Abstract
The theory and experimental protocol for distinguishing between various three-substrate enzymic mechanisms by use of alternative substrates and isotopic substrates is presented in this paper. The rate equations describing alternative substrate inhibition and isotope competition are presented for 10 different three-substrate enzymic mechanisms. Various mechanisms can be distinguished by evaluation of their inhibition patterns.
