Abstract

Objective To explore the roles of microRNA (miR)-21 in the thyroid papillary carcinoma (TPC-1) cells. Methods Transit in the TPC-1 transfection of miR-21,Immunohistochemical detection of programmed cell death 4 (PDCD4) expression after transfection;transfected with miR-21 TPC-1 cell proliferation measured by tetrazolium (MTT) assay; flow cytometry detection of transfected TPC-1 cells withereddie. Results By transfection group were detected by immunohistochemistry, cytoplasmic brown color, the control group showed no staining, transfected group programmed cell death 4 (PDCD4) expression was significantly higher than the methyl thiazol MTT assay transfected cell viability was significantly be promoted (P < 0.05).Transfection group apoptosis rate was (3.13 ± 0.34) %,and a blank group, the apoptosis rate of the empty plasmid group (8.03 ± 0.02) % (9.51 ± 0.54) % (P < 0.05),apoptosis of the transfected cells is significantly reduced. Conclusion PDCD4 is a miR-21 target genes of high expression of miR-21 can promote the development of papillary thyroid carcinoma. Key words: Thyroid carcinoma; MicroRNA-21 ; Programmed cell death 4; Apoptosis
