One of the biggest pending issues in clinical medicine is the bone injury caused by trauma, cancer, deformity, etc. Research suggests that the periosteum around epiphysis is a dense connective issue membrane. It has a dynamic structure and not looks like what it is seemingly. It provides favorable living environment for stem cells with regeneration potential. Besides, it also plays an important role in bone nutrition, cell supply, pain sensation and growth repair. Therefore, deep study in periosteum structure and function, as well as in periosteum response mode will benefit the clinical application of bone defect repair. Key words: Bone injury; Bone defect repair; Osteogenesis; Periosteum; Periosteal reaction
