Abstract

In the current study, Trilazad mesylate-loaded cellulose acetate scaffolds were developed in order to promote diabetic wound healing. Trilazad mesylate at 3 different concentrations of 1% w/w, 3% w/w, and 5% w/w was loaded into polymeric solution of cellulose acetate and then electrospun. The produced scaffolds were studied regarding their microstructure, cell proliferation, water uptake capacity, mechanical properties, hydrophobicity, gas exchange, drug release, hemocompatibility, porosity, antibacterial property, anti-oxidant activity, cytoprotection, and protein adsorption. Cellular studies showed that the scaffolds loaded with 3% w/w Trilazad mesylate had the highest cell proliferation and cell viability under normal and oxidative stress conditions. Therefore, this concentration was chosen for treating diabetic wounds. Wound healing study showed that drug-loaded dressings could significantly improve the level of wound healing and could significantly reduce the gene expression profile of Glutathione peroxidase and NFkB genes. These preliminary results suggest that Trilazad mesylate-loaded cellulose acetate wound dressings have contributed in diabetic wound healing through modulation of immune response and protection against oxidative damage.
