Abstract

Objective To study curative effects and mechanism of bone marrow stromal cells (BMSCs) transplantation on rat models of Parkinson's disease (PD). Methods BMSCs were induced into TH-positive cells, which were stained by immunofluorescence with TH and detected by flow cytometry. The 6-hydroxydopamine (6-OHDA) induced hemiparkinsonian rats were selected and allocated randomly into 3 greups,which were. BMSCs labeled with brdu induced into TH positive cells (1.2×106 cells/each model), BMSCs without induced and physiological saline (12 μl) were injected into right striatum of PD rat respectively. The rotation test was taken at 6 week after transplantation. Brain frozen sections were car-fled out immunofluorescence double staining (BrdU/GFAP,BrdU/NSE and BrdU/TH) at 6 week of BM-SCs. Meanwhile the level of dopamine in nigrostriatal system were observed by High-performance liquid chromatography. Results The percentage of TH-positive cells was 10.3% by detecting with flow cytome-try. Frequency of changed rotation were significantly more in induced than non-induced BMSCs group and normal saline group at 6 week after transplantation. Some BrdU/NSE, BrdU/TH positive cells could be found in the implanted region of BMSCs induction group at 6 week after transplantation. At this time, Br-dU/NSE and BrdU/TH positive cells are not found in the implanted region of BMSCs group but there are BrdU/GFAP positive cells, the level of dopamine in nigrostriatal system in the implanted region of BMSCs induction group at 6 week after transplantation, were more than BMSCs group observed by High-perform-ance liquid chromatography. Conclusion BMSCs induced into TH-positive cells had better effects on treating PD rat models than that of BMSCs without induced group, and physiological saline group.
