Abstract

Objective To approach the role and expression of IL-21/STAT3 pathway in the pathogenesis of murine with ulcerative colitis(UC).Methods 20 BALB/C mice were randomly divided into normal group and DSS model group,10 mice in each group.UC mice were induced by drinking dextran sodium sulfate solution. The mouse disease activity index score(DAI score)was calculated.Mice were killed at the eighth day,the colon tissues were conducted with pathological section and score, serum content of IL-6 and IL-21 were assayed by Elisa kits, the expression of STAT3 protein in colonic mucosal was detected by immunohistochemical method. Results The score of DAI and histological damage score, serum IL-6 and IL-21 content and the expression level of STAT3 in colonic mucosal were higher in model group than those in normal group(P < 0.05),the difference had statistical significance(P < 0.05).Conclusion The serum contents of IL-6 and IL-21 and the expression level of STAT3 intestinal tract were increased in mouse with UC,we presume that IL-21/STAT3 pathway may participate in the process of UC,blocking IL-21/STAT3 pathway in illness intestinal tract and reducing STAT3 expression may be the drug treatment target of UC. Key words: Ulcerative colitis(UC); Disease activity index; Interleukin21; Dextran sulfate sodium
