Abstract

The guinea pig polymorphonuclear leucocytes were incubated with rabbit IgG double labeled with 131I and fluorescein isothiocynate. The amount of ingested protein was sufficiently great (approximately 10 7 molecules per cell) to be detected by radioimmunodiffusion . The IgG fixed was distributed in all the subcellular fractions, the highest percentage being associated with the leucocytic ‘granules’. At this level the IgG was hydrolysed up to the stage of the Fab fragment. The Fc fragment was not pointed out here, nor the trichloracetic acid-soluble radioactive peptides. The action of the ‘granular’ fraction was found to be identical in vitro also.
