Abstract

Abstract The influence of a finite source and detector on the three-dimensional imaging of an incident light fluorescent confocal scanning optical microscope is investigated on the basis of its three-dimensional optical-transfer function. It is shown that the optical resolution in such set-ups remains the same as with punctiform pinholes if the source and the detecting pinhole radii are both smaller than the size of the Airy disc.
