Abstract
Lens mRNA obtained from polysomal RNA was further purified by either poly(rU)-agarose, oligo(dT)-cellulose or cellulose chromatography. Stimulation of [ 3H]leucine incorporation into CCl 3COOH-precipitable material was obtained with mRNA isolated with all of the chromatographic media. The mRNA was shown to direct the production of the polypeptide chains of alpha-crystallin and also served as templates for the synthesis of complementary DNA (cDNA) in the presence of oligo(dT) primer and avian myeloblastosis virus (AMV) RNA-dependent DNA polymerase. The cDNA products were found to have sedimentation constants of 7·68 and 8·25 and hydridized to their respective lens mRNA template giving a RNA-DNA hybrid of buoyant density, 1·56 g/ml.
