Abstract

A microsomal ATPase activity isolated from etiolated wheat leaf protoplasts required Mg 2+ and was stimulated by K + . The activity showed a pH optimum of about 6.5 with Mg 2+ , while the K + -stimulated activity was optimal at pH 5.5–6.0. The ATPase was inhibited by 50 μM vanadate. Vanadate inhibited the swelling response of protoplasts to red light, and also the reduction in size following transfer of protoplasts from 0.5 M to 0.6 M sorbitol. However no consistent effects of red light on microsomal ATPase could be detected, either when light treatment was applied to the protoplasts before membrane isolation or to the isolated membrane fraction.
