Abstract

• Single molecule FRET studies reveal details of the folding of STIM1 in its resting state and unfolding activated by Ca 2+ store-depletion. • STIM1 is folded with SOAR inverted and held between the CC1α helices in a domain-swapping configuration close to the ER membrane. • Store-depletion causes the STIM1 TM-helices to bind together, squeezing SOAR from between the CC1α1 helices, allowing it to flip up and activate Orai channels SEE ABSTRACT in Query Response
