Abstract
The objective of the study was to vitrify mouse embryos with the cryoloop technology using a new combination of vitrification mediums. Embryos were exposed to a 2- step loading of CPA, ethylene glycol and propylene glycol, before being placed on the surface of a thin filmy layer formed from the vitrification solution in a small nylon loop. After warming, the CPA was diluted out from the embryos by a 3-step procedure. Our data show that a high percentage of embryos survived (92.7%) vitrification in the mixture of EG and PG combined with cryoloop carrier and developing normally (89.1%) in vitro after thawing.

The objective of the study was to vitrify mouse embryos with the cryoloop technology using a new combination of vitrification mediums. A tradicionálisnak mondható „lassú mélyhűtés” mellett az embrió mélyhűtés egy másik változata a vitrifikáció, amikor a sejtek jégkristályképződés nélkül fagynak meg. A fagyasztóoldatnak nagy mennyiségben kell tartalmaznia intra- és extracelluláris védőanyagok keverékét, 2. A sejteket kis térfogatú oldatban kell mélyhűteni (néhány μl), 3. Az utóbbi években a hűtési sebességet sikerült nagymértékben növelni, azáltal, hogy a védőoldat térfogatát csökkentették, ami annak köszönhető, hogy új típusú szállító/hordozó eszközöket fejleszttettek ki (OPS straw: Vajta és mtsai, 1997; Oberstein és mtsai, 2001; flexipet-denuding pipetta: Liebermann és mtsai, 2002; hemi-straw: Kuwayama és Kato, 2000; nejlonháló: Matsumoto és mtsai, 2001; nejlonhurok: Lane és mtsai, 1999; Oberstein és mtsai, 2001; SSV: Dinnyés és mtsai, 2000). Ennek eredményeként az embriók/petesejtek nagyon kis mennyiségű védőoldatban fagyaszthatók
