Abstract

An unusual and heretofore undescribed alteration of rough ER membranes of rat hepatic cells after CCl4 injury has been observed. The lesion is detectable as early as 30 min after oral administration of CCl4 and is fully developed within 2 to 5 h. It is characterized by the detachment of ribosomes, narrowing of cisternal spaces, fusion of membranes and eventual collapse. By 10 and 20 h, large masses of smooth vesicular membranes with deposition of electron-opaque flakes were observed. These masses appear to have a direct connection to the altered rough ER. The significance of these lesions in respect to the mechanism of CCl4 induced injury is briefly discussed.
