Abstract

The maintenance of cell cultures on Nucleopore polycarbonate filters during growth, embedding, sectioning, and viewing has proved to be a very useful technique for in situ ultrastructural observations. The greatest advantages of this substrate are sufficient transparency to permit observation of living cells prior to fixation, choice of either a smooth or a rough surface as substrate, highly regular channels of even bore, and good stability in the electron beam.
