Abstract

abstract This chapter is divided into two sections. The first section describes protocols to produce eggs and embryos of Xenopus laevis to investigate the organization and composition of tight junctions (or other structures). The second part describes approaches to study interactions between tight junction (or other) proteins using recombinant proteins and lysates of cultured epithelial cells (from Xenopus or mammalian organisms), reticulocytes, and insect cells. Combining studies on protein expression in vivo with studies on protein interactions in vitro can help to elucidate the molecular mechanisms involved in tight junction biogenesis. The reader is referred to review articles (1–5) for a suitable introduction to the structure, function, and molecular composition of epithelial tight junctions.
