Abstract

UDP-glucose:1,3-β-glucan synthase was recovered in a particulate fraction from potato tuber tissue ( Solanum tuberosum L. cv. Eba) (D.G. Robinson and H. Quader (Eds.), Cell Walls 81, Wiss. Verlags GmbH, Stuttgart, 1981, p. 84). This enzymatic activity was promoted by digitonin and partially solubilized by 0.5% of this detergent. Other detergents tested were detrimental to the enzymic activity. The activity of the particulate fraction and of the solubilized preparation was very labile. Eighty percent of the solubilized activity was lost after 2 h at 25°C. The stability of the enzyme preparation could be improved by a stepwise extraction with digitonin. The partially lost activity of the particulate fraction and the solubilized enzyme preparation could be restored by the addition of lipids extracted from tuber tissue or from the particulate fraction.
