Abstract

• Two new eremophilane-type sesquiterpenes were isolated from the fresh roots of Rehmannia glutinosa . • The structures were established based on spectroscopic data analyses. • Compounds 1 and 2 exhibited protective effects against LPS-induced damage to NRK-52e cells. Two new eremophilane-type sesquiterpenes, remophilanetriol A ( 1 ) and remophilanetriol B ( 2 ), together with two known compounds, diincarvilone C ( 3 ) and diincarvilone D ( 4 ), were isolated from the fresh roots of Rehmannia glutinosa . The structures were determined based on analysis of the IR, UV, MS, and NMR data. The absolute configurations of the new compounds were resolved by comparing their experimental and calculated ECD spectra. All compounds were tested for their protective effects on normal rat kidney tubule epithelioid (NRK-52e) cells injury induced by lipopolysaccharide (LPS). The results showed that compounds 1 and 2 exhibited protective effects against LPS-induced damage to NRK-52e cells.
