Event Abstract Back to Event Transcription factors involved in the development of Purkinje cell dendrites Isabelle Dusart1* 1 CNRS UMR7102, UPMC, France Purkinje cells (PCs) in the cerebellum generate a very peculiar dendritic tree: very large, highly branched and planar. During the first two postnatal weeks, mouse PCs undergo a 2-phase remodelling process of their dendrites. The first phase is characterized by the retraction of the primitive dendrite and the formation of multiple small processes arising from the cell body, disappearing with the formation of the mature dendrites. In the second phase, the mature dendrites grow rapidly and branch out.In order to identify TFs involved in the dendritic development of PCs, we analyzed the gene expression profile of a cerebellar compartment enriched in PCs. Comparing the transcriptome at different maturation stages; we found that several TFs were differentially expressed during the two phases of dendritic remodelling. Six of them were validated by quantitative RT-PCR and single cell RT-PCR analyses. Among these factors, Ebf1 and Ebf2 were expressed during the first phase of PC dendritic remodelling and downregulated in the second phase. To investigate the contributions of these factors in dendritic development, we generated lentiviral vectors allowing overexpression in organotypic cultures counteracting their developmental downregulation. We show that overexpression of Ebf2 maintained PCs in a morphologic immature stage inhibiting the retraction of the primitive dendritic tree whereas Ebf1 overexpression in PCs resulted only in minor effects on dendritic development.
