Abstract

Susceptibility to insulin dependent diabetes mellitus in the diabetes prone BB (BB-DP) rat is associated with lymphopenia, which is characterized by a lack of the T cell subset expressing the differentiation marker RT6. RT6 is an alloantigenic system characterized by two highly polymorphic alleles ( RT6 a and RT6 b ) of a single gene on chromosome 1, whose expression is restricted to T cells during a post-thymic stage of development, as well as to intraepithelial lymphocytes and a subset of natural killer cells. The RT6 a gene of BB-DP rats is intact, and lymphopenia has been shown to be due to a defect in a so far uncharacterized gene ( Lyp) mapped to chromosome 4. To determine whether RT6 expression is influenced by the Lyp gene product, we are currently engaged in characterizing the regulatory elements of the RT6 gene.
