Abstract

This study was designed to evaluate the effects of leukocytes contamination determined by peroxidase-positive leukocytes on semen characteristic of leukocytospermic subjects. Fifty (25 leukocytospermic vs. 25 non leukocytospermic) infertile couples collected from Al-Hussein Teaching Hospital/Thi-qar Health Directorate enrolled in this study and semen samples were analyzed before and after in vitro sperm preparation technique. Furthermore, sperm concentration, sperm motility, progressive sperm motility, normal sperm morphology, and seminal leukocytes counts were evaluated according to standard WHO criteria (2010). For preparation technique, sperm prepared and incubated for 30 minute in 5% CO2 at 37oC. The results of present study shows that leukocytospermic men significantly (P<0.001) decreased and differences sperm functions compared with non leukocytospermic men may be due to reactive oxygen species generation by leukocytes. It was concluded that the increase seminal leukocyte counts lead to impaired sperm parameters and decrease fertilizing ability of human spermatozoa and sperm function improved and augmented after sperm processing technique. Further studies are recommended to assess the harmful outcomes of ROS on DNA damage and embryo quality after intracytoplasmic sperm injection (ICSI-ET).
