Abstract

Objective To detect the expression level of leucine-zipper tumor suppressor 1 (LZTS1) in normal renal tissues and renal cell carcinoma tissues and evaluate its significance.Methods Twenty-eight cases of normal renal tissues and renal cell carcinoma tissues were harvested.The expression level of LZTS1 mRNA was detected by reverse transcriptase-polymerase chain reaction (RT-PCR),realtime reverse transcriptase-polymerase chain reaction (RT-qPCR) and in situ hybridization (ISH).Results The relative level of LZTS1 mRNA in renal tumor group was (0.39 ± 0.04,n =28) by RT-qPCR via 2-△△ct method.Meanwhile,the relative level of LZTS1 mRNA in normal renal tissue group was (1.40 ± 0.06,n =28).The expression level of LZTS1 mRNA in renal tumor tissue was down-regulated as compared with that in normal renal tissue with the difference being significant (P < 0.05).The in situ hybridization showed only less brown precipitate in renal tumor tissue than in normal renal tissue.The positive expression rate of LZTS1 mRNA was 92.9% (26/28,n =28) in normal renal tissue,and that was 14.2% (4/28,n =28) in renal tumor tissue.The results of ISH were consistent to the PCR results.Conclusion The loss of LZTS1 maybe participate in the molecular process of carcinogenesis in renal cell carcinoma.
