Abstract

Abstract Two methods used to induce protoplast fusion, involving polyethylene glycol (PEG) and high calcium/high pH, respectively, have been investigated in procedures for the inoculation of isolated tobacco protoplasts with TMV. It has been shown that inoculation in the presence of fusion agents involves two stages. Firstly, virus precipitation on to the protoplast surface and secondly, entrapment of virus particles between protoplasts in zones (putatively) of transient protoplast fusion.
