Abstract

A modified alkaline sucrose gradient technique for the fractionation of single-stranded mammalian DNA has been developed. The method involves the use of a reorienting gradient zonal centrifugation system of large sample zone capacity. The data obtained support the concept that the DNA in mammalian chromosomes exists as a linked array of subunits (10).
