the uptake of 5HT was significantly increased (without change in K m ) following treatment with OSSMe and paraquat. Following ANTU treatment the V max for the uptake of 5HT was unchanged (5 mg/kg) or increased (10 mg/kg, K m also increased). These results indicate that in lung slices the response to lung injury may be associated with an increased accumulation of 5HT. The V max for the uptake of putrescine was significantly decreased (without change in K m ) following treatment with OSSMe and paraquat. Following ANTU treatment the V max for the uptake of putrescine was unchanged (5 mg/kg) or decreased (10 mg/kg, no change in K m ). These results suggest that a decreased putrescine uptake is a sensitive index of pulmonary epithelial damage.

Suppression and potentiation of 5-hydroxytryptophan-induced hypoglycaemia by alpha-monofluoromethyldopa: correlation with the accumulation of 5-hydroxytryptamine in the liver.

Experiments were done to examine whether the accumulation of 5-hydroxytryptamine (5-HT) in the liver is responsible for the hypoglycaemia induced in mice by 5-hydroxytryptophan (5-HTP) and lipopolysaccharides (LPS). (+/-)-alpha-Monofluoromethyldopa (FMD), a potent irreversible inhibitor of aromatic amino acid decarboxylase, suppressed the 5-HTP-induced accumulation of 5-HT in the liver at a dose of 2 mg kg-1 or more, but potentiated the accumulation at lower dose of 0.4 mg kg-1. Corresponding to these effects, the hypoglycaemic response was prevented by the higher doses of FMD and potentiated by the lower dose. These contrasting effects of FMD were explicable by the amounts of 5-HTP entering the liver. In contrast, FMD did not prevent either the hypoglycaemia or the accumulation of 5-HT in the liver induced by LPS. These results further support the hypothesis that the accumulation of 5-HT in the liver is causally related to the hypoglycaemia induced by 5-HTP and indicate that the LPS-induced 5-HT accumulation in the liver is not derived from stimulation of 5-HT synthesis. It is still not clear whether the accumulation of 5-HT in the liver is involved in the hypoglycaemic response to LPS.

A Comparison of Biochemical Indices of 5‐Hydroxytryptaminergic Neuronal Activity Following Electrical Stimulation of the Dorsal Raphe Nucleus

The activity of 5-hydroxytryptaminergic neurons has been estimated from measurements of: concentrations of 5-hydroxyindoleacetic acid; the ratio of the concentrations of 5-hydroxyindoleacetic acid to 5-hydroxytryptamine; the rate of accumulation of 5-hydroxytryptophan following the administration of an aromatic L-amino acid decarboxylase inhibitor (e.g., NSD 1015); the rate of accumulation of 5-hydroxytryptamine, and the rate of decline of 5-hydroxyindoleacetic acid following the administration of a monoamine oxidase inhibitor (e.g., pargyline). The purpose of the present study was to compare these different methods under conditions of changing neuronal impulse traffic produced by electrical stimulation of 5-hydroxytryptaminergic neurons. Male rats anesthetized with chloral hydrate were killed following 0, 15, or 30 min of electrical stimulation of the dorsal raphe nucleus at a frequency of 0, 5, or 10 Hz. The concentrations of 5-hydroxytryptamine, 5-hydroxyindoleacetic acid, and 5-hydroxytryptophan in nucleus accumbens, amygdala, suprachiasmatic nucleus, and dorsomedial nucleus were measured using HPLC coupled to an electrochemical detector. In each brain region, stimulation elicited an increase in the concentration of 5-hydroxyindoleacetic acid and the 5-hydroxyindoleacetic acid/5-hydroxytryptamine concentration ratio in saline-treated animals and an increase in 5-hydroxytryptophan accumulation in NSD 1015-treated animals, but did not alter the concentration of 5-hydroxytryptamine or 5-hydroxyindoleacetic acid in pargyline-treated rats. The results o f this study indicate that although the first three methods serve as valid indices of 5-hydroxytryptaminergic neuronal activity, the pargyline-dependent techniques are not responsive to changes in the rate of 5-hydroxytryptamine nerve firing.

Metergoline, pirenperone and pizotifen alter dopamine and 5-hydroxytryptamine synthesis in discrete rat brain nuclei

The effects of the 5-hydroxytryptamine receptor antagonists metergoline, pirenperone and pizotifen on 5-hydroxytryptamine and dopamine synthesis were determined by measuring the rate of accumulation of 5-hydroxytryptamine and 3,4-dihydroxyphenylalanine, respectively, after administering l-tryptophan and m-hydroxybenzylhydrazine, an inhibitor of aromatic-l-amino acid decarboxylase. 5-Hydroxytryptophan, 3,4-dihydroxyphenylalanine and l-tryptophan were measured in four forebrain regions, the caudate putamen, nucleus accumbens, nucleus septi lateralis, and nucleus amygdaloideus centralis, which contain terminals of 5-hydroxytryptamine and dopamine neurons. Metergoline increased 5-hydroxytryptophan and 3,4-dihydroxyphenylalanine accumulation, and decreased l-tryptophan concentration in a dose- and time-dependent manner. Pirenperone increased 5-hydroxytryptophan and 3,4-dihydroxyphenylalanine accumulation, but had no effect on l-tryptophan levels. These effects of pirenperone were time- and dose-related. Finally, pizotifen increased 5-hydroxytryptophan accumulation in a dose-related and time-dependent manner, but did not alter 3,4-dihydroxyphenylalanine or l-tryptophan concentrations. All the drug effects generally occurred in all four nuclei. These results suggest that 5-hydroxytryptamine receptor antagonists may affect synthesis in 5-hydroxytryptamine and/or dopamine neurons after l-tryptophan treatment and aromatic-l-amino acid decarboxylase inhibition.

Influence of Chlorpromazine on the Accumulation of 5-Hydroxytryptamine by Interaction with ATP

5-Hydroxytryptamine accumulates at pH 6.0 when ATP is present on one side of an artificial lipoidal barrier. In the presence of chlorpromazine, the diffusion of ATP through the barrier and a decrease in the accumulation of serotonin can be noted. The diffusion rate constants of ATP and serotonin are related to the concentration of chlorpromazine.

Platelet-induced neurogenic coronary contractions due to accumulation of the false neurotransmitter, 5-hydroxytryptamine.

The purpose of this study was to determine if 5-hydroxytryptamine released from aggregating platelets could be accumulated and released by canine coronary adrenergic nerves, and if the false neurotransmitter resulted in an abnormal response of the smooth muscle to nerve stimulation. Isometric tension was measured in rings of epicardial coronary suspended in organ chambers filled with physiological salt solution. The response to electrical stimulation or exogenously added norepinephrine was elicited after contraction with prostaglandin F2 alpha. Electrical stimulation and exogenous norepinephrine caused beta-adrenergic relaxation of control rings. However, after rings were exposed for 2 h to aggregating platelets or 5-hydroxytryptamine, electrical stimulation caused frequency-dependent contractions. These contractions were prevented by the serotonergic antagonists, cyproheptadine or ketanserin, or by the neuronal uptake inhibitor, cocaine. The relaxation caused by exogenously added norepinephrine was unchanged after exposure to platelets or 5-hydroxytryptamine, indicating that smooth muscle alpha- and beta-adrenergic responsiveness was unchanged. The electrically stimulated overflow of radiolabeled norepinephrine from superfused strips of coronary artery was not altered by prior exposure to 5-hydroxytryptamine, indicating that the effect of exposure on the response to electrical stimulation is primarily at smooth muscle serotonergic receptors. Canine coronary arteries accumulated and metabolized radiolabeled 5-hydroxytryptamine in vitro. The accumulation of 5-hydroxytryptamine was inhibited by cocaine or by adrenergic denervation with 6-hydroxydopamine but unaffected by removal of endothelium, indicating that the adrenergic nerves were the primary site of accumulation. Electrical stimulation of superfused strips of coronary artery preincubated with radiolabeled 5-hydroxytryptamine caused the release of the intact indoleamine; this was blocked by the neurotoxin, tetrodotoxin. These studies suggest that 5-hydroxytryptamine liberated from aggregating platelets may be accumulated by coronary adrenergic nerve endings. Upon its release from the nerves as a false transmitter, the amine can activate serotonergic receptors on the smooth muscle and reverse the action of the adrenergic nerves from dilator to constrictor.

5-Hydroxytryptamine transport in cells and secretory granules from a transplantable rat insulinoma.

Mechanisms of transport of 5-hydroxytryptamine in the pancreatic B-cell were investigated by using cell suspensions and secretory granules prepared from a transplantable rat insulinoma. (1) Cells incubated with 5-hydroxy[G-3H]tryptamine at concentrations ranging from 0.1 microM to 5 mM accumulated the radioisotope principally by a simple diffusion process. The incorporated radioactivity was recovered principally as the parent molecule and was recovered predominantly in soluble protein and secretory-granule fractions prepared from the tissue. (2) Isolated granules incubated in buffered iso-osmotic medium without ATP accumulated the amine to concentrations up to 38-fold that of the medium. This process was insensitive to reserpine and occurred over a wide range of 5-hydroxytryptamine concentrations (0.075 microM-25 mM). Above 5 mM, 5-hydroxytryptamine accumulation decreased in parallel with the breakdown of the delta pH across the granule membrane. Uptake was favoured by alkaline media and was reduced by the addition of (NH4)2SO4. In both cases a close correlation was observed between uptake and the transmembrane delta pH, a finding that suggested that 5-hydroxytryptamine permeated the membrane as the free base and equilibrated across the membrane with the delta pH. Binding of 5-hydroxytryptamine to granule constituents also played a part in this process. ATP caused a further doubling of granule 5-hydroxytryptamine uptake by a process that was sensitive to reserpine (0.5 microM). Inhibitor studies suggested that amine transport in this instance was linked to the activity of the granule membrane proton-translocating ATPase. (3) It was concluded that the uptake of amines driven by proton gradients across the insulin-granule membrane could account for the accumulation in vivo of amines in the B-cell.

The effects of some antipsychotic drugs, D-amphetamine, and reserpine on the concentration and rate of accumulation of tryptamine and 5-hydroxytryptamine in the mouse striatum.

The mouse striatum contains about 2 ng/g of tryptamine and 600 ng/g of 5-hydroxytryptamine. No significant changes in mouse striatal tryptamine were observed after the administration of chlorpromazine, haloperidol, spiperone, or alpha-flupenthixol. The levels of 5-hydroxytryptamine were moderately reduced by chlorpromazine, spiperone, and alpha-flupenthixol but not by haloperidol. The administration of antipsychotic drugs to mice pretreated with a monoamine oxidase inhibitor (pargyline) produced an increase in the rate of accumulation of striatal tryptamine compared with that of pargyline-treated mice. In contrast, the rate of accumulation of 5-hydroxytryptamine after monoamine oxidase inhibition was reduced by chlorpromazine, spiperone, and alpha-flupenthixol but not haloperidol. D-Amphetamine administration did not change either tryptamine or its 5-hydroxyderivative while reserpine increased tryptamine and reduced 5-hydroxytryptamine. The results suggest that changes in striatal tryptamine may be controlled by the availability of tryptophan, the amino acid precursor of tryptamine.

NMR studies of 5-hydroxytryptamine transport through large unilamellar vesicle membranes.

Nuclear magnetic resonance techniques developed to study membrane permeability in closed membrane systems have been used to investigate transport of 5-hydroxytryptamine across the phospholipid membranes of large unilamellar vesicles. The vesicles, modeling the 5-hydroxytryptamine storage organelles of blood platelets, contained a high internal level of ATP buffered at a pH low relative to the external solution. The resultant pH gradient drove accumulation of 5-hydroxytryptamine to a level consistent with selective transport of the neutral amine. The upfield shifts of the 5-hydroxytryptamine resonances resulting from complexation with internally confined ATP were utilized to resolve and, simultaneously, to observe the internal and external amine. Simulation of the time evolution of the 5-hydroxytryptamine concentration allowed measurement of a permeability coefficient of 1.4 +/- 0.5 X 10(-5) cm/sec for the neutral amine.

Action of immobilized neuraminidase on the accumulation of 5-hydroxytryptamine by human platelets.

Abstract The accumulation of 5-HT was studied in human platelets following graded desialylation with neuraminidase immobilized on macrobeads. Platelets were removed from the soluble sialylglyco-proteins of the plasma by gel filtration on Sepharose 2B; the uptake of 5-HT by gel filtered platelets over four hours was similar to that found using platelet rich plasma. Platelet aggregation induced by 10 −5 M 5-HT and 10 −6 M ADP was decreased following gel filtration and the use of Sepharose 2B insolubilized apyrase did not ameliorate this effect. Removal of up to 30 per cent of the platelet sialic acid, which results in changes in the glycoprotein pattern as demonstrated by two dimensional gel electrophoresis, appears to accelerate the rate of uptake of 5-HT. This is followed by a significantly decreased rate once 50% desialylation has been achieved. Native platelets and those from which 30 per cent of the sialic acid had been removed were refractory to resialylation with exogenous rat liver sialyltransferase though this enzyme preparation catalyses the resialylation of soluble asialoglycoprotein acceptor.

Kainic acid-induced wet dog shakes in rats

Following the intracerebroventricular administration of kainic acid (KA), rats showed wet dog shakes (WDS) in a dose-dependent manner. dl-α-aminoadipic acid and l-glutamic acid diethylester blocked WDS behavior induced by 0.05 μg of KA. Noradrenaline, clonidine, yohimbine and apomorphine also significantly blocked KA-induced WDS. Phentolamine and propranolol did not affect WDS. FLA 63, 6-OHDA lesion and bilateralis electrolytic lesion to locus coeruleus markedly enhanced, but l-Dopa blocked WDS behavior. Moreover, the KA-induced shaking behavior was blocked by α-methyl-p-trosine and haloperidol. Cyproheptadine and methergoline also blocked WDS. p-Chlorophenylalanine, 5,6-DHT lesion, electrolytic lesions to dorsal and medial raphe nuclei showed no effect on WDS behavior, but l-5-hydroxytryptophan efficiently blocked it. Atropine and morphine considerably blocked KA-induced WDS behavior, but pilocarpine and nalorphine showed no effect. Bicuculline significantly enhanced, but aminooxyacetic acid blocked WDS. Intracerebroventricularly administered KA dose-dependently decreased the concentrations of noradrenaline and dopamine in the whole rat brain. The brain concentration of 5-hydroxytryptamine was unchanged. In contrast, the concentration of 5-hydroxyindoleacetic acid increased. KA was ineffective regarding the GABA concentration and GAD activity. KA dose-dependently accelerated the disappearance of brain noradrenaline and dopamine after inhibition of catecholamine synthesis. KA, following inhibition of monoamine oxidase, increased the accumulation of 5-hydroxytryptamine, but failed to change the rate of decline of 5-hydroxyindoleacetic acid. KA failed to change the disappearance of brain 5-hydroxytryptamine after inhibition of its synthesis by PCPA. It is suggested that KA-induced WDS behavior is independent from the increased activity of serotonergic neurons in the central nervous system. KA-induced WDS appears to be under the inhibitory control of noradrenergic and GABA-ergic activity. The weaker inhibitory effect upon this behavior showed also dopaminergic and serotonergic neurons. The present experiments showed the close relationship between KA-induced WDS and shaking behavior in morphine abstinence, but basic differences in WDS behavior caused by excessive stimulation of serotonergic receptors.

Inhibition of adenosine triphosphatase, 5-hydroxytryptamine transport and proton-translocation activities of resealed chromaffin-granule 'ghosts'.

1. Highly purified resealed chromaffin-granule ;ghosts' were assayed for ATPase and ATP-driven H(+)-translocation and 5-hydroxytryptamine-uptake activities, and for 5-hydroxytryptamine uptake driven by an imposed transmembrane H(+)-gradient. The effects of several inhibitors on these activities were studied. 2. Dicyclohexylcarbodi-imide inhibits all of these activities, but not in parallel; at low concentrations it decreases the permeability of the membrane to protons. 3. 4-Chloro-7-nitrobenzofuran (Nbf-Cl) and silicotungstate inhibit ATP-dependent activities, without effect on 5-hydroxytryptamine uptake driven by an imposed H(+)-gradient. 4. Tributyltin chloride inhibits all of the activities. 5. Treatment of the ;ghosts' with low concentrations of urea inhibits 5-hydroxytryptamine uptake and ATP-dependent generation of a transmembrane H(+)-gradient, without inhibiting ATPase activity. 6. Nbf-Cl and silicotungstate are without effect on the rate of leakage of 5-hydroxytryptamine from preloaded ;ghosts', whereas dicyclohexylcarbodi-imide and tributyltin chloride accelerate the rate of leakage. 7. Treatment of the membranes with (14)C-labelled Nbf-Cl labels several proteins; membranes treated with dicyclohexyl[(14)C]carbodi-imide are labelled predominantly in a protein of low molecular weight, which may be analogous to the mitochondrial H(+)-conducting proteolipid. 8. It is concluded that Nbf-Cl and silicotungstate inhibit the H(+)-translocating ATPase of the granule membrane; that dicyclohexylcarbodi-imide inhibits the ATPase, and inhibits 5-hydroxytryptamine accumulation by accelerating leakage of the amine; and that the effects of tributyltin chloride are due to inhibition of the ATPase, and collapse of the transmembrane H(+)-gradient through OH(-)-anion exchange.

Lithium restores abnormal platelet 5-HT transport in patients with affective disorders.

Kinetic analysis of the transport of 5-HT into the blood platelets of depressed patients and recovered depressive patients has shown that the rate of accumulation of 5-hydroxytryptamine (5-HT) is significantly decreased both before and after recovery from the illness. This abnormality is corrected by both short and long-term lithium treatment. As a corollary to these studies, the effect of lithium in vitro on 5-HT uptake has been studied and the results are opposite to those reported in vivo. These findings suggest that lithium acts indirectly, and possible mechanisms of its action are discussed.
