Abstract
The time course of physiological disposition of 3H-labeled 7-hydroxychloropromazine and 8-hydroxychlorpromazine has been examined in rats after a single dosage, and the accumulation of each compound has been studied after 6,14 and 28 doses on a bid schedule. The 8-hydroxycompound decays at a slower rate and shows a greater degree of accumulation on repeated dosage than does the 7-hydroxychlor-promazine. Both compounds enter the brain (the 7-hydroxy to a greater extent) and distribute uniformly, with no significant overt behavioral effects. Both compounds caused significant liver and kidney damage as demonstrated by histological evaluation.
