Abstract

Objective To research the relationship between calpain small subunit 1(Capn4) gene and clear cell renal cell carcinoma and the influence of Capn4 expression change to invasiveness of 786-0 cells. Methods Capn4 expression was measured in 75 pairs of clear cell renal cell carcinoma(ccRCC)and matched adjacent non-tumor tissues by real-time quantitative polymerase chain reaction(Real-time PCR)and immunohistochemistry.Specific small interfering RNA(siRNA) for Capn4 gene was designed and synthesized, and then 786-0 cells were transfected with siRNACapn4 to inhibit the expression of Capn4 gene.After RNA interference,invasiveness of 786-0 cells was detected. Results Capn4 expression in the ccRCC tissues was significantly higher than in the adjacent non-tumor tissues(2. 125±0. 793 vs.1. 480±0. 584, P< 0. 05); the expressions of Capn4 were closely related with lymphatic metastasis of clear cell renal cell carcinomain[odds ratio(OR)= 0. 598, 95% confidence interval(CI)0. 389-0. 918, P< 0. 05].After RNA interference, the invasiveness of 786-0 cells reduced significantly(166±52 vs.448±47, P< 0. 05). Conclusion There is a relationship between Capn4 gene with metastasis of renal carcinoma,and the inhibition of Capn4 gene expression can inhibit the invasiveness of 786-0 cells.
