Abstract

HeLa cells were exposed to dimethyl sulfoxide (DMSO) at +4 and at +22°C for periods of 30, 60, 90, and 120 min. At concentrations of DMSO below 15% there appeared to be no significant difference in the two temperatures of exposure as measured by confluency of cultures at the end of 96 hrs. Exposure at the lower temperature, however, did permit survival of cells in the higher concentrations of DMSO. The morphological effects of DMSO as examined with coverslip preparations stained with Maximow's hematoxylin eosin-azure II are also described. Pathological changes consisted of a typical mitotic figures, giant cells, and cells with ragged basophilie pseudopod-like projections. The number of abnormal cells increased with an increase in concentration of DMSO. The percentage of necrotic cells increased with an increase of DMSO concentration almost independent of the time of exposure or the time of subsequent culture.
