Abstract

Objective To investigate the relationship of bone morhogenetic proteins 4 (BMP4)and chemotherapy sensitivity in glioma.Methods The tissue microarry (TMA) of human brain astrocytoma was constructed and analyzed for the expresion of BMP4,B lymphocytes/leukemia-2 (bcl-2) and MnSOD by using SP immunohistochemistry.Temozolomide (TMZ) resistant cell line was constructed and the expression of BMP4 was detected.BMP4 plasmid was transfected into the TMZ resistant cells,and chemotherapy sensitivity was tested.Results The average absorbance of BMP4 ( WHO Ⅰ:0.3320 ± 0.0065,WHO Ⅱ:O.2485 ±0.0066,WHOⅢ:0.1701 ±0.0083,and WHOⅣ:0.0831 ±0.0077) was decreased with the increased grade of astrocytoma ( all P < 0.05 ).The expression of BMP4 was negatively correlated with MnSOD (r =- 0.821,P < 0.05) and BCL-2 (r =- 0.711,P < 0.05 ).TMZ resistant cell line was constructed and the expression of BMP4 was decreased compared to U251 cells.The resistance was abolished when plasmid of BMP4 was transfected into the TMZ resistant cells.The TMZ inhibitory rate in blank group was ( 2.10 ± 0.04 ) %,and ( 45.70 ± 2.20 ) % in transfected group ( 45.70 ± 2.20 ) % ( P <0.05 ). Conclusion BMP4 was associated with chemotherapy sensitivity of TMZ. Key words: Glioma; Bone morhogenetic proteins 4; Chemotherapy sensitivity
