Abstract

Objective To investigate the toxic effects in human lung carcinoma cell line A549 in-duced by methyl methacrylate (MMA) monomer.Methods A549 cells were divided into five concentration of MMA groups ( 1,10,40,80 and 120 mg/L) and control group.The A values of different groups were measured hy using methyl thiazol tetrazolium (MTT) assay at 30 min,1st day,3rd day and 5th day after administration.Annexin V/PI method was used to test the percentage of apoptosis and necrosis at let day after administration.Results There were significant differences in A value in 10 mg/L group at 5th day,and 40,80 aud 120 mg/L groups at 1st,3rd and 5th day when compared to control group (P <0.05).The percentage of apoptosis and necrosis at 10,40,80 and 120 mg/L groups were (1.78 ±0.09)%,(3.97 ±0.22)%,(4.58 ±0.36)% and (5.27 ±0.14)%,and (3.43 ±0.08)%,(6.70 ±0.21 ) %,( 7.93 ± 0.40 ) % and ( 9.51 ± 0.64 ) %,respectively,which was significantly different from that in control group [(1.14±0.08)% and (1.24±0.09)%] (P<0.05).Conclusion MMA monomer has cytotoxicity on A549 cells.
