Abstract

Objective To analyze the proteomic components of the serum from the patients with lung adenocarcinoma and healthy donors, in search of the specific tumor markers of lung adenocarcinoma for diagnose. Methods The serum total protein of human lung adenocarcinoma serum and healthy donors serum were separated by immobilized pH gradient based on two-dimensional polyacrylamide gel electrophoresis(2-DE).The differentially expressed proteins were analyzed using PDQuest analysis software, then identified using MALDI-TOF-MS and Mascot differentially expressed in serum from lung adenocarcinoma patients and healthy donors. Seven protein spots of interest were incised from gel and digested in-gel by trypsin, seven PMF maps were obtained by MALDI-TOF-MS.The typical peptide masses were searched in the NCBInr database by Mascot software,one protein was identified succesfully.It was serum amyloid A(SAA). Conclusions two-dimensional polyacrylamide gel electrophoresis to separate and identify the between lung adenocarcinomma patients and healthy donors.It will be useful for biomark of lung cancer.
