Exploring the hepatoprotective effects of silymarin in experimentally induced- diclofenac sodium toxicity in rabbits

The study was aimed to investigate the hepatoprotective effects of varying doses of silymarin against liver damage caused by diclofenac sodium in rabbits. Silymarin, extracted from the Silybum marianum plant, is known for its antioxidant properties and is used to treat liver diseases. The rabbits (n=40) were divided into 4 groups. Group A served as the control (with vehicle solution), group B, as toxin control with diclofenac sodium (50mg/kg intra-peritoneally), group C received both diclofenac sodium and protective dose of silymarin (50mg/kg orally), while group D was administered diclofenac sodium along with higher dose of silymarin (100mg/kg orally). At the study's conclusion, rabbits were humanely euthanized and blood samples were collected for analysis. Enzymes such as Alanine Aminotransferase (ALT), Alkaline Phosphatase (ALP), Aspartate Aminotransferase (AST), direct bilirubin, serum urea, and total bilirubin were measured. The study revealed that higher dose of silymarin (100mg/kg) was more effective in reducing serum enzyme levels compared to lower dose, indicating stronger hepatoprotective effect. This suggests silymarin's potential as the therapeutic agent against liver damage and toxicity induced by substances like diclofenac sodium

The Circumglobal Transport of Massive African Dust and Its Impacts on the Regional Circulation in Remote Atmosphere

Abstract Atmospheric dust from North Africa, the largest and most persistently active dust source over the world, spreads widely in the Northern Hemisphere and plays essential roles in the Earth environment evolution. During 7–24 June 2020, an extremely strong dust event occurred with its westward spreading modulated by the North Atlantic Oscillation (NAO) and its eastward spreading regulated by European blocking, ultimately resulting in the circumglobal transport of African dust. The Mediterranean low pressure linked to the European blocking dipole was the key to facilitating the eastward transport of dust. This record-breaking African dust episode caused a notable diurnal precipitation decrease of 0.98 mm day−1 over northeastern India and a decrease of 1.55 mm day−1 over central North America, which was ascribed to the effect of dust-induced radiative heating on large-scale circulation. It triggered a Rossby wave train and caused anomalous high pressure over northeastern India, which weakened the India summer monsoon and consequently inhibited the occurrence of precipitation. Dust-induced radiative heating also supported the stability in the anomalous warm high over North America, further repressing import of moisture from Atlantic. Ambient moisture and atmospheric instability also presented consistent variation over North America and India characterized as strengthen descending motion and sharply reduced moist convection. This study reports, for the first time, the strong modulation of regional circulation by circumglobally transported African dust, especially in Asia and North America. The new aspects on the unexpected consequences on moisture convection indicate broader roles that the dust may play in the global climate change.

Association of genetic polymorphism of glutathione S-transferases with colorectal cancer susceptibility in snuff (Naswar) addicts.

The current study aimed to investigate the relationship between polymorphisms in detoxifying (GSTM1, GSTT1, and GSTP1) genes and their association with colorectal cancer (CRC) in tobacco addicts of Pashtun ethnicity. Polymorphisms in the selected genes were genotyped in a case-control study consisting of 100 histologically confirmed male CRC patients and 100 birth-year and gender-matched healthy controls using the PCR-RFLP method. The GSTM1 null, and GSTT1 null genotypes were significantly contributed to the risk of CRC in the cases (OR= 3.131, 95% CI: 1.451-6.758, P = 0.004, and OR= 3.541, 95% CI: 1.716-7.306, P = 0.001, respectively), whereas the association observed for GSTP1 Val/Val (1.139, 95% CI: 0.356-3.644, P = 0.826) did not show statistical significance. The combined GSTM1 null and GSTT1 null showed a 41-fold increased risk (95% CI: 4.945-351.950, P = 0.001), while, the combined GSTM1 null and GSTP1 Ile/Val or Val/Val variant genotypes exhibited about 3-fold (95% CI: 1.196-7.414, P = 0.019) increased risk to CRC. Similarly, the combined GSTT1 null and GSTP1 Ile/Val or Val/Val variant genotypes showed about a 3-fold (95% CI: 1.285-8.101, P = 0.013) increased risk of CRC. In the combination of three GST genotypes, the GSTM1 null, GSTT1 null, and GSTP1 Ile/Val or Val/Val variant genotypes demonstrated a more than a 22-fold (95% CI: 2.441-212.106, P = 0.006) increased risk of CRC. Our findings suggest that GSTM1 and GSTT1 polymorphism and its combination with GSTP1 may be associated with CRC susceptibility in the Naswar addicted Pashtun population of Khyber Pakhtunkhwa, Pakistan.

Anti-hyperglycemic and anti-hyperlipidemic effects of a methanolic extract of Debregeasia salicifolia in Alloxan-induced diabetic albino mice.

Diabetes mellitus (DM), an endocrine syndrome characterized by high blood glucose levels due to abrogated insulin activity. The existing treatments for DM have side effects and varying degrees of efficacy. Therefore, it is paramount that novel approaches be developed to enhance the management of DM. Therapeutic plants have been accredited as having comparatively high efficacy with fewer adverse effects. The current study aims to elucidate the phytochemical profile, anti-hyperlipidemic, and anti-diabetic effects of methanolic extract D. salicifolia (leaves) in Alloxan-induced diabetic mice. Alloxan was injected intraperitoneally (150 mg kg-1, b.w), to induced diabetes in mice. The mice were divided into three groups (n=10). Group 1 (normal control) received normal food and purified water, Group II (diabetic control) received regular feed and clean water and group III (diabetic treated) received a methanolic extract of the plant (300 mg kg-1) for 28 days with a typical diet and clean water throughout the experiment. Blood samples were collected to checked serum glucose and concentration of LDL, TC, TG. The extract demonstrated significant antihyperglycemic activity (P<0.05), whereas improvements in mice's body weight and lipid profiles were observed after treatment with the extract. This study establishes that the extract has high efficacy with comparatively less toxicity that can be used for DM management.

Tracing probiotic producing bacterial species from gut of buffalo (Bubalus bubalis), South-East-Asia.

Due to extensive application of antibiotics as growth promoters in animal feed, antimicrobial resistance has been increased. To overcome this challenge, rumen microbiologists search for new probiotics to improve the rate of livestock production. The present study was aimed to isolate and evaluate breed-specific lactic acid bacteria (LAB) as potential animal probiotics. The current study was conducted during 10 months from July 2020 to April 2021, in which a total of n=12 strains were isolated from different samples including milk, rumen, and feces of Nilli Ravi Buffaloes. These isolates were evaluated for their antimicrobial potential against common animal pathogens (Bacillus spp., E. coli, Staphylococcus aureus, Salmonella spp., Listeria spp.). All the isolates were identified using 16S rRNA gene sequencing and the phylogenetic analyses inferred that these strains showed close relations to the species of various genera; Enterococcus lactis, Pediococcus pentosaceus, Bacillus subtilis Weissella cibaria, Weissella soli, Bacillus tequilensis, Weissella bombi, Bacillus licheniformis, Lactococcus lactis, Bacillus megaterium, Lactobacillus ruminis, and Lactococcus lactis. NMCC-Ru2 has exhibited the enormous potential of antimicrobial activity, 28 mm, for Salmonella typhimurium;23 mm for Listeria monocytogenes 21 mm for E.coil. Highest resistance was seen in NMCC-Ru2 agasint test antbiotic, like 25.5 mm for Tetracycline. Overall results revesl that the probiotic profile of isolates was achieved using standard criteria, particularly with animal probiotic properties.

Brassica oleracea L. (Acephala Group) based zinc oxide nanoparticles and their efficacy as antibacterial agent.

Zinc oxide nanoparticles were synthesized from the leaf extract of Brassica oleracea L. Acephala group (collard green) followed by their characterization using Scanning Electron Microscope (SEM), and Energy Dispersive X-ray (EDX). The antibacterial properties of zinc nanoparticles were tested against Gram-negative bacteria, Pseudomonas aeruginosa (ATCC ® 9027™), Escherichia coli (ATCC ® 8739™), Klebsiella pneumoniae (ATCC® BAA-1705™) and Gram-positive bacteria, Staphylococcus aureus (ATCC ® 6538™) and Listeria monocytogenes (ATCC ® 13932™), at four different concentrations (50.00 µg/ml, 100.00 µg/ml, 500.00 µg/ml and 1 mg/ml) of zinc oxide nanoparticles suspension. Results revealed that the synthesized nanoparticles exhibit strong antibacterial effects against Pseudomonas aeruginosa, Listeria monocytogenes, Klebsiella pneumonia, Staphylococcus aureus and Escherichia coli at 500.00 µg/ml-1 mg/ml concentrations. An increase in efficacy of nanoparticles with the decrease of their size was also evident. This is a first ever report on Brassica oleracea, L. based nanoparticles which demonstrates that 500.00 µg-1 mg/ml conc. of zinc oxide nanoparticles have antibacterial activity against both Gram -ve and Gram +ve bacteria and have the potential to be considered as an antibacterial agent in future.

Ethnomedicinal uses of plants for various diseases in the remote areas of Changa Manga Forest, Pakistan.

This study aims at reporting the indigenous knowledge of the medicinal flora from the inhabitants of surroundings of the World's largest artificial planted forest "Changa Manga", Pakistan. Data were collected by direct interviews and group meetings from 81 inhabitants including 32 local healers having information regarding the use of indigenous medicinal plants over a period of one year. Different statistical tools were applied to analyze the data including Frequency citation (FC), Relative frequency citation (RFC), Use Value, Factor of informants consensus and fidelity level. This study reported 73 plant species belonging to 37 plant families and 46 genera. The majority of plant species belong to compositae family. The most commonly used medicinal plants were P. hysterophorus L., P. dactylifera L., S. indicum L, P. harmala L., P. emblica L., and A. indica A.Juss. The greatest number of species was used to cure gastrointestinal disorders. The highest fidelity level (68.18%) was of E. helioscopia to cure gastrointestinal disorders. Maximum fresh uses (17) were reported by C. dactylon (L.) Pars. While the highest number of species reporting fresh uses in similar number was 13. In this study, five novel plants are being reported for the first time in Pakistan for their ethnomedicinal worth. Our data reflect unique usage of the medicinal plants in the study area. The statistical tools used in the study proved useful in pointing the most important and disease category specific plants. High use value plant and the new reported medicinal plants might prove an important source of the isolation of pharmacologically active compounds.

Species diversity pattern of mosquitoes (Diptera: Culicidae) breeding in different permanent, temporary and natural container habitats of Peshawar, KP Pakistan.

To determine the species composition, relative abundance and seasonal variation of different mosquitoes Genera (Aedes, Anopheles, Armigeres, Culex, and Culiseta) in different habitats the present research work was carried out in Entomology Research Laboratory of The University of Peshawar. Sampling performed from variety of permanent and temporary breeding habitats was carried out on monthly basis from targeted breeding sites for two consecutive years through dipping method. Species diversity in the survey sites was noted. Collection from these seventeen various types of potential larval habitats, yielded a total of 42,430 immature constituting 41,556 larvae and 874 pupae. Among these only 19,651 adult mosquitoes emerged comprising 11,512 female and 8,139 male mosquitoes. 78% (n= 15333) of mosquito larvae were from permanent and 22% (n=4318) were from temporary breeding sites. This study showed that Peshawar valley harbours 15 species from the genera Aedes, Anopheles, Armigeres, Culex and Culiseta. When the density of each species was examined, Culex quinquifasciatus was found to be dominant (79%) and constant in distribution. Among the temporary habitats Aedes albopictus was found as the most prevalent species particularly from tree holes and water cisterns. The highest intensity of mosquitoes was in June (2243 emerged adults) and November (2667 emerged adults) while the lowest was in January (203 emerged adults). A perfect positive correlation (r = +0.8) was found between temperature and population of mosquitoes (df 10 and α 0.05). The species diversity index for mosquitoes remained between 0.12 and 1.76. The Margalef's richness components was noticeably low for bamboo traps (0.2) and fairly high for rice fields, Percolating water and Animal tracks (1.3) which shows the abundance of mosquito species in these habitats. Similarly Pielou's Evenness was highest for bamboo traps (E=1) showing species uniform distribution. Animal tracks were presumed not only the diverse habitat rather also possessed high value for species richness and species evenness. Temperature, rainfall, humidity and other related attributes responsible for species variation and abundance need to be analysed further to pave way for controlling vector species in their oviposition targeted sites.
