Abstract

Introns from eukaryotic pre-mRNA transcripts are removed by the spliceosome. Splicing is accomplished only after the spliceosomal RNA active site is formed from snRNAs, pre-mRNA, and several proteins. Base pairing between U4 and U6 snRNAs is disrupted during activation, allowing for a conformational change in the U6 snRNA necessary for catalysis at later stages of splicing. The process by which the U4/U6 duplex base pairing contacts are destabilized during activation is unknown but results in irreversible release of U4 with its associated proteins.
