Abstract

(Nuclear maturity of bovine oocyte after 24 hours ovary preservation)ABSTRACT. The objective of the research was to investigate their meiotic competence or nuclear maturity of bovine oocytes maturity in vitro after 24 hour preservation on 5°C. Oocytes were collected by slicing the ovaries in modified phosphate buffer saline (m-PBS). Selected cumulus-oocyte complexes (COCs) homogenous ooplasm were cultured in maturity medium at 38°C in humidified atmosphere of 5% CO2 incubator. After 24 hours, oocytes stained for nuclear maturity’s evaluation. The proportion of oocytes at metaphase II (MII) was significantly difference (P 0.05) on oocytes that 24 hours preservated (44.21 ± 3.04%) vs oocytes from fresh ovary (73.97 ± 9.32%) (P0.05). These results indicated that 24 hour’s preservation bovine’s ovary on 50C cause decreases of nuclear oocyte maturity.

MATERI DAN METODE

Media koleksi adalah NaCl fisiologis ditambah penisilin 100 IU/ml, streptomisin 100 μg/ml. Koleksi ovarium dan koleksi oosit Ovarium sapi yang diperoleh dari RPH lokal disimpan dalam termos dengan media koleksi (NaCl fisiologis) suhu 30-35°C. Ovarium dicuci dengan media koleksi, oosit dikoleksi dengan metode slicing dan dicuci 3 kali, pencucian terakhir dengan media pematangan TCM 199. Setelah disimpan selama 24 jam pada suhu 4°C ovarium dicuci dengan media koleksi. Gelas obyek yang berisi oosit tersebut dimasukkan ke dalam larutan fiksasi yang mengandung asam asetat dan Tingkat Kematangan Inti Oosit Sapi Setelah 24 Jam Presevasi Ovarium (drh.Rini Widyastuti, M.Si dan Dr.agr.Siti Darodjah Rasad, M.S). Evaluasi tingkat kematangan inti yang diamati pada penelitian ini adalah dengan cara menghitung jumlah oosit pada setiap pembelahan meiosis mulai dari Germinal Vesicle (GV), Germinal Vesicle Break Down (GVBD), Metafase I (M-I) dan Metafase-II (M-II).
