Abstract

The colorimetric determination of Kjeldahl nitrogen has been modified. The method utilizes the addition of Nessler's reagent directly to the Kjeldahl digest. As a color stabilizer 0.015 M KCN is added to the Nessler reagent of the Koch-McMeekin formula. Three to thirty micrograms of nitrogen can be conveniently determined by this method with a standard deviation of less than 2% of the mean. The entire procedure is carried out in a test tube and the method is particularly suited to running large mumbers of samples. An application of the method to the micro determination of nitrogen in proteins, lipoprotein, and other amide-containing molecules is also presented.
