Abstract
Objective To explore the method of separating gastric epithelium mucosae cells and analyze the cell membrane proteins. Methods Get rid of the mucus of the gastric tissues by 0. 1% pepsin solution, scrape the mucous membrane, shear the tissues by mechanical method and then after filtering and centrifugation, the gastric epithelium mucosae cells was obtained. Extract membrane proteins by Triton-114,perform SDS-PAGE electrophoresis and choose some protein straps to do mass spectrography analysis. Results We obtained gastric epithelium mucosae cells successfully,the cell plasma membrane proteins were separated from total proteins, and we acquired some membrane proteins information by mass spectrography analysis. Conclusion Gastric epithelium mucosae cells can be separated by the combination of physical and chemical methods,plasma membrane proteins can be extracted by lyses buffer containing Triton-114.
